1 The right plate for your appliation Choosing the right plate is the first step to a suessful experiment. We offer a wide range of plates to support different appliations and levels of throughput. The following information will help you hose the most suitable plate material, olour, base and shape: Frame Materials Polypropylene has very low biomoleular binding harateristis, a high temperature tolerane and resistane to many standard laboratory hemials, inluding DMSO. It is mainly used for storage appliations. Polystyrene is a hard material with optial larity. It is used for ell ulture, fluoresene, luminesene, ELISA and homogeneous assays. Well shape Round wells are suitable for most appliations as they redue droplet effets and wiking. Square wells ensure the best use of spae and improve sample mixing partiularly when used for baterial ulture growth. Plate olour Clear plates offer the best solution for absorption, Elisa, spetrophotometri and storage appliations. Blak solid base plates are reommended for top reading fluoresene instrumentation beause of their low bakground and minimised light sattering. Blak lear bottom plates are suitable for bottom reading instruments and mirosopy appliations. White plates give the best results for luminesene detetion beause they maximise signal intensity. Well bottoms Flat bottoms are most suitable for optial imaging and ell ulture appliation while U-bottoms are better for washing, mixing and pelleting and give high surfae area. Conial V-bottoms are best for preipitation, entrifugation and small volume reovery. 84

12 Vision Plates Vision Plates are our best plates for your most advaned appliations. When visualising fluoresent signals from anything between a single moleule to a whole ell, Vision Plates will give aurate and onsistent results. Suitable for ell based high ontent sreening, onfoal mirosopy, FRET and homogeneous assays, results show an optimum signal to noise ratio and a high onsisteny essential for automated high throughput. Using state-of-the-art manufaturing tehnology 4titude have developed a produt whih offers key advantages to the end user. Laser Welding Tehnology 4titude s Vision Plates are assembled using unique patented laser welding tehnology. The use of loalised welding heat dramatially redues base film distortion during prodution. This improves base flatness, whih in turn redues instrument auto-fousing time. Optial Quality of the Polymer Film The lear base omponent of our Vision Plate demonstrates superior properties in terms of optial larity (low absorbane and high transmission), low bakground fluoresene and onsisteny of material thikness. The latest extrusion tehnology is used for manufaturing an ultra lear base of 190µm thik, to provide optimum results with onfoal mirosopy and laser based detetion systems. Variation aross the plate is minimised so the time required for omplex sreening appliations an be redued dramatially. In addition, ell plate types are available with a 700µm moulded base. 13 Redution of Autofluoresene 4titude s Vision Plates are assembled using unique patented laser welding tehnology whih redues autofluoresene and does not inhibit ell growth. Other manufaturers assemble lear base miroplates by gluing a lear film to the frame or heat-welding the omponents together. Both tehniques an ause problems with ell growth and the subsequent mirosopi or fluorometri analysis. Heat welding of the two plate omponents under high pressure results in autofluoresene at the well edges, alled halo effet. Gluing uses organi solvents within the adhesives, whih an also ause autofluoresene. These solvents an also have ytotoxi effets, whih may lead to inhibition of ell growth or even ell death. Additionally, inomplete glue lines or weld lines often result in well-to-well leakage. 14 Improved Cell Adhesion Polymers, suh as polystyrene are very hydrophobi and need to be surfae treated to inrease wettability and to allow for ell adhesion. To introdue the neessary harges to the plasti surfae, sreening plates are ommonly undergoing orona treatment or similar low ost modifiations. The drawbaks of suh treatments are they are partly reversible and the density of the harges varies aross the surfae area, resulting in uneven ell growth and a short shelf-life of the produts. 4titude uses a unique low pressure plasma proess for treating the plasti surfae whih produes a more onsistent and stable surfae. The treatment has been espeially designed to improve ell adhesion under diffiult onditions (e.g. redued serum onditions). It is also useful for growing ells with low adhesion properties suh as transfeted ells overexpressing proteins. 4titude offers oatings with Collagen 1 and Poly D-Lysin. Collagen 1 Collagen 1 is a protein of the extraellular matrix (ECM), an interellular substane whih, in vivo, influenes adhesion, migration and proliferation among other proesses. Collagen 1 is one of the most important ECM proteins for in vitro ell ultures. Diffiult to ultivate ells adhere to Collagen 1 and display positive growth. Collagen 1 an also influene differentiation and morphology. Poly-D-Lysin Poly-D-Lysin is a syntheti moleule that improves the adhesion of different ell types to polystyrene surfaes. 20 Mimetix TM saffolds for 3D ell ulture: Mimetix TM saffolds are highly porous and mimi the extraellular matrix, providing an ideal environment to support the growth of ells in 3D. They are reated by eletrospinning the medial grade polymer poly-l-latide (PLLA) into fibres. The Mimetix TM saffold is highly onsistent well-to-well with respet to fibre diameter and pore size, resulting in exellent reproduibility of ell-based assays. Mimetix TM has been validated with a number of primary ells, ell lines and stem ells and is available in a range of ready to use, sterile laboratory onsumable formats. Mimetix TM saffold resembles the extraellular matrix. Why Use 3D Models in Drug Disovery? Cells grown in a 3D miro-environment look and behave more like ells in human tissues than those ultured in 2D.
