8 Sample requirements and shipping information (solid biopsy) Only one tumor block will be sent and preferably the block that has been used to determine the diagnosis (if there is enough material) or the most representative block of the patient s diagnosis. If several blocks are sent, the process will be interrupted until the most representative block of the patient diagnosis is identified. A sample will be considered as valid if: 1. The percentage of tumor cells is > 10% of the entire sample 2. The size of the region where the tumor cells are located is > 5mm 2 3. The percentage of lymphocytes is not too high (<20% in the region where the tumor cells are located) Shipping instructions: When a tumor block is sent a) Add a code sticker identifying the block. b) Place the tumor block in the small transparent bag provided and that has a barcode sticked. c) Place the block in the OncoSTRAT&GO Kit. d) Attach the corresponding Pathology Report, BioSequence Application and Informed Consent completed. When sheets are sent Please note that for technical reasons it is preferable to receive FFPE block. The unused remains of the block will be sent back at the end of the process. a) Prepare the tools for H & E and unstained sheets.. Use a new microtome blade for each case. Wash the bath water between cases Wear gloves and change them frequently to prevent contamination of the samples. b) Prepare slides of 5 µm. c) Make sure that sections of each layer are similarly oriented. d) Do not place coverslip on slides unstained. e) Insert the blades into the containers included in the OncoDEEP Shipment kit. f) Add a code sticker identifying the slides containers. g) Attach the corresponding Pathology Report, BioSequence Application and Informed Consent completed. This treatment degrades DNA. Recomendations of the.... processing: All FFPE samples must be fixed in neutrally buffered formalin 10% for 30 minutes after surgical excision. The time between excision biopsy and fixation should be as short as possible (less than 15 min). The optimal time for fixing is 24h in formol (between 18 y 24 hours max) and then be soaked in paraffin IHC-grade. Important Please make sure that the above criteria are carried out. 9 Sample requirements and shipping information (liquid biopsy) The blood samples will be drawn in the Streck tubes (placed inside the plastic tubes identified with a code) that contain a solution that stabilizes the DNA at room temperature. The extraction protocol is the following: a) Remove the blood by venipuncture according to CLSI H3-A6 recommendations b) Fill two tubes Streck completely. c) Remove the adapter tube and immediately mix by gentle inversion between 8 and 10 times. Inadequate or delayed mixing may result in inaccurate test results. d) After the drawal, enter the Streck tubes in the plastic tubes. e) Place the tubes in the small bubble envelope we provide. Then place this envelope in the kit the same way it came. NOTE: Perform the blood extraction from Monday to Tuesday to ensure the arrival of the sample by the laboratory in the same week of the extraction Please make sure that the above criteria are met.
