3 NutriDisk Dehydrated Media Products Orange Serum Nutrient Pad Intended Use: Medium for the isolation, cultivation and enumeration of acid-tolerant, putrafacation microorganisms in fruit juices and fruit juice concentrates, especially those from citrus fruits. This culture medium is in accordance with the recommendations of the German Institut für Lebensmitteltechnologie und Verpackung (Institute for Food Technology and Packaging 1974). Recommended Incubation Time: 3 5 days at C Incubate the inoculated culture medium up to 3 days; if fungi are suspected to be present, incubate up to 5 days. Determine the microbial count. Further tests can be performed to differentiate and identify the colonies. Additional information: The culture medium is optimally adapted to the special requirements of the microbial flora present in citrus juice (e.g. Bacillus, Lactobacillus, Leuconostoc species, fungi, etc.) due to the fact that it contains orange extract. It is therefore especially useful for production control in the fruit juice industry. Orange Serum Nutrient Pad Ingredients per liter of demineralised water adjusted to ph 5.5 ± 0.2 at 25 C. Mold develops velvety or fluffy whitish to greenish colonies which can take on various colors after conidiospore production. Yeast and bacteria colonies have smooth surfaces. Acid forming sugar fermenters have a whitish to yellow coloration; non acid formers are, by contrast, greenish to blue green. Select characteristic colonies from Schaufus Pottinger plates and perform additional physiological and serological tests as required. Note that colonial morphology as observed macroscopically is a presumptive aid and does not provide information for specification. Recommended Incubation Time: 2 3 days at C. 4 Products NutriDisk Dehydrated Media Standard Plate Count Nutrient Pad Intended Use: Standard Plate Count Media is recommended for use in determining the Standard Plate Count of viable bacteria in milk, dairy products, and other material of sanitary significance. Standard Plate Count Media is formulated according to specifications from the APHA for use in performing the Standard Plate Count for bacteria in dairy products, and the same formulation described for Plate Count Agar, dehydrated (Tryptone Glucose Yeast Agar) in the Standard Methods for the Examination of Water and Wastewater 13 th, Edition 4. It is also the same formulation as the Tryptone Glucose Yeast Agar from the Association of Official Analytical Chemist (AOAC) and conforms to the formulation specified by the U.S. Pharmacopea. Recommended Incubation Time: 2 5 days at 25 C. Historical background: Standard Plate Count Media is a modification of Bowers and Hucker Tryptone Glucose Skim Milk Agar. It was shown by Dr. Yale (approx. 1938) to be more effective in plate count procedures for milk and dairy products. The American Public Health Association (APHA) in their Standard Methods for the Examination of Dairy Products recommends the use of Standard Plate Count Media for the performance of the Standard Plate Count on dairy products. Standard Agar is the same formulation as Plate Count Agar recommended by the APHA for use in the Standard Plate Count for water. Standard Plate Count Nutrient Pad Ingredients per liter of demineralised water adjusted to ph 7.0 ± 0.2 at 25 C. Peptone (Pancreatic Digest of Casein) Yeast Extract Dextrose 5.0 g 2.5 g 1.0 g Standard TTC Nutrient Pad Intended Use: For the determination of the colony count of water and waste water; for pure culture and as a base medium for further additions to demineralised water (serum, antibiotics etc.) by wetting the pads. Additional information: 2, 3, 5-Triphenyltetrazolium chloride (TTC) is rapidly reduced to red formazan by almost all coliform organisms including Escherichia coli. For the cultivation, isolation and enumeration or enrichment of fungi, especially of yeast. Recommended Incubation Time: 2 5 days at 25 C Additional information: The accompanying bacterial flora is weakly suppressed by a ph value of 5.0. Evaluation: Yeast usually develop as smooth white or colored colonies. Mold, as a rule, form velvety or fluffy, cotton-like colonies which are white in the early growth phase and may later take on various colors after conidiospore production. Select characteristic colonies from wort plates and perform additional physiological and serological tests as required. 9 Rapid Tests Contamination tests using the membrane filtration method Yeast and Lactobacillus Rapid Test Quantitative Rapid Tests The Yeast and Lactobacillus Rapid Tests are based on the membrane filtration method. In addition the quantitative results are available within 24 hours, rapid determination of yeast contamination within 8 hours. Products Yeast Rapid Test For the rapid detection of yeast contamination in soft drinks. Yeast concentrations of 1000 yeast cells/ml can be detected in only 8 hours. The membrane filter is incubated on a nutrient medium and colored blue with reagent solution. After only 24 hours incubation even a single colony can be detected. Package includes: 50 membrane filters, reagent solution, special nutrient media pads for 50 tests. Yeast and Lactobacillus Rapid Test. Lactobacillus Rapid Test For the rapid detection of Lactobacillus contamination in soft drinks. In the microbiological spoilage of soft drinks, carbonated soft drinks in particular, Lactobacilli play an important role. In the LST-45 test the membrane filter is incubated on a nutrient medium and the colonies are then colored blue with a reagent solution. Even dense growth can be easily recognized. 11 Swabs SwabCheck Citrus-S Products SwabCheck Citrus-S For the presumptive identification of Escherichia coli on production and product contact surfaces in the citrus industry. Description: This media was specifically developed for the detection of Escherichia coli on all surfaces that a product may come into contact with, such as peeling knives, rotary cutters, filling heads, cappers, pulpers, squeezers etc. The media is modified from that published by Feng and Hartman by the addition of MUG. The test kit uses neutralizing buffer as the sample collection medium. The neutralizing buffer inactivates chlorine and quaternary ammonium based detergents that may be present on a surface after the implementation of a cleaning protocol. It allows residual bacteria whose growth may be suppressed, but not killed, to develop and be identified. 13 Swabs Neutralizing Buffer Swabs and SwabCheck Products Neutralizing Buffer Swabs Neutralizing buffer inactivates the bactericidal and bacteriostatic effects of chlorine and quaternary ammonium detergents. However neutralizing buffer exhibits no toxic effects on microorganisms. This permits the transfer of swabbed organisms to the laboratory without loss in viability. Description: Neutralizing buffer swabs are used in the monitoring of surfaces for total bacterial count. Neutralizing buffer is not designed to culture and enumerate microorganisms. Description: SwabCheck changes color from purple to yellow. The color change is based on an acid reaction with the indicator. The more rapid the color change the higher the level of bacteria in the sample. SwabCheck is useful in determining the sanitation levels of preparation surfaces, filling ports and processing areas in beverage and food processing plants, dairies, restaurants and healthcare facilities. 14 Products Swabs Letheen Swabs Letheen Swabs Letheen swabs are used for the determination of bactericidal activity of quaternary ammonium compounds. Description: Letheen broth, on which the swab systems are based, is a modification of the AOAC formula. Letheen broth incorporates lecithin to neutralize the effects of quaternary ammonium compounds and sorbitan monooleate to neutralize phenolic disinfectants, hexachlorophene and formalin. Lecithin and sorbitan monooleate synergistically react to neutralize the effects of ethanol. Inaccurate counts would be obtained if the disinfectants were allowed to mask the presence of organisms by means of their bacteriostatic action. Description: Buffer swabs contain no bacteriostatic or bactericidal compounds and cannot suppress the action of detergents None required. Description: Bromocresol purple is a ph indicator which demonstrates a color change from purple to yellow in the presence of acid. Lactose fermenting organisms produce acid which initiates the color change. The presence of coliforms is detected with greater sensitivity by use of a relatively large sample volume (100 ml) in a single bottle. Yellow color in the sample after 24 hours incubation at 35 C indicates the presence of coliform organisms. A red sample after 24 hour incubation at 35 C should be incubated for further 24 hours. A red sample at 48 hours incubation at 35 C indicates no coliform organisms present in the sample. 16 Products ColiCheck with MUG ColiCheck with MUG Used for the presumptive identification of coliforms and the determination of the presence of Escherichia coli in water samples by a presence/absence technique. Description: Bromocresol purple is a ph indicator which demonstrates a color change from purple to yellow in the presence of acid. Lactose fermenting organisms produce acid which initiates the color change. The presence of coliforms is detected with greater sensitivity by use of a relatively large sample volume (100 ml) in a single bottle. The addition of MUG (4-Methylumbellifryl-β-D-glucuronide) which is a fluorogenic enzyme allows the media to selectively identify Escherichia coli. MUG is hydrolyzed by the Escherichia coli specific enzyme β-glucuronidase to release 4-Methylumbelliferone which fluoresces under ultraviolet light (approx. 366 nm wavelength). Yellow color in the sample after 24 hours incubation at 35 C indicates the presence of coliform organisms. A red sample after 24 hour incubation at 35 C should be incubated for further 24 hours. A red sample at 48 hours incubation at 35 C indicates no coliform organisms present in the sample. Fluorescence under ultraviolet light is specific for the presence of Escherichia coli.
