4 ELISPOT Instrumentation and Reagents The enzyme-linked immunospot (ELISPOT) assay is a powerful tool for detecting and enumerating individual cells that secrete a particular protein in vitro. Based on the sandwich enzyme-linked immunosorbent assay (ELISA), the ELISPOT assay derives its specificity and sensitivity by employing high affinity capture and detection antibodies and enzyme-amplification. Originally developed for analyzing specific antibody-secreting cells, the assay has been adapted for measuring frequencies of cells that produce and secrete other effector molecules, such as cytokines. The sensitivity of the assay lends itself to measurement of even very low frequencies of cytokineproducing cells (e.g., 1/300,000). Recent developments in assay plate design and in ELISPOT plate-reader instrumentation have significantly improved the utility of the ELISPOT method for objective and rapid analysis of cytokine-producing cells. BD Biosciences Pharmingen is pleased to announce the availability of optimized ELISPOT reagent sets and instrumentation, in collaboration with Cellular Technology Ltd (CTL) of Cleveland, OH. BD Biosciences Pharmingen is now marketing CTL s state-of-the-art automated ELISPOT plate reader, the ImmunoSpot Series 1 Analyzer, along with high sensitivity ImmunoSpot ELISPOT plates and ELISPOT-optimized antibody pairs. The ImmunoSpot Analyzer offers a degree of sensitivity and specificity which is not possible using other approaches, as well as unique features for data analysis, storage and high throughput screening. Primed human PBMCs were restimulated in culture with PMA and ionomycin in the microwell of an ImmunoSpot ELISPOT plate, which was precoated with unlabelled NA/LE anti-human IL-10 antibody (Human IL-10 ELISPOT Reagent Set, Cat. No ). Biotinylated anti-human IL-10 antibody was used to detect the captured IL-10. Spots were visualized using avidin-hrp enzyme and AEC substrate, followed by image analysis and spot enumeration using an ImmunoSpot. 5 BD OptEIA ELISA Kits Developed for the highest accuracy with serum, plasma, and other biological fluids, the BD OptEIA ELISA Kit II format is the latest product of BD Biosciences Pharmingen s ongoing assay R&D efforts. The Kit II format demonstrates mean spike recovery improvements of 7% to 41% in serum and plasma. With cell culture supernatant samples, values in the original kits and the Kit II s are equivalent. The BD OptEIA Kit II contains a new ELISA diluent, standard/sample diluent, and one-step TMB substrate reagent, with an improved protocol. 6 BD OptEIA ELISA Sets BD OptEIA Sets can be used to measure cytokine (and other soluble protein) levels in serum, plasma and cell culture supernatants. The Sets utilize specifically fragmented antibodies to reduce interference by autoantibodies and plasma proteins such as complement components, acute phase proteins, and fibronectin.
