2 Label-free Bio-Affinity Analysis Surface Plasmon Resonance (SPR) is an established tool in the life science sectors. It offers a new generation of label-free biomolecular analysis, providing information on kinetic processes (association and dissociation), binding affinity, analyte concentration and real time molecule detection. A large variety of bio-interactions can be monitored, such as antibody/antigen, peptide/antibody, DNA/DNA, antibody/bacteria etc... It measures modifications of the refractive index at the surface of the biochip, which can be correlated to mass variations. It detects interacting molecules in real time to determine analyte concentration and to characterize the biomolecular affinity. Biomolecular interaction After the injection of the sample solution, several hundred different molecular affinities can be monitored in real time. Principle The phenomenon of SPR occurs when light interacts at the interface between the biochip s metallic surface and the liquid medium.

5 Biomolecular Applications SPRi allows rapid generation and analysis of numerous simultaneous molecular interactions. Thanks to SPRi, many types of ligands and analytes (peptides, proteins, nucleic acids, carbohydrates, bacteria, cells,...) that are involved in life-science mechanisms can be monitored as never before in real time. Also, potential targets found in complex mixture solutions such as serum, plasma or cell cultures can be injected directly through the fluidic system. Carbohydrate-Protein Interactions The parallel detection of carbohydrate-protein interactions provides information on glycobiology research and it helps to understand oligosaccharide-protein complex formation in biological systems. DNA-DNA Interactions Label-free SPR imaging enables detection of DNA point mutations. In addition the effects induced by the variation of one or several parameters can be compared in a single run. Protein-Peptide Interactions SPRi allows to monitor clinically related protein-peptide interactions in real time. It is a powerful technique for pharmaceutical and medical screening and for basic research applications.
