2 PERFECT SOLUTIONS Focus on the user Our customers have been involved in the development process since the production of special blood bank centrifuges began decades ago. Ever since then, novel approaches have been continuously conceived, tested and implemented in association with our customers. As a result of this information acquisition and exchange we have developed high-performance products that have made everyday routines in blood banks and transfusion centres even more efficient and simpler. ROTO SILENTA 630 RS ROTO SILENTA 630 RS is a typical highquality centrifuge for blood bank use. It can achieve high RCF values (up to 6,520) thanks to its advanced rotor technology without a windshield. The comprehensive range of accessories caters for almost all applications, both standard and special ones. For example, blood bag insert 4529 is available for the preparation of thrombocyte concentrate or platelet rich plasma (PRP). It is suitable for all blood bag systems up to 500 ml. The blood bag can be suspended from hooks which are attached, in accordance with the customer s requirements, at one of 3 different heights on the insert. This allows the bag to be stabilized, regardless of its fill quantity, and prevents the accumulation of erythrocytes. Suitable accessories for all applications If the standard accessories are not suited for special applications, customized accessories to meet specific requirements can be manufactured within a very short time. Also, customized accessories have been developed for blood banks and stem cell banks, where they are used to process placental blood and stem cells in blood bag systems of lower volume. The perfect team: ROTO SILENTA 630 RS and the PRESSOMAT Optimally matching accessories facilitate shorter processing times, better preparation quality and higher yields. The ROTO SILENTA 630 RS with blood bag insert 4598-P and the PRESSOMAT plasma separator form a perfect combination. Centrifugation and separation take place in the same insert. Since it is not necessary to manipulate the blood bag system during transportation from the centrifuge to the separator, any disturbance of the erythrocytes is avoided. The HettInfo documentation system logs the processing data and the quality of the blood products derived. The range of blood bank equipment is completed by the ROTOLAVIT cell washing system which is used to test the compatibility of donor and recipient blood. 4 SEPARATION OF BLOOD COMPONENTS Blood is centrifuged to separate it into its basic components - erythrocytes (red blood cells), leucocytes (white blood cells), thrombocytes (platelets) and plasma. These constituents of the blood sediment at different rates according to their density. The erythrocytes collect at the bottom above them the leucocytes, followed by the thrombocytes. The leucocytes and thrombocytes together form a thin white layer known as the buffy coat. The plasma is above the buffy coat. Buffy Coat method 1a Since the standards that govern the demands on the preparation of blood products and their quality vary between countries, different methods are adopted. The most common methods are outlined below: 1b 1. Buffy Coat method Preparation of platelet poor plasma and concentrated red cells from unfiltered whole blood with subsequent production of platelet concentrate from the buffy coat Centrifugation at a high RCF separates the blood into platelet poor plasma (PPP), buffy coat (BC) and concentrated red cells (CRC) with a small proportion of leucocytes. A plasma separator presses the platelet poor plasma (PPP) into one satellite bag, the concentrated red cells (CRC) into another. The buffy coat (BC) remains in the primary bag (1a). The remainder of the leucocytes in the CRC can be removed by filtration. In a subsequent centrifugation step at a low RCF the BC is separated into platelet concentrate (PC), a residual BC and a small volume of sediment containing leucocytes and erythrocytes. The PC is pressed into a satellite bag. The remainder of the constituents are left in the primary bag (1b). In countries where it is not prohibited, the platelet concentrate is prepared from pooled buffy coat. Pooled BC comprises 4 to 6 individual buffy coats that are allowed to flow into one bag. The pool is separated by centrifugation at a very low RCF into PC, a residual buffy coat and a sediment comprising erythrocytes and leucocytes. Upon subsequent expression of the PC it flows through a filter, removing most of the leucocytes and erythrocytes still present in the PC. This yields platelet concentrate that is low in leucocytes. alternative: 1c 1d PCs prepared from pooled buffy coat have higher numbers of thrombocytes (i.e. a higher therapeutic dose) and fewer variations in quality. 4

Pre-filtered whole blood Preparation of platelet poor plasma and concentrated red cells from filtered whole blood Pre-filtered whole blood (e.g. application of the PRESSOMAT) 2a 2b With this method the buffy coat is filtered from the whole blood prior to centrifugation (2a). The subsequent centrifugation step at a high RCF separates the filtered blood into platelet poor plasma (PPP) and concentrated red cells (CRC). The constituents separated by a plasma separator are then collected in satellite bags (2b). 3. PRP method Preparation of platelet rich plasma and concentrated red cells from unfiltered whole blood with subsequent preparation of platelet concentrate and platelet poor plasma In a first centrifugation step at a relatively low RCF unfiltered whole blood is separated into platelet rich plasma (PRP) and concentrated red cells (CRC). The PRP is expressed into a satellite bag (3a). The leucocytes still contained in the CRC are removed through filtration. 6 CONTROL PANELS Simple to program With the S and C control panels, even the most complex centrifuge runs can be programmed quickly and easily. The individual parameters are selected by way of symbol keys, the desired values are set with the rotary knob and stored by pressing the STO key (S control panel) or the START key (C control panel). The operating and display elements are clearly laid out and easy to read. The actual status of all parameters is displayed during operation. The display/entry of RPM can be converted to display/entry of RCF by pressing a button. S CONTROL PANEL (ROTO SILENTA 630 RS and ROTIXA 50 RS) KEYPAD START Starts centrifugation run, short-time centrifugation. Pre-cooling can be assigned to one program location. A PIN code is entered in centrifuges with a C control panel to prevent unauthorized entry or modification of the stored programmes. Models with an S control panel feature a key-operated switch for locking. Centrifuges with C control panel further permit the centrifugation time to be switched from at start (commencing upon start) to at speed (start when the set speed has been reached). STOP PROG STO RCL RCF Stops centrifugation manually. Selects the program. 89 programmable memories are available. Stores entries, changes and programmes. Calls up the selected program. The integral over the RCF indicates the accumulated RCF acting on the centrifuged material up to that particular time. The braking characteristics of the rotor have a decisive influence on the quality of separation achieved, especially at low centrifugation speeds, such as those commonly run in the preparation of thrombocyte concentrate (TC) or platelet rich plasma. The regular R-ramps brake the rotor uniformly over the entire t down time. Loosely packed sediment may be resuspended during this critical run-down phase. In contrast to this, the B-ramps brake the rotor rapidly, at first, then slowly so that it gently comes to standstill. STOP Stops centrifugation manually / OPEN Opens the lid. PROG RCF Entry / recall of the program number. 98 programmable memories are available. Pressing the RCF key several times results in display of the integral over the RCF. 17 Certificates and registrations Quality and safety Hettich centrifuges comply with all relevant EU standards in effect. All stages from design to end-product are continuously subjected to the strictest controls. Evidence is provided by national and international test marks such as IEC or the CE conformity. The ISO 9001, ISO and ISO certificates accredited to the company bear witness to the extreme care and responsibility Hettich puts into the manufacturing of centrifuges and their accessories. This is why Hettich centrifuges are known for safety and reliability all over the world. Marks of conformity for the use of Hettich centrifuges in the US and Canada, on request. according to directive 98/79/EC IvD stands for In-vitro Diagnostics. Hettich centrifuges conform to IvD standards, i.e. they comply with the European IvD guideline s requirements (98/79/EC) and guarantee a standardised level of quality and safety. Non-polluting waste disposal Since , Hettich has been registered in Germany under WEEE Reg. No. DE with the Elektro-Altgeraete Register 1) - Foundation for the implementation of the 2002/96/EC directive (ElektroG 2) ), regarding the disposal of electrical and electronic equipment in the EU. With the symbol of the crossed-out bin, it is made clear that centrifuges may not be disposed of with household waste. The disposal regulations in the individual EU countries can vary. Please ask your supplier, if necessary. 18 HettInfo HettInfo guarantees reliability, transparency and flexibility of data documentation. Data documentation and traceability have to satisfy increasingly stringent requirements. As a consequence, more and more time has to be spent on documentation. With HettInfo transparency can be increased and at the same time processes can be streamlined and rationalized. Packing room with external barcode unit The HettInfo data documentation system consists of combinable modules and can be easily adapted to the required number of centrifuges. Up to 29 centrifuges and 3 external barcode units can be integrated into the network, which is reliable and absolutely immune to disturbances. HettInfo enables logging of all centrifugation data. The user can log the date, end of the centrifugation run, program number and the temperature, and can select and de-select additional optional parameters. If barcode kits are used, the barcodes of blood bags and users can also be logged. In addition, the result of each run is automatically stored with all logged data. This simplifies any search for incorrect centrifuge runs. Modern data back-up mechanisms increase data integrity. To avoid duplication and to ensure that higher-level databases are kept up to date, all data acquired using HettInfo can be integrated into these databases. For this, the data is stored in CSV format at the desired storage location. Time-consuming and costly programming of the interface is not required. 19 Participants The data collected through HettInfo by the participants is stored on a PC with a RS 232 interface. Participants are centrifuges with an online kit or a barcode online kit, external barcode units and additional distributors. In principle any centrifuge with an S or C control panel can be connected to HettInfo through an online kit. Barcodes can also be captured and documented through the use of a barcode online kit or an external barcode kit. To serve the function of a barcode online kit, an external barcode kit must be used in combination with an online kit. In addition to barcodes on blood bags, user data can also be captured via barcodes. This ensures full traceability and saves time and labour. 20 PRESSOMAT The plasma separator with innovative functionality The PRESSOMAT was developed for the production of leucocyte-depleted plasma and leucocyte-depleted erythrocyte concentrate. Its functional principle is quite unique: centrifugation as well as separation take place in the centrifuge insert (= two-chamber insert 4598-P). During transportation from the centrifuge to the PRESSOMAT, the blood bag system is not manipulated, and dispersion of the erythrocytes is thus avoided. 21 The blood bag system An increasing number of blood donation facilities are taking advantage of the benefits of whole blood inline filtration, as long as the buffy coat is not required for the preparation of platelet concentrates. The PRESSOMAT was specially developed for pre-filtered whole blood in a ml T&T blood bag system. 22 ROTOLAVIT Automatic cell washing system for serological testing In blood banks and transfusion centres, it is routine practice to test the compatibility between donor and recipient prior to every blood transfusion. This test is quite labourintensive. Especially the washing of the erythrocytes takes a lot of time. The ROTOLAVIT cell washing system carries out the washing process automatically, hence reducing the workload and saving valuable time. The appropriate quantity of saline solution is filled equally into 12 or 24 tubes, depending on the rotor, and mixed with a previously pipetted drop of blood. It is then centrifuged and the supernatant is decanted. The washing cycle is repeated several times and the anti-human globulin (Coombs) serum is then added manually, in order to assess possible agglutination following the final centrifugation.
