2 Automated GPC-System, Tye VARIO Main characteristics Benefits for the user The rocessing sequences for the samle rearation for esticide analysis are described within the AOAC method No , USEPA SW-846 method 3640A, EN 12393, EN 1528 and within the L method in accordance with 35 LMBG. After extraction, these methods usually require the clean-u of the extract via a gel ermeation chromatograhy (GPC) system. LCTech have develoed a suitable GPC-system for this rocessing ste. The system, however, can be utilised in many different areas, for examle in universities or in R&D of industry. This has been realised through the remarkable flexibility of hardware and software emloyed in the system design. Flexible... The GPC-system, tye VARIO, allows for variable set-us, for examle with regard to the column size used, injection volume, flow rate and other imortant arameters. The samle carousel of the combined autosamler/fraction collector can be easily exchanged. Consequently, either more samles with smaller main fractions or fewer samles with larger main fractions can be rocessed. Samles can be transferred quantitatively. The samles are rovided in 5 ml vials (rack P60-5), which are rinsed additionally to assure a comlete transfer. Easy to oerate... Terminal Great effort has been ut into the design to make it uncomlicated and easy to understand and oerate. Via menu-driven terminal inut, all imortant arameters can be defined. To facilitate individual samle rearation, each single samle can be arameterised and saved as a method. U to 97 methods can be stored. Parameterisation is erformed via terminal inut on the system with automatic data storage. Desite the large flexibility, rocessing of samles or samle sequences may be defined within only a few minutes. The system is initiated when switched on. 3 Robust for oeration without suervision To warrant 24-hour-oeration without suervision, all comonents have been designed to be very robust. All arts wetted by solvents are solvent resistant; materials used are glass, PTFE, stainless steel and PPS-E2. All solvent carrying ies are visible and therefore easily insected and likewise easily exchanged. As a benefit of the robust design, no daily or weekly service needs to be carried out. User rotection... The system design is articularly considerate of user rotection. Samle vials and recetacles for fractions can be (but don't have to be) sealed with seta, such that solvent evaoration is reduced to a minimum. For this reason, the GPC can be oerated indeendent of an exhaust hood. In addition, sealing with seta revents evaoration and, consequently, the unintended concentration of the samles rior to their rocessing.... and just in case the software udate or suort from LCTech is quickly and inexensively available via modem. 4 Processing Sequence and Configuration Comrehensive automatisation according to individual requirements The automated GPC-system, Tye VARIO by LCTech comrises the following stes: Suction of the samle out of a closed vial Transfer into a samle loo Clean-u via the GPC-column and Filling into a closed container with the otion of fractionation in u to 26 vials Flexibility through modular system and intelligent software The GPC-system contains all necessary comonents, which include a high quality LCum, a combined autosamler/fraction collector and a GPC-column (unfilled). Process engineering and safety engineering of all relevant comonents have been undertaken in such a way that unattended over-night oeration is ossible. The range of alication of the GPC-system can be easily adjusted to the analytical needs by choosing the aroriate carousel rack. Deending on the caacity of the carousel rack 12 crude extracts (P ) can be rocessed in one fraction of u to 250 ml or 26 crude extracts (P ) in one fraction of u to 110 ml, or 52 crude extracts (P-60-16) in one fraction of u to 60 ml, or 8 crude extracts (P R) in one fraction of u to 250 ml (standard round-bottomed flask), or 12 crude extracts (P-100S-16) in one fraction of u to 100 ml (ointed flask) or quantitative samle transfer 52 samles (P-60-5) quantitatively from 5 ml to 60 ml vials can be rocessed. age 4 of 14

5 Carousel rack With each tye of carousel rack, the timecontrolled division of the main fraction in u to twenty-six fractions can be re-set (volume results from flow rate x time). Overfilling of the vials is automatically revented by an internal lausibility enquiry of the system. Each fraction is filled into a searate vial. Additionally, it is ossible to arameterise unrelated fractions and to vial these searately. Thereby, nearly all thinkable GPC alications should be covered. The carousel racks are easily removed and exchanged through undoing the locking device. Before every sequence, the used rack is queried by the system, which the user confirms by a keystroke. age 5 of 14

6 Handling and System Control Simle handling and fast rogramming The rack is inserted into the system and loaded. The method to be alied to the samles is simly re-selected from the system memory - either individually er samle or, in case of the same rocessing requirements, for the entire sequence. Thereby, the rocessing stes can be defined individually within a very short eriod of time. System arameterisation Parameterisation is conducted via terminal inut on the system followed by automatic storage. Further set-u otions of the GPC-system, Tye VARIO are: Pum flow rate; the system is offered inclusive of a high-quality, rearative or analytical um. Forerun and tailing from s Main run (can be divided into u to 26 fractions) from s, whereby in combination with the set flow rate, a total volume in resect of the container sizes cannot be exceeded. The PTFE-rinsing ort including waste function, the needle rinsing ort and all contaminated comonents are cleaned in the rinsing mode. A cross-contamination can be reliably avoided due to the comrehensive rinsing rocesses. The rinsing rocesses require a minimal amount of solvents. Should a run be interceted (e.g. by a ower-cut), then an integrated rinsing rogram can be called u. The aaratus is cleaned and reared for the next samle searation. 7 Technical Data / System Comonents Measurements and weight, electrical connection 750 x 600 x 1000 mm (W x D x H), ca. 60 kg Connection 1x three-in lug 230 V AC; 50 Hz; CE-certified GPC Pum The system contains a robust SPS-controlled dulex um, which has already roved itself many times to be excellently suited for continuous running. This is either a rearative (to be set anywhere between 0.40 and ml/min) or an analytical (from ml/min) um. The um flow rate can be set, ulsation lies below < 1 %. Automatic cut-off on reaching certain thresholds for overressure or underressure is art of the system standard equiment. The um is solvent resistant and low in maintenance. Chromatograhy Column Refillable, DIN-comliant secial column (unfilled; standard delivery), materials subjected to wetting with solvents are glass and PTFE, 1/8 -PTFE-tube at inlet and outlet site, rigid PTFE-base and adjustable lunger, stable construction, high overressure stability, wearing arts are individually available. If required, any other column may be built in, if it is designed and ermitted for use under ressure of at least 200 si. The column is not filled. Filling with Bio-Beads, Tye S-X3, is recommended. 8 Software VarioSoft (otional) The LCTech PC-software VarioSoft rovides a comfortable solution for the control of the GPC VARIO system. Process arameters can be visually assisted reselected and stored. A samle rocessing list can be created and stored offline as a job list. Facing the control at the system, now an essential extension is the definite correlation of samle identification and job list (consecutive and nonmaniulable samle ID). Protocols of the results can be exorted as ASCI-files. Samle information such as laboratory internal identification, customer data or information regarding samling and samle origin (free text inut), resectively, can be read in via barcode reading. Admission to the system is assword secured via a multi-level authentication hierarchy The Software can also be used for the integration of an UV detector for quality assurance. A fixed wavelength detector (254 nm) is recommended. Simle chromatograms will be generated, that can be used for the evaluation of the system erformance. Windows 98 or higher is required. A free disk sace of about 100 MB will be needed for the software on the hard drive. Fixed Wavelength UV Detector (otional) The UV detector offered by LCTech is otimally suitable for utilization with the LCTech software VarioSoft. It is a 254 nm fixed wavelength detector, which is tuned for the quality assurance of GPC columns. age 8 of 14

9 Combined Samler/Fraction Collector and Injection Port A robot swivel arm with a stable, sring-loaded stainless steel needle and aeration device takes u the samle, injects it into a stainless steel injection ort with needle guide and then conducts fractionation. The carousel racks can be easily mounted and taken off. The solid construction of the racks kees the vials safely in lace. Normally, vials with seta are being used that the user is not exosed to health damaging solvent vaours escaing from oen vials. Samle Loo The sulied standard samle loo is made from PTFE and is set to 5.0 ml (rearative system) or set to 1.0 ml (analytical system). A measuring rotocol is sulied. Other volumes are available on request. The samle loos can be exchanged without any effort. What is next? If the samles need to be concentrated after rocessing, the fractionating vials can be directly transferred into the analytical rearation system EVA III. This system concentrates the samles fully automatically and without the need for suervision to a defined end volume (1.0 ml, 5.0 ml or 10 ml). Solvent exchange may be incororated into this rocess. The exchange is normally conducted in the liquid hase. However, the samles may also be evaorated to dryness and then taken u by a different solvent. age 9 of 14
