2 RESEARCH EXPERIENCE: 1. Ph.D. Thesis Submitted ( ): (Lab: Dr. Bikash Mandal, IARI, New Delhi) Thesis entitled Development of plant viral vectors for expression of heterologous protein in plant system : Advanced Centre for Plant Virology, Division of Plant Pathology, Indian Agricultural Research Institute (IARI), New Delhi-12. The full-length CGMMV (6.4 kb) genome was cloned and developed as an infectious clone based on two methodology, in vitro transcription and binary vector construction. The full-length infectious clone was utilized as a gene expression vector to clone GFP and PRSV light chain antibody gene at the end of CGMMV CP terminal and successfully expressed in the plant system. Expression vector of CGMMV was constructed by deleting partial gene sequences of coat protein. Validation of construct was done with GFP and PRSV light chain antibody gene. Both genes were highly expressed compared with the full-length expression vector and proved by real-time PCR and this construct has virus free protein expression in the plant. The purification of PRSV light chain antibody protein was done from the recombinant CGMMV full-length and CP partial deleted vector infected tissue separately and the recombinant proteins can thus be used as antibodies for detection of PRSV in the virus-infected tissue by ELISA and western blot. 2. Currently working as SRF in the project entitled Development of genetically engineered vaccines against economically important poultry viral diseases associated with Dr. Bikash Mandal, Principal Scientist, IARI, New Delhi. This project mainly focuses on application of RNA plant viruses as expression vector in the production of plant-based pharmaceutical proteins without genetic modification of plant. In this work, I have developed full-length infectious CGMMV based expression vector for gene expression. Vaccines were developed against Newcastle disease virus (NDV) Hemagglutinin- Neuraminidase (HN) and Bursal disease virus (IBDV) VP1 and successfully expressed in plant system. The Indian isolate of PVY (9.7 kb) was successfully amplified by overlapping extension PCR and developed as an infectious clone. The clone will be use as an expression vector for future purpose. DNA virus based CYVMV (2.7 kb) genome was utilized as replicon based gene expression vector, confirmed by GFP. 3. Positioned as SRF in ICAR funded a project entitled Genomics & Molecular Breeding of Linseeds / Flax from 10/11/10 to 31/03/2012, associated with Dr. Anand Kumar, NRC on Plant Biotechnology, New Delhi.

3 Differential expressed (DE) genes were found in the subtraction hybridization from the drought stress linseed, DE genes were cloned and sequenced. The DE genes were reconfirmed by realtime PCR in the drought stress filed sample The bri1 gene has an essential role in BR-regulated plant growth and development. The fulllength bri1 gene was isolated from the linseed, mustard and cucumber by genome walking and rapid amplification cdna ends. The full-length bri1 gene was cloned in binary vector and was successfully over expressed into bri1 mutated dwarf arabidopsis and rescues a weak bri1 mutant. The CKX3 and CKX5 genes were isolated from the linseed and mustard plant through genome walking and RACE methodology. Both full-length genes were clone in bacterial expression vector and were over expressed recombinant proteins were purified from the bacteria. The purified protein was exhibits the CKX activity compared to control total plant protein. 4. Positioned as RA in DBT funded project entitled Referral center for molecular diagnosis of transgenic planting materials from 04/11/09 to 22/09/10, associated with Dr. Gurinder Jit Randhawa, National Bureau of Plant Genetic Resources, New Delhi. This project mainly focused on detection of GMO crop based on high throughput method of Real-time, multiplex PCR, qualitative and quantitative analysis of transgenic plants on field levels. This project helped in identification of GMO contamination in the field levels. Apart from that low-cost multiplex PCR-based detection kits were developed and commercialized for GMO crops detection. Dissertations: Post M.Sc., Diploma Title: Host Range Studies on the Soybean Isolate of Mungbean yellow mosaic india virus from Madhya Pradesh. Supervisor: Prof. R. USHA, Madurai Kamaraj University, Tamil Nadu, India This work targeted at understanding the host range of the soybean isolate of Mungbean yellow mosaic india virus (MYMIV) from Madhya Pradesh. The plants tested were cotton, chili, bhindi besides the natural host, soybean. The result of the study is useful for selecting plant species that can be explored by using VIGS vectors based on MYMIV.
