Both cdna synthesis and PCR reactions are performed in a single tube using sequence-specific primers corresponding to the target RNAs from total RNA or mrna. The Enzyme Mix in this kit is an optimized blend of HelixCript TM Thermo Reverse Transcriptase and a specially designed automatic hot-start version of PCR enzyme, and ensures reliable results in terms of sensitivity, accuracy and specificity. The 2x Buffer Mix contains all of the required components including optimized buffer components, Mg 2+, dntps, and RNase inhibitor. Two types of qrt-pcr kits are available. qrt-pcr [Intercalator type] is dedicated for SYBR Green detection, and qrt-pcr [Probe type] is for the detection using sequence-specific probes. ROX passive reference dye is also provided with separated tube for normalization of the fluorescence signal. 2 qrt-pcr Kit [Intercalator type] Protocol 1. Program a real-time PCR instrument as follows in order to synthesize cdna and PCR amplification. The excitation and emission maxima of the included SYBR Green I are at 494 nm and 521 nm, respectively. <2-step cycling protocol> If annealing temperature(at) of primers used in real-time RT-PCR is between 58 and 62, thermo cycling can be performed using 2-step cycling protocol for PCR Amplification Step as follow. Add following components for a single 50 μl reaction volume.* Components RNA Template (1 ng ~ 1 μg)** 2x qrt-pcr Buffer Mix [S] qrt-pcr Enzyme Mix [S] Forward primer (10 pmoles/μl) Reverse primer (10 pmoles/μl) ROX Passive Reference Dye (Optional) DEPC-treated water Volumes X μl 25 μl 1 or 0.1 μl*** Adjust to final 50 μl * For multiple reactions, prepare a master mix by adding the required volumes of each above components (except the RNA template) and dispense appropriate volumes into PCR tubes or plates. * The reaction volume for a reaction could be adjusted according to the recommendations for the instruments. ** Recommended amount of RNA : Total RNA, 10 ng ~ 1 μg, Enriched mrna, 1 ng ~ 500 ng *** See the following table for amount or concentration of ROX Passive Reference Dye for your specific instrument.
