1 Applied Biosystems Thermal Cycler Remote Access User Guide A step-by-step guide to setting up secure access to your thermal cycler with Thermo Fisher Connect

2 Check run status from any mobile device This guide will walk you through the process of setting up your Applied Biosystems SimpliAmp Thermal Cycler and ProFlex PCR System for remote accessibility through Thermo Fisher Connect. 3 Ensure instrument is cloud-enabled Any SimpliAmp Thermal Cycler or ProFlex PCR System can be connected to the Thermo Fisher Cloud as long as it is using firmware version 1.2 or higher. You can check your current firmware version by: 1. Going to the Settings menu. 2. Selecting About Instrument. If the instrument does not have firmware version 1.2 or higher, you will need to upgrade your firmware, using the USB provided or by going to thermofisher.com/simpliamp or thermofisher.com/proflex 1. Insert USB preloaded with new firmware into the USB port. 2. Go to the Settings menu, or touch the shortcut to the Upgrade Firmware screen if you have enabled the USB pop-up. 3. Go to Maintenance & Services, then Software Update. 4. Select the appropriate file and touch Update and Yes to confirm that you want to upgrade. IMPORTANT! Do not power off the instrument during the upgrade. When the upgrade process is complete, the instrument will restart automatically. Create Thermo Fisher Connect account 1. From your Internet browser, go to thermofisher.com/connect 2. If you already have an account with Thermo Fisher Scientific, log in using the same name and password. 3. If you do not already have an account, follow the prompts to create one. 4. Once you have logged in to thermofisher.com/connect, an will automatically be sent to you to verify your address. 5. Open your and click on the link to visit the Connect page to verify your account. 6. If you have done this successfully, you will receive a confirmation telling you that you have connected to Thermo Fisher Connect. Connect instrument to the internet SimpliAmp and ProFlex Thermal Cyclers can be connected to the Internet using a wireless (using the High-Power USB Wi-Fi Module, Cat. No. A26774) or wired connection. See the product manual for detailed instructions on setting up either connection type. 4 5

4 Connect instrument to Thermo Fisher Cloud 1. Touch Sign In. 2. Select Administrator when asked to choose screen name. 3. Log in. (The default password for the Administrator account is password.) 4. You will then be redirected back to the home screen. Go to Settings. 5. Select Remote Service. 6. Turn on Remote Service. 7. Touch My Profile on the home screen. 8. Touch the Cloud tab and enter your Thermo Fisher Connect account login and password. 9. Select Link Account. Upon successful authentication, the Administrator is linked to Thermo Fisher Connect. Set up mobile device to connect to instrument Download the Instrument Connect Mobile Application on your mobile device. 1. For an ipad or iphone mobile device, download the application from the App Store by searching for Instrument Connect. 2. For Android devices, download the application from Google Play by searching for Instrument Connect. 3. Log in to the Instrument Connect mobile application using your Thermo Fisher Connect login and password. Subsequently, any user can log in and link the instrument to their Thermo Fisher Connect account by following steps 3 9 above. NOTE: The administrator account must be activated first to connect the instrument to the Cloud. Afterward, multiple users can create unique user profiles using their own login information. Each unique profile must have a unique Thermo Fisher Connect account. 6 7

5 Check run status from mobile device 1. Open the Instrument Connect app. If login is required, enter your Thermo Fisher Connect login and password. 2. The dashboard is displayed upon login, showing the status of all registered instruments. 3. To check the time remaining and the protocol stage for a running instrument, click on it. 8 9

6 Design methods from desktop computer and upload to instrument 1. From your Internet browser, go to thermofisher.com/connect 2. Log in using your Thermo Fisher Connect login and password 3. Select the PCR Designer application from the dashboard. 5. Follow these steps to create a new experiment (method): a. Enter experiment properties. Click Next. b. Enter the stages and steps in the method. Click Next. 4. Select Create new experiment. (At a later time Open existing experiment may be selected to view templated protocols and those you ve already written.) 10 11

7 c. Define plate layout details. Click Next.
