Overview In the demonstration of tissue antigens by Immunohistochemistry, the evaluation of specific staining is sometimes impeded by the presence of endogenous biotin, which may lead to an erroneous interpretation or false positive results. This blocking system is intended for use in immunohistochemical staining to block nonspecific staining due to endogenous biotin during the IHC detection of antigens. Incubate tissue sections with avidin block for minutes Wash 2 times with buffer Incubate tissue sections with biotin block for minutes Wash 2 times with buffer Apply primary antibody to the tissue sections ab64212 Endogenous Avidin/Biotin Blocking Kit 3

4 2. Materials Supplied and Storage Store kit at 4 C immediately on receipt and check below for storage for individual components. Kit can be stored for 1 year from receipt, if components have not been reconstituted. Aliquot components in working volumes before storing at the recommended temperature. Avoid repeated freeze-thaws of reagents. 8 6. Assay Procedure 6.1 Blocking/staining protocol: 1. Incubate tissue sections with avidin block for minutes. 2. Wash 2 times with buffer. 3.
