3 The History of The Binding Site The Digital RID Reader is designed to simplify & speed-up the process of reading precipitin radial immunodiffusion rings. The Binding Site is built on the firm foundations of scientific evidence and a flexibility that allows us to respond to the needs of clinicians and researchers alike. Radial Immunodiffusion is a well-established technique based on the complex formed between antigen and antibody, producing a visible precipitin ring in the gel. Since the early 1970 s, we continually strive to expand our knowledge and understand the challenges facing patients and medical professionals in our industry. The concentration of specific proteins can be determined efficiently and accurately by measuring the precipitin ring diameter using our RID Reader. The Binding Site provides a solution to determine the precipitin ring diameter using the RID Reader or a jewellers eye piece. Using a range of quantitative calibration methods, the RID Reader provides a diameter reading tailored for each assay kit. 53 4

5 Bringing Simplicity to a Proven Science The Binding Site RID Reader provides you with 3 methods of Calibration Curve. This allows you total control and confidence in the concentration generated. The linear curve fit allows reading for RID plates that have been allowed to run to their stated completion time before reading the rings. The point-to-point curve fit is an alternative to the second order curve fit, for non-linear curves. The points of the curve are joined by straight lines. The second order curve fit allows reading when RID plates are to be read prior to their stated completion time, producing a binomial curve. The second order (binomial) curve fit is used to calculate a non-linear curve. 7 8

7 Complement The Complement system is a complex part of the immune system comprising of numerous proteins which act as a cascade. These assays are efficient as screening tools to detect complement deficiencies. Complement is involved in initiating an inflammatory response and destroying certain bacteria and viruses. Where a complement deficiency is suspected it maybe necessary to test for the specific components of the complement system. Immunoglobulins are a critical part of the immune response by recognising and binding to a particular antigen. Determination of individual subclasses is also relevant in assessing primary immunodeficiency or immune responses. 9 Human Complement Functional Assays Functional assays are effective as screening tools to detect complement deficiencies and aid in the monitoring of total complement activity. These assays are based on the haemolysis of red blood cells following activation of the complement system. Human Coagulation Proteins Coagulation proteins are the core components of the coagulation system. Coagulation testing is carried out to determine if an individual has enough coagulation activity to control the blood clotting process and to measure the function or amount of these proteins in the blood.
