Experience the speed, simplicity, and scalability of semiconductor sequencing with the Ion PGM sequencer. Ion Torrent has pioneered an entirely new approach to sequencing. Instead of using light as an intermediary to sequence DNA, Ion Torrent uses semiconductor technology and simple chemistry. Ion Torrent translates chemical signals into digital information, much as a CMOS chip translates photons into pixels. The time was reduced approximately threefold in just six months, thanks to key breakthroughs at each stage of the workflow: The new Ion Xpress Fragment Library Kit offers a novel enzymatic fragmentation process that has reduced library preparation to 3.5 hours, 60 percent faster than six months ago. The Ion OneTouch System* for template preparation, shipping in the third quarter, has added a component to automate enrichment, the Ion OneTouch ES.* Now the entire template preparation process is fully automated and can be completed in three hours, 70 percent faster than six months ago. The new Ion 316 chip has a two-hour run time for 100 base pair reads, just like the Ion 314 chip. Run times are reduced to less than one hour when sequencing short reads. The new Torrent Suite 1.4 analysis software cuts analysis time to just 30 minutes on the Ion 314 chip and one hour on the Ion 316 chip, while also delivering a significant improvement in base-call accuracy. 4 nothing gets better faster than Ion semiconductor sequencing Six months to 10-fold improvement In the six months since the Ion PGM sequencer launched, its throughput has increased tenfold, its workflow has become the fastest in the industry, and it s now the fastest-selling sequencer in the world, with placements in more than 40 countries. In the second half of the year you ll see more of the same. Read lengths will double to 200 base pairs and throughput will increase by tenfold again. All of these advances are a direct result of Ion s semiconductor sequencing technology and native chemistry, which make sequencing faster, simpler, and more scalable than ever before. ION RAPIDLY IMPROVING ACCURACY More Throughput, Better Value The scalability of Ion semiconductor sequencing will enable a hundredfold increase in throughput this year. The new Ion 316 chip has 100 Mb of throughput, tenfold more than the original Ion 314 chip. The Ion 318 chip, available in the fourth quarter, will deliver 1 Gb of throughput. Each of the three chips will work on the Ion PGM sequencer, giving users the flexibility to buy only as much throughput as they need for an experiment, from 10 Mb to 1 Gb. Ion semiconductor sequencing offers exceptional value as well as performance. The Ion 314 chip is now available for just $99 USD, which enables the sequencing community to develop novel methods and applications. Ion Torrent is able to lower the price because of the significant sales volume generated by the Ion 314 chip and the economies of scale inherent in semiconductor manufacturing. Better Accuracy The Ion PGM system s accuracy has improved by an order of magnitude since it was launched six months ago, as a result of the system s speed and simplicity. Ion Torrent has capitalized on the inherent accuracy of its simple sequencing chemistry, achieving more signal, better signal processing and improved base calling. Accuracy improved quickly in part because the Ion PGM sequencer s two-hour run times enable Ion Torrent R&D teams to quickly test and refine ideas. Improvements in accuracy will help Ion Torrent reach 200 base pair reads in the second half of this year and 400 base pair reads in 2012, making many new applications possible for researchers. More New Products Additional new products now available include an early-access version of the Ion DNA Barcoding 1-16 Kit* for multiplexed sequence analysis, the Ion Library Quantitation Kit for TaqMan - based quantitation of Ion DNA libraries, and the Ion Sphere Quality Control Kit for template bead quality control. The Ion PGM Sequencer in Customers Hands Scientists turned to the Ion PGM sequencer to identify a deadly strain of E. coli in Europe in June because the system s two-hour run time was faster than anything else on the market. Scientists at BGI sequenced the bacterium within three days, and a team at the University of Münster sequenced the strain and developed a field-deployable test in just five days. Scientists around the world also have reported exceptional throughput results with their first runs on the Ion 316 chip. Early-access users at Baylor College of Medicine recently broke Ion Torrent s internal record for highest output, after reporting 287 Mb from a single Ion 316 chip run, and the Broad Institute reported up to 250 Mb. Most recently, the Wellcome Trust Sanger Institute reported that one of the first runs of an Ion 316 chip on their Ion PGM sequencer produced a total yield of 194 Mb.
