1 AP Biology Reading Guide Name Chapter 16: The Molecular Basis of Inheritance Concept 16.1 DNA is the genetic material 1. What are the two chemical components of chromosomes? 2. The search for identifying the source of Mendel s heritable factors and Morgan s genes on chromosomes began in the early 20 th century. Scientists believed the two likely candidates were the two chemicals you named above. Until the 1940 s, most believed proteins were responsible for heredity. However, the experiments of Griffith, Avery, and Hershey and Chase, caused others to question whether it was something other than proteins. Read through each experiment and summarize the design of their experiments, the results, and their final conclusions. Experimental Design Results Final Conclusion Hershey & Chase Avery Griffith 1

Given that the DNA of a certain fly species consists of 27.3% adenine and 22.5% guanine, use Chargaff s rules to deduce the percentages of thymine and cytosine. 10. Name the five nitrogenous bases, and put a checkmark in the correct column for each base. Also indicate if the base is found in DNA (D), RNA (R), or both (B). Explain how Meselson and Stahl confirmed the semiconservative mechanism of DNA replication. 17. Where does replication of a DNA molecule begin? What is unique about this starting point? 18. Distinguish between the leading and the lagging strands during DNA replication. 3

Now let s go back and review the enzymes used in DNA replication and their functions. Identify which enzyme does each of the following: Function a. untwists and separates strands Enzyme b. holds DNA strands apart c. synthesizes RNA primer d. adds DNA nucleotides to new strand e. relieves strain caused by unwinding f. joins DNA fragments together g. removes RNA primer and replaces with DNA 22. Put it all together! Make a detailed list of the steps that occur in the synthesis of a new strand. 4
