3 Canonical Wnt Signaling TIPS The Wnt pathways are involved in the control of gene expression, cell behavior, cell adhesion, and cell polarity. The Canonical Wnt Signaling pathway is the best studied of the Wnt pathways and is highly conserved through evolution. In this pathway, Wnt signaling inhibits the degradation of β-catenin, which can regulate transcription of a number of genes. Wnt signaling is activated via ligation of Wnt proteins to their respective dimeric cell surface receptors composed of the seven transmembrane frizzled proteins and the LRP. Upon ligation to their receptors, the cytoplasmic protein disheveled (Dvl) is recruited, phosphorylated and activated. Activation of Dvl induces the dissociation of GSK-3β from Axin and leads to the inhibition of GSK-3β. Next, the phosphorylation and degradation of β-catenin is inhibited as a result of the inactivation of the destruction complex. 7 Complement Activation Pathways TIPS The complement system is an enzyme cascade that is a collection of blood and cell surface proteins to help the abilities of antibodies to clear pathogens from an organism. The complement system that comprises 30 different proteins, including serum proteins, serosal proteins, and cell membrane receptors is an important part of the innate immune system. Some complement proteins bind to immunoglobulins or to membrane components of cells. Others are proenzymes that, when activated, cleave one or more other complement proteins and initiate an amplifying cascade of further cleavages. The endresult of this cascade is massive amplification of the response and activation of the cell-killing membrane attack complex. The complement system has four major function, including lysis of infectious organisms, activation of inflammation, opsonization and immune clearance. 11 Death Receptor Signaling TIPS Death receptors are cell surface receptors that transmit apoptotic signals initiated by specific ligands and play a central role in instructive apoptosis. Death receptors belong to the tumor necrosis factor receptor (TNFR) gene superfamily. Eight members of the death receptor family have been characterized so far: TNFR1, CD95, DR3, TRAILR1, TRAILR2, DR6, EDAR and NGFR. When these receptors are triggered by corresponding ligands, a number of molecules are recruited to the death domain and subsequently a signaling cascade is activated. Two types of death receptor signaling complex can be distinguished. The first group comprises the death-inducing signaling complexes (DISCs) that result in the activation of caspase-8, which plays the central role in transduction of the apoptotic signal. The p53 protein is a nuclear transcription factor that regulates the expression of a wide variety of genes involved in apoptosis, growth arrest or senescence in response to genotoxic or cellular stress.
