1 Procedure for Stool Routine & Microscopy : To perform Gross & Physical examination of Stool Variables : Specimen collection, Labelling of specimen, Name, Identifying number, date, time Gross Examination : Consistency & form : Normal consistency : well formed Abnormal consistency : Look for the following,whether Pale, bulky, frothy,hard, flattened and ribbon like, semi solid, watery, rice water stool. Presence of adult worms or their parts. Color : Normal color Light to dark brown Abnormal color Black, Bright red, fresh blood, mucus, clay colored, white,
2 Procedure for ph of Stool Specimen : Determination of ph of Stool Specimen : Reagents and chemicals : ph paper (Range 4-9 or 2-10) Procedure : Dip ph paper in small quantity of the faecal material Observe the color Compare with the color chart & record the ph
3 Procedure for Occult Blood Test in Stool Specimen Name of method : Benzidene test : To perform Occult Blood Test in Stool specimen. Specimen Procedure : Glass slides, Applicater Stick or wire loop, Glacial Acetic Acid, Hydrogen Peroxide (30 % V/V), Benzidene powder : Faeces : Take pinch of benzidene powder in small test tube Acidify it with 2-3 drops of, Glacial Acetic Acid and mix well Add about 1.0 ml of Hydrogen Peroxide and mix well Place small quantity of stool specimen on a clean and dry glass slide Place 1 or 2 drops of Benzidene Glacial Acetic acid- Hydrogen Peroxide mixture on the stool specimen on the glass slide Observe change in color Observations : 1) No change in color : Occult blood absent 2) Color changes to Green to Bluer : Occult blood Present
4 Procedure for Urine Routine & Microscopy Variables Procedure : To perform Chemical examination of urine:-by using Uristix Reagent Strip For Protein,Glucose and Ketones. : Uncentrifuged urine. Uristix reagent strip. : Specimen collection, Labelling of specimen, Name, Identifying, number, date, time : Dip the test areas of the strip in the urine specimen(fresh, well mixed and uncentrifuged). Remove the excess of urine by tapping the edge of the strip against the container. Compare the test areas closely with the corresponding color charts on the bottle at the times specified. Note : The strip should be compared in reasonably good light.
5 Procedure for Stool Routine & Microscopy Specimen Variables : To perform Microscopic examination of Stool : Glass slides, Cover slips, Normal saline, lugol s Iodine Solution, : Morning stool specimen collected in a wide mouth clean & dry container : Specimen collection, Labelling of specimen, Name, Identifying number, date, time Procedure : Saline specimen Preparation Place a drop of normal saline on a glass slide Take a little faecal material with wooden stick and mix with normal saline Place a cover slip over it (avoid air bubble) Iodine specimen praparation Place a drop of lugol s iodine on the glass slide mix little faecal material with drop of iodine solution Place a cover slip on it (avoid air bubble) Observation : observe the slide under high power in microscope for Ova, Cyst & Trophozoit form of the various organism.
6 Procedure for Concentration method Specimen Variables : To perform Microscopic examination of Stool for concentration method : Glass slides, Cover slips, Saturated saline Solution, small container : Morning stool specimen collected in a wide mouth clean & dry container : Specimen collection, Labelling of specimen, Name, Identifying number, date, time Procedure : Fill the half of the small container with saturated saline solution Place small amount of faecal material in it with wooden stick & mix well Fill the container completely with saturated saline solution Place a slide on it (avoid air bubble) Leave for 15 minute Lift the slide gently (By taking care that no spillage occure) Place the coverslip on a glass slide Observation : observe for the fertile & non fertile ova of various organism
