Here s what we ve heard I have more confidence in my results if I use two kits I need to trust that my profiles are real & not due to contamination I don t want to have to redo work I ve already done SE33 is an important marker- but it s so big! I find it hard to recover loadable profiles from degraded samples Resolving mixtures is the bane of my existence QC what s important to me I need to get more out of less 2

leading to Product Development Goals Build a high quality, robust STR multiplex kit for casework customers in central Europe, UK & the Nordic region: Developed specifically for use in laboratories that will utilize the expanded European Standard Set of loci. Optimize kit configuration to maintain small amplicon sizes for original ESS loci. Address the dual amplification requirements many labs have when combined with the NGM SElect Kit. With increased sensitivity for casework samples with low DNA concentrations. Meeting the requirements for forensic DNA consumables, specifically ISO

Announcing NGM Detect PCR Amplification Kit a new kit under development Powered by 6-dye, the NGM Detect kit builds on the well-established NGM Select kit format and is being developed specifically for users who require loci based on ENFSI and EDNAP requirements. With complementary marker positions in the two kits, NGM Detect and NGM Select will build a powerful combination to gather maximum information from challenging casework samples. 4

Enabling Increased Database Searches SGM+ SGM+ EESS 6 Dyes: Facilitates smaller marker sizes 7 mini STRs (whole marker range <230bp) SGM+ EESS EESS EESS EESS SE33: Locus with highest discrimination power Now completely under 350bp Moved to stronger dye channel Prioritization of markers with the highest representation in databases will enhance recovery of searchable information from casework samples. * - Original European Standard Set EESS- Expanded European Standard Set Detect more from less 5

Dual Amplification Compatibility NGM Detect NGM SElect An advanced partner for the NGM Select kit- same loci, different primer sequences with different marker placement. In combination, the two chemistries will allow recovery of additional searchable information from challenging samples with 12 independent mini STRs (<230bp) across both kits. Detect more from less 6

Improved Sensitivity New master mix formulation coupled with the ability to add additional DNA volume to the reaction (15 microliter vs. 10 microliter) will enable higher recovery of allelic data. Experiment set up: 11 replicates for each kit at each DNA input. Run parameters as per manufacturer s recommendations. Detect more from less 7

Additional Y-marker Amelogenin Y null alleles can occur due to deletions. Leads to a false designation as a female profile. An additional gender marker can highlight Amelogenin Y null allele: Recovery of true designation. The Y indel is on the long arm of the Y chromosome to minimize the risk of a double deletion. p q Amelogenin Y indel Inclusion of a Y indel marker will enable gender confirmation giving you confidence in your results. 9 A Passion for Delivering Products You can Trust My assessment has shown that there is a good correlation between the production of consumables at the Thermo Fisher Scientific sites in Warrington and the requirements stipulated in the ISO standard. Kevin Sullivan PhD, Director at Principal Forensic Services We are proud to announce that our manufacturing facilities are ISO compliant and delivering high-quality Forensic DNA Grade products. Thermo Fisher Scientific is the first manufacturer of HID products in the industry to have its compliance inspected and verified by a third party auditor. 9

Summary The new NGM Detect chemistry builds on the well-established NGM Select format with new primer sequences that will enable optimal marker positioning developed specifically for users who require loci based on ENFSI and EDNAP requirements. Will enable you to detect more with: Reconfiguration of markers, enabling more database searches. Dual amplification compatibility when partnered with NGM Select. Improved sensitivity due to ability to add in more input volume. Additional Y- marker for gender confirmation. Designed to deliver answers you can trust through our commitment to quality. 11
