MASTER CLASS ATP HYGIENE MONITORING & RAPID TESTS FOR COLIFORM/E.COLI/TOTAL COUNT 9TH FOOD SAFETY AND & QUALITY SUMMIT NEW DELHI 2&3 DECEMBER 2014

ATP Hygiene Monitoring What is it? A test method to verify good cleaning wherever you want it, whenever you want it Portable, easy and ready for use Tests can be done at line, giving instant feedback on status of cleanliness Ideal for industries with a need for rapid cleaning control and who want to improve their hygiene and quality standards Key advantage is speed and ease-of-use

PURPOSE OF CLEANING Primary purpose is to remove organic soiling i.e. Environmental Cleaning (Stage 1) Also reduced microbial bioburden Ideal test is a direct, instant test for organic residue e.g. ATP Only after good cleaning can disinfectants can be applied with confidence Disinfection Cleaning ( Stage 2) Hygiene monitoring is an assessment of the cleaning regime over time i.e. supervise and inspect Are standards being maintained? Can we improve it?

PROVEN TECHNOLOGY ATP tests have been around since late 80 s and are approved as rapid cleaning verification test British Retail Consortium have developed a global standard for food safety and quality, involving ATP testing. Edition 6, page 40 advises the use of ATP for cleaning verification. Compliant with HACCP Value for money

ATP HYGIENE MONITORING SYSTEM Two part system composed of 1. Hardware = Detection instrument supported by data analysis software 2. Swab device for sample collection and testing Designed for: Simplicity ( easy of use) Convenience Robustness Affordability Sustainability Multi-functional

COMPARISON WITH OTHER METHODS What is clean? Visual Microbiology ATP Fast? Sensitive? Detection of product residues? Simple?

WHAT IS ATP? ATP stands for Adenosine-Tri-Phosphate, which is the energy source of all living organisms and food. If ATP is detected on a surface, it means it has not been cleaned properly. Sources of ATP: Food Microorganisms People

MEASUREMENT PRINCIPLE Detection of ATP by bioluminescence reaction ATP reacts with luciferine/luciferase enzyme complex to generate a light reaction, which is measured in a sensitive and dedicated luminometer The ATP test is the most common used rapid test in the world to determine cleaning efficiency.

ATP VS CLEANING RESULT high ATP (expressed in RLU) low Cleaning result excellent very poor The more food residues or microorganisms, the more ATP is detected, the higher the light output.

TWO FORMATS Ultrasnap & Supersnap For surface hygiene monitoring For flat as well as for irregular surfaces Immediate result Aquasnap For water samples (like rinse water after Cleaning in Place, CIP) Dip Aquasnap into the water Immediate result

PROCEDURE Swab Snap Squeeze Read

SIMPLE INTERPRETATION Interpretation is done by pass/fail limits, which are already pre-set in the instrument. For the most common food contact surface (stainless steel), these pass/fail limits are: Pass: <10 RLU ( ) Caution: RLU (!) Fail: >30 RLU (X)

BENEFITS OF ATP SYSTEM Simple and easy to use by anyone, anywhere, any time Providing instant objective quantifiable data Permits immediate corrective action Identify hot spots and potential risk areas Optimises cleaning processes Demonstrates and reinforces Best Practice Provide real data for Quality monitoring, evidence of due diligence, value for money Supports training and awareness Enables team building through more effective direct communication Restores pride, ownership and commitment of cleaning staff Encourages behaviour change Clean it Prove It!

MICROSNAP RAPID DETECTION OF COLIFORMS/E.COLI & TOTAL COUNT

MICROSNAP RANGE These are applications that require sample preparation and handling and therefore cannot be done at line. But nevertheless very simple and reliable. Microsnap Coliform/E.coli a very rapid and low cost screening test for Coliforms and E.coli. Total test time 6-8 hours. MicroSnap Total a rapid low cost screening test for total viable counts. Total test time 7 hours.

MICROSNAP COLIFORM/E.COLI SPECIFIC DETECTION Sensitive luminometer Microbial detection Specific enzyme detection AOAC approved

SAMPLE TYPES Surface swabs Raw material and finished product Solids, liquids and filterable products Food & beverage; dairy; cosmetics & pharma and water

TEST PROCEDURE After incubation, transfer 100µl from the enrichment device (A) and transfer to the detection device (B). 100µl Activate detection device (B) with a snap and a squeeze. Read RLU in luminometer Interpret RLU results Incubate for 10 minutes at 37 C

INCUBATION TIME Incubation Time (37C) (h) Lowest Inoculum Detected with 95% certainty 1 < < < < <100 6 <10 7 <5 8 1 VIDEO

MICROSNAP TOTAL Sensitive luminometer Two-step procedure 7 Hours incubation (Step 1), followed by immediate measurement (Step 2) AOAC approval pending

SAMPLE TYPES Surface swabs Raw material and finished product Solid and liquid samples Food & beverage; dairy; cosmetics & pharma and water

TEST PROCEDURE 100µl After incubation, transfer 100µl from the enrichment device (A) and transfer to the detection device (B). Activate detection device (B) with a snap and a squeeze. Read result in luminometer Interpret RLU results

INTERPRETATION OF RESULTS Correlation between CFU/ ml and average RLU After 7 hour Incubation at 30 C CFU per ml/100cm2 EnSURE RLU < 100 < 10 < 200 < 20 < 300 < 30 < 500 < 50 < 1,000 < 100 < 10,000 < 1,000 < 100,000 < 9,999 VIDEO
