7 Description (Taq DNA Polymerase) Taq DNA polymerase is purified from E. coli expressing cloned DNA polymerase from Thermus aquaticus. The enzyme consists of a single polypeptide with molecular weight of approximately 94kDa. It has 5-3 DNA polymerase activity and 5-3 exonuclease activity. It lacks 3-5 exonuclease activity. This enzyme is suitable for routine amplification. Features (Taq DNA Polymerase) Expression rate is about 1-2 kb/min. Template-independent A can be generated at the 3 end of PCR product. Amplification of genomic DNA fragment up to 4 kb. 8 Features (Pfu DNA Polymerase) 18-fold fidelity as compared to Taq DNA polymerase. Expression rate is about 0.5 kb/min. PCR Product can be directly cloned in to Blunt Vectors. Amplification of genomic DNA fragment up to 6 kb. Amplification of plasmid DNA fragment up to 10 kb. Since, it is not hot-start, we recommended to add enzyme last during PCR. 10 Tips for optimizing PCR reaction A final concentration of 2mM MgSO4 is sufficient for most targets amplification.
