3 PRINCIPLE OF THE METHOD (1) The MC Media Pad ACplu i a tet pad coated with gowth medium and thee edox indicato fo detection of aeobic bacteial colonie. Once the liquid ample i inoculated onto the tet pad, the ample diffue though the whole pad by capillay action, econtituting the medium. If aeobic bacteia ae peent, they gow a ed coloed colonie on the tet pad. Fo mot food, a 50 g o 50 ml tet potion i homogenized with 450 ml Buttefield Phophate Buffe. Seial decimal dilution ae pepaed a needed. Fo daiy poduct, an 11 g tet potion of ceam cheee i added to 99 ml 2% odium citate wamed to C and homogenized. An 11 ml tet potion of yogut dink i diluted into 99 ml Buttefield phophate buffe. Seial dilution of daiy poduct ae pepaed with 1 ml o 11 ml of homogenate diluted into 99 ml Buttefield phophate buffe. A 1 ml aliquot of diluted ample i applied to the tet pad, the cove film i eplaced, and the plate incubated at 35±1 C fo 24±2 h (apid uage) o 48±2 h (tandad uage). Even though the apid uage can be ued fo poceed food etc. afte veified by ue, it doe not apply to food containing lage amount of lactic acid bacteia o pychophilic bacteia. Altenatively, a 10 g tet potion of matix i diluted into 90 ml Maximum Recovey Diluent (MRD). Seial dilution ae pepaed with 10 ml homogenate diluted into 90 ml MRD. A 1 ml aliquot of diluted ample i applied to the tet pad, the cove film i eplaced, and the plate incubated at 30±1 C fo 72±3 h (altenate method). DISCUSSION OF THE VALIDATION STUDY (1) The MC Media Pad ACplu method pefomed a well o bette than the elevant efeence method teted in the matix tudy. In all matixe teted, thee wa le than a 0.5 log 10 cfu/g diffeence between the candidate and efeence method and the epeatability of the method wa vey imila. The eult howed that the altenative method wa vey veatile in it application. It howed compaable pefomance to the USA efeence method unde tandad uage (48±2 h at 35±1 C) fo all 10 matixe and unde apid uage (24±2 h at 35±1 C) fo 7 matixe. In addition it howed compaable eult to the ISO efeence method (72±3 h at 30±1 C) fo all 10 matixe. The eult of thi validation tudy indicate that the MC Media Pad ACplu method can be ued fo apid and accuate enumeation of total aeobic count in a vaiety of food and unde a vaiety of tet condition.
