16 What is TGF-β Transforming growth factor beta (TGF-β) is a family of cytokines first identified in 1978 and is now known to be highly conserved throughout the animal kingdom. There are 3 isoforms of TGF-β (TGF-β1, TGF-β2, and TGFβ3) secreted in virtually all cell types. Because TGF- β is involved in a wide range of biological processes, the mechanism of TGF-β signaling in both extracellular and intracellular interactions are studied in immunity and diseases such as cancer, cardiovascular, and fibrosis.

17 TGF-βrelated research Cancer Cancer is a disease characterized by uncontrolled growth and proliferation of abnormal cells. It is now known that certain cancers evade the immune system by secreting TGF-β into the extracellular matrix to hide their presence from the cancer killing immune cells. Thus, making TGF-β an attractive therapeutic target to treat the disease. Inflammation and Immunity Small concentrations of TGF-β activates monocytes, stimulating the synthesis of a variety of cytokines (such as IL- 1β and TNF-α), chemokines (like MCP-1), and growth factors (including basic FGF and PDGF-BB) increasing integrin expression. In contrast to the activating effects of TGF-β on blood monocytes, its actions on mature macrophages are predominantly suppressive. Measuring TGF-β along side cytokine molecules will be common in this area of research. Cardiovascular After a heart attack, members of the TGF-β family are strikingly induced and rapidly activated. Understanding the exact role of TGF-β is important in order to design novel therapeutic strategies targeting the TGF-β signaling cascade in healing tissue affected by loss of blood supply and cardiac remodeling Fibrosis TGF-β is an essential growth factor that regulates cell proliferation, differentiation and the extracellular matrix formation in the wound healing process. When dysregulated, it can result in excessive scarring and tissue fibrosis that can lead to organ failure and death. Currently, a significant unmet medical need exists for safe and effective antifibrotic therapies to treat fibrotic diseases of the lung, liver, kidney, eye and cardiovascular system.
