2 Cell Health Overview CELL HEALTH No experiment is complete without all its components and one key component is confidence in the health of your cells. Determining the health of your cells is a crucial component in ensuring reproducibility of your experiments. Whether you are assessing viability of cells in culture, cell proliferation in response to different stimuli, are trying to understand which treatments induce apoptosis or autophagy, or simply want to improve your flow cytometry data, we have the right reagent for you. CELL CULTURE Cell Viability Measure general cell health by monitoring parameters such as redox potential and membrane integrity. 3 Cell Viability Cell Viability Determining cell viability is vital for many experiments. It can be the data in toxicity assays, be used to determine side effects of experimental treatments, provide accurate information about cellular activity, or simply be used to ensure that you are studying a live cell population. We offer a range of versatile and easy-to-use cell viability assays, including convenient fixable dyes that allow you to measure the viability of cells using flow cytometry, microscopy, and ELISA. Featured : VivaFix Cell Viability Assays Fixable cell viability assays can be used to easily discriminate dead cells from live cells in both fixed and unfixed samples using flow cytometry. Assays are available in a range of excitation and emission spectra for convenient addition to multicolor flow cytometry panels (see table below). 4 Cell Proliferation Cell Proliferation An increase or decrease in cell growth and division can be a key result in cytotoxicity and apoptosis assays and can be used to determine the effect of experimental treatments on cellular replication. We offer a broad range of antibodies, kits, and dyes to monitor proliferation. Proliferation can be measured through dye incorporation into live cells, by quantifying key proliferation markers, or by assessing incorporation of the thymidine analog BrdU into DNA. Featured : CytoTrack Cell Proliferation Assays CytoTrack Dyes allow you to measure cell proliferation in live cells. As the labeled cells divide the concentration of the dye is halved. Proliferation can be measured and cell generations distinguished using this cell-division dependent decrease in fluorescence. 5 Autophagy Autophagy Autophagy is the orderly degradation and recycling of cytoplasmic components via the lysosome. Autophagy can be induced by factors such as starvation or growth factor suppression, ER stress, infection, or through immune signals. Autophagy is thought to be a repair and survival response during stress and infection and has been linked closely to apoptosis. We offer a range of antibodies and kits to detect and measure markers of autophagy. These include reagents to detect the lysosome, the autophagosome, and the most-studied macroautophagy pathway. Featured : MAP1LC3A/B Antibody MAP1LC3 family proteins are widely used as reliable markers for monitoring autophagy. LC3-I is the cytosolic form of LC3, which is converted into the active, membrane-bound form LC3-II, during autophagy. Tracking the level of conversion of LC3-I to LC3-II provides an indicator of autophagic activity. The rabbit anti human MAP1LC3A/B (N-Terminal) antibody recognizes both the LC3-I and LC3-II forms of MAP1LC3A and MAP1LC3B in immunofluorescence and western blot experiments. Fig. 3. Autophagy can be monitored by immunofluorescence using AHP2167, a MAP1LC3A/B antibody. HeLa cells were fixed with 3% paraformaldehyfe and permeabilized with 0.3% Triton X-100. Nuclei were stained with 1.5 µg/ml DAPI. Immunofluorescence was carried out using AHP2167, a rabbit anti human polyclonal MAP1LC3A/B antibody at a 1:25 dilution. STAR36D488GA, a sheep anti rabbit DyLight 488 antibody was then used at 1:50. Cells were visualized on the Bio Rad s ZOE Fluorescent Cell Imager at 60x magnification. 6 Apoptosis Apoptosis Apoptosis is the process of programmed cell death characterized by distinct morphological and biochemical changes within a cell. Featured s: Mitochondrial Membrane Potential Kit and Bcl-2 PrecisionAb Antibody An early indication of apoptosis involves a collapse of the electrochemical gradient across the mitochondrial membrane. Loss of mitochondrial membrane potential can be detected by a unique fluorescent cationic dye known as JC-1 (5,5,6,6 -tetrachloro-1,1,3,3 -tetraethylbenzamidazolocarbocyanin iodide) that has been incorporated into our MitoPT JC-1 Mitochondrial Permeability Transition Kit. Bcl-2 is a member of a family of anti-apoptotic proteins that prevent the release of sequestered mitochondrial cytochrome c and subsequent caspase 9 and 3 activation to promote cell survival.
