2 OBJECTIVES: General Sampling Guidelines Chain of Custody Essentials Then Q&A

3 GENERAL SAMPLING GUIDELINES 1. Representative sampling different for each matrices food, water, environmental 2. Sampling Equipment be flexible & practical 3. Maintain sample integrity sampling plan, labelling, temperature control, packaging, time to lab, 4. Collect enough sample to cover all analysis parameters 5. Consistent techniques training your staff reduces sampling variability 6. Frequency of sampling regular intervals needed to trend patterns

4 GENERAL SAMPLING GUIDELINES 1. Representative sampling matrices: food, water, environmental Food Send in original packaging when possible Sample aseptically into leak-proof wide mouth jars or sample bags (food grade or sterile) Weight needed is dependent on # test parameters (100 to 800 gm) Send separate packages for chemistry and microbiology to avoid subsampling Water Use sterile microbiology water bottle 250 ml (contains NaThio to neutralize Cl) Processing waters can use well sealed bags with wire closures. Chemistry needs many bottle types (consult lab)

5 GENERAL SAMPLING GUIDELINES Environmental Samples Surfaces - stick swabs, allergen swabs, sponge swabs and contact plates Air - settle plates, measured air volume cubic metre (m 3 ) plates ie) MAS-100

6 2. SAMPLING EQUIPMENT BE FLEXIBLE & PRACTICAL FOOD GRADE PLASTIC ZIP-LOCK WORKS FOR BULK FOOD SAMPLES SEAL SETTLE PLATES BEFORE PUTTING IN BAG/BOX BASIC HYGIENE WASH HANDS AND THEN GLOVE NEW CLEAN GLOVES.

7 GENERAL SAMPLING GUIDELINES 3. Maintain Sample Integrity Environmental - Where to sample? Divide your facility in Zones. Zone 1 & 2 have highest FS risk level.

8 CREATE ROTATIONAL SAMPLING PLAN REGULAR INTERVALS: Zone 1: Food Contact Surfaces FCS (conveyors, knives, rollers, cutting boards, overhead pipes) Adjacent - Nonfood Contact Surfaces NFCS (floor area, safety guards, shelving, handwashing sink) Zone 2: Building NFCS Support areas Workstations, lunchroom, locker room, warehouse, supply carts PREPARE order supplies & forms, write a location list, pre-label, let lab know volumes & timetables to minimize time to results.

9 3. MAINTAINING SAMPLE INTEGRITY During processing: Settle plates & air samplers, stick swabs, wash waters Sponge swabs in adjacent areas only as need to clean after sampling. Post Clean: Pathogens 1 sponge swab/pathogen (DE broth neutralizes cleaner), Dry swabs or sponge swabs SPC/YM, Contact or settle plates Cleaning is needed after sampling for sponge swabs, agars* Record time settle plates opened-closed (4 hours max)

12 SAMPLE INTEGRITY TEMPERATURE CONTROL & PACKAGING Best practices: Keep samples in fridge <10C & send to lab same day as sampling (24hr) Send to the lab in coolers with ice packs/ice - maintain conditions Enclose paperwork that matching samples & specifying test parameters

13 GENERAL SAMPLING GUIDELINES 4. Collect enough sample Sample both FTC, and NFTC areas & critical Zones 1&2 One swab per Pathogen type Listeria species, EcoliO157:H7, Salmonella sp, generic E. coli One shared swab for SPC/YM/Staph ok Sample the air settle plates or plates from air samplers

Consistent techniques Use a rotational sampling plan with regular intervals (* site dependent) Train your staff to use sampling equipment to reduce variability Avoid composite site swabs if using & have positive result need to trigger resampling of individual sites to find source.

Frequency of sampling Regular monitoring data will enable patterns to be trended Frequency is site dependent (daily, weekly, monthly) *Risk based Ie) RTE facility has high risk control by sampling as visual inspections cannot detect microbial conditions Ie) Closeness of contaminated areas to processing line increases Risk

18 CHAIN OF CUSTODY ESSENTIALS: Four Main Areas: 1. Client Contact Information 2. Sample Information 3. Test Parameters 4. Tracking Info (Sampler/Shipper/Receiver)

20 1. CLIENT CONTACT INFORMATION Who gets the report? Where does the COA report get sent? How paperless/ /fax or snail mail? Who gets the invoice? This section minimizes delays in result reporting which increases the effectiveness of your environmental sampling. Responsiveness

SAMPLE INFORMATION # Samples list one per row Sample Description (Matrix-Location) Time/Date/Sampler - This information can be in shorthand on the COC and can attach the detailed #,descriptions, times that are needed for the report - Remember this info needs to link directly back to the label on your samples - Otherwise, Lab Receiver can t match sample+info and anomaly triggers delay

22 3. TEST PARAMETERS If the form has prefilled headers resist checking off all tests Multiple methods may exist for the same testing parameter. Contact the lab ahead of time if certain methods are required & know the minimum TAT. TC/Ecoli MFHPB-34 Petrifilm 48 hrs SPC generally 48 hrs all methods Y/M 3 day preliminary with 5 day final most methods Listeria 3 days minimum (screening via PCR & cultural confirmation+) Salmonella 2 days minimum (screening via PCR & cultural confirmation+)

24 4. TRACKING INFORMATION Helps if questions arise by IDing Who gave the sample to courier? When did the courier receive the sample? What time did it arrive at the lab? This represents the Chain of Custody (COC) of the sample on a test request form. Helps trace integrity of the sample cold chain if logger in box or at least at the beginning & end of the journey.
