2 What is STH-PAS? STH(Single-stranded Tag Hybridization) Method: 1Conduct PCR amplification of target DNA using a labeled primer and a primer with single-stranded tag DNA 2Spread the amplified solution on the membrane strip without heat denaturation (singlestranded) 3Trap the amplified PCR product using strong hybridization reaction between singlestranded complementary tag DNA and single-stranded tag immobilized in a line shape 4Labeling agent for the trapped amplified PCR product enables the line visualized by coloring 5A new genetic testing method to determine the presence or absence of the target DNA in a sample PAS(Printed-Array Strip): Membrane strip used in the above STH method with several kinds of complementary DNA tag immobilized in a line Basic width of the line is 0.5mm which enables the visualization (March 2014:The STH method patented as a new genetic testing method)

5 Comparison with the conventional nucleic acid chromatography method Heat denaturation process is not required, since the single-stranded DNA tag is captured.
