4 IDEXX BioResearch 3R Advantage Program Environmental Purpose: improve Detection possibilities for agents not well transmitted to sentinels Method: Real-Time PCR Real-Time PCR is sensitive - detects as little as 1-10 molecules Specific can distinguish between related microbial species Infectious agents don t have to be viable/cultivatable Easy sample collection and no need for special storage conditions Easy to ship; fast results Steps For direct environmental sample - Swab the plenum or exhaust air filters. - Alternatively, directly swab the interior of the cage(s) - Swab the most 'dirty' places. - Place the swab into tube labelled "Environmental sample". For cage filters Option 1 - Place a piece of the filter (max. 2 cm x 2 cm) into the big tube labelled "Cage Filter". Option 2 - Swab the filter and place the swab into tube labelled "Environmental sample". For plenum and AHU sampling For capturing devices specifically designed for environmental testing (eg.tecniplast Interceptor or Allentown Sentinel ) please follow instructions provided by the supplier and send the devices to IDEXX BioResearch for testing. 4

5 Faeces Purpose: Detection Real-Time PCR sampling detects parasites, bacteria and viruses in faeces. Real-Time PCR is sensitive - detects as little as 1-10 molecules Specific can distinguish between related microbial species Infectious agents don t have to be viable/cultivatable Easy sample collection and no need for special storage conditions Easy to ship; fast results Steps Collect 2 faecal pellets; either directly from the animal or from the cage, using decontaminated forceps Place faeces into labelled faeces sterile tube To identify the exact source, the faeces should be collected directly from the animal 5

6 Blood Purpose: Antibody Screening Serology testing screens for antibodies produced as a result of infection by viral, bacterial and fungal pathogens Opti-Spot We ve revolutionized serology testing by simplifying rodent blood collection now it s easy and fast. IDEXX BioResearch s Opti-Spot collection method has reduced the amount of blood required for serology to only a few drops providing an easy and time-saving method to collect serology samples. 7 Steps Technique: Tail Venipuncture Place mouse on wirebar lid Hold tail base Remove lancet or 25 gauge needle from package Lift tail to expose ventral tail vein Poke vein with needle/lancet Blot 2-3 blood drops onto the circled area of the Opti-Spot strip (blood spot ø at least 6mm) Allow spotted Opti-Spot card to dry Other blood collection techniques that work well for Opti-Spot include: Lateral Saphenous Venipuncture Facial or Temporal Venipuncture Note: We recommend to label the Opti-Spot cards before collecting the samples 7

8 Fur Sample Collection Purpose: Detection Real-Time PCR sampling detects ectoparasites and cutaneous bacteria Real-Time PCR is sensitive the most recommended test for the detection of fur mites. Specific can distinguish between related microbial species Samples easy to collect and store no need for special storage conditions Easy to ship Fast results Steps Place mouse on wirebar lid and gently restrain Rub over the fur against the direction of the hair coat Swab the face, back, around tail base and belly It s important to swab all areas noted Place swab tip into labelled fur sterile tube 8

9 Oral Sample Collection Purpose: Detection Real-Time PCR sampling detects most respiratory bacteria and viruses Real-Time PCR is sensitive detects respiratory microbes such as Mycoplasma and Pasteurella pneumotropica in the oral cavity. Specific can distinguish between related microbial species Infectious agents don t have to be viable/cultivatable Samples are easy to collect and store no need for special storage conditions Easy to ship Fast results Steps Restrain the mouse with one hand so that the head is not able to move from side to side Insert the oral swab starting at the corner of the mouth to encourage the mouse to open its mouth Slowly twirl the swab to collect the sample Place swab tip into the labelled oral sterile tube 9

10 Packing & Submission Fur, Oral and Faeces Samples Package according to sample type in sealable plastic bags one bag each for fur, oral and fecael samples. Label bag with sample type and submitter name. PCR samples are stable and can be held at room temperature if packaged the same day or stored in the refrigerator until ready for packaging the next day. Blood Samples Allow Opti-Spot strips to dry for al least one hour at room temperature. Package into labelled plastic bag with a desiccant packet (1 packet per 50 strips). Shipping Place all plastic bags in a cardboard mailer or styrofoam box and ship to our laboratory by overnight courier (address below), or contact idexxbioresearch-europe@ idexx.com to help you to arrange a courier collection.
