4 Filtration of culture media and additives Volumes up to 100 ml Puradisc syringe filters Whatman Puradisc syringe filters from GE Healthcare are available with a wide selection of sizes and membrane types to provide full flexibility for each application. For cold sterilization we recommend the Puradisc sterile syringe filter with a 0.2 µm membrane. The membrane (polyethersulfone or cellulose acetate) is inherently hydrophilic, has low extractables, and exhibits low protein binding. These features allow samples to be filtered without any risk of contamination. The housing is made of pigment-free material ensuring high-quality sample preparation for critical applications. Two options are available: Puradisc 25 mm, with polyethersulfone (PES) membrane Puradisc FP 30 mm, with cellulose acetate (CA) membrane Puradisc features: Clean room manufacturing/production Pore sizes of 0.2 µm and 0.45 µm Nonsterile options available Other membrane types available Fig 1. It combines a glass microfibre (GMF) prefilter and a nylon membrane, to create a high performance capsule that improves flow rates and prolongs filter life. Users can filter large volumes (up to 20 l) and difficult samples more easily. Polycap capsules can filter buffers or heavily loaded biological solutions including virus suspensions, biological suspensions, admixtures, and enzymes. 5 Volumes of 250 to 1000 ml Klari-Flex filtration system Whatman Klari-Flex Bottle-Top Filtration System provides an easy-to-use stand-alone filtration device that improves overall efficiency and minimises the risk of spills and contamination. Klari-Flex includes the low protein binding PES membrane. Klari-Flex system features Drop-Connect Technology, which requires just one hand to connect the unit to the base. There is no need to detach/attach tubing because the vacuum can be controlled using the vacuum knob. Product is gamma sterile and individually bagged. Connect vacuum to the docking station only once and plug in the Klari-Flex filtration unit Pour in your culture media or additives, close the lid and... start filtration by turning on the knob on the docking station Remove the bottle, close it with the cap done! 6 Venting during cell growth Standard flasks (125 to 2500 ml) BugStopper sterile closure devices BugStopper devices prevent bacteria and viruses from entering or exiting a culture vessel, while allowing air and gas to pass through the membrane. BugStopper devices (BugStopper and BugStopper 10) will withstand 50 standard autoclave cycles without loss of performance. Repeated use reduces cost per use significantly. Needles may be used to penetrate the silicone portion of the BugStopper device for use as a sample port or for gas infusion. This is not recommended for BugStopper 10 because of the wall thickness. BugStopper/BugStopper 10 devices are easy to use; you simply push the device into the neck of the flask and autoclave the entire vessel. 7 Alternative culture vessels HEPA-VENT and HEPA-CAP filters The design of these HEPA depth filters gives a high flow rate and low pressure drop, ensuring that clean air passes into and out of vessels. HEPA-VENT and HEPA-CAP prevent bacterial, algal, or fungal contamination in fermentors and incubators and are suitable for particle removal from air and gases. 8 Clarification of cultures Syringe filters for smaller volumes Whatman GD/X syringe filters Whatman GD/X syringe filters contain multiple glass microfibre prefilters in addition to a layer of PES membrane. Filtering suspensions that contain a high loading of particulate matter becomes easier and a higher sample volume can be filtered than with standard syringe filters. 9 Capsule filters for larger volumes Polycap TC capsule filters Critical filtrations of virus suspensions and cell culture media require outstanding products that do not contaminate the sample. The PES filtration membrane, together with the test methods used during the manufacturing process, ensure that Polycap TC capsule filters are one of the most suitable capsules for these applications. Polycap TC capsule filters are disposable, dual-layer PES membrane filtration capsules that provide efficient filtration for critical aqueous solutions. The PES membrane is inherently hydrophilic, has excellent flow rates, has low extractables, is biosafe, and exhibits low protein binding. Polycap TC 36 has a filtration area of 400 cm 2. 10 Cell observation Critical optical measurements Glass bottom microplates Optically clear glass bottom microplates are suitable for use in a wide range of critical optical measurement techniques including enzyme studies, cell-based assays, and DNA-protein interaction. The plates are available with a choice of clear or black polystyrene plate bodies that have a premium quality slip glass of a uniform mm thickness. The uniformity of the glass is important to ensure a uniform background for all wells in the plate. Black plate bodies isolate the excitation or emission energy to the individual wells so that there is no interference from well to well. This can be important for sensitive probes, light-sensitive cells, and background subtraction. A further modification to the plate, which enhances its use for these applications, is the inclusion of a low profile skirt. Normally the microplate skirt lifts the well base slightly. In this case the skirt isolates the wells from outside light energy while allowing direct contact of the glass bottom with the optics of the microscope or reader. 11 Cell growth and observation Nuclepore membranes Cells can be readily grown on a matrix. This matrix needs to be a solid substrate that does not interact with cell growth and permits easy transportation of liquid throughout the substrate into the cells. A suitable matrix for this application is a polycarbonate membrane such as Nuclepore. Nuclepore membranes are manufactured without the use of surface tension improvers, so extractables are low and do not interact with cells during the growth process. Nuclepore membranes have a smooth and flat surface offering good visibility of cells during and after cell culture. Simply place your Nuclepore membrane on the culture media and spike your cells on the surface. Fig 16. Nuclepore membrane.
