3 Two Ways To Get There Mutation Discovery (Amplicon Scanning) Mutation Scanning Data Analysis Four distinct groups can be identified by the change in the shape of the curve and by the difference compared to the wild type. Mutations in PCR products are detected by the presence of heteroduplexes optimally detected by our LCGreen Plus dye. Variations are identified by changes in the shape of the melting profile compared to a reference sample. Hi-Res Melting on the LightScanner is also being used for mutation discovery in tumor samples, yielding information on mutations present in low allele fractions (as low as 5%) and allelic imbalances between tumor and matched normal samples. Genotyping Genotyping with small amplicons can be successfully performed for SNPs, small deletions and insertions that are located between the 3 ends of the forward and reverse primer. No probe is required for difficult base pair neutral changes (A>T, G>C). The High Sensitivity Master Mix can overcome challenges in detecting subtle differences between these homozygous alleles. Heterozygous Double Heterozygous Homozygous G allele C Variant Single Heterozygous Wild Type Genotyping with unlabeled LunaProbes is a homogeneous end-point assay easily accomplished using the LightScanner System. The probe is included in the PCR reaction but is not consumed during amplification. Genotyping is accomplished by monitoring the melting temperature of probe-target duplexes post-pcr. G allele Heterozygous C Variant The fully matched probe-target hybrid (C variant) generates a melting peak with higher Tm (shown in blue), while the mis-matched hybrid (G allele) generates a peak at a lower Tm (shown in gray). The heterozygous samples demonstrate a profile with peaks at both Tm s, representing the presence of both alleles (shown in red).
