Wide range of products for numerous applications MagnaMedics separation solutions are based on a proprietary nano- and micro-bead technology incorporating super-paramagnetic properties into a silica matrix. The proven characteristics of silica are also combined with innovative surface modification methods covering a broad range of separation applications. Our standard products have sizes of 600 nm, 1 µm and 1.2 µm. For most applications the 1 µm beads are ideal. Beads with a mean size of 600 nm give more surface area per mg of bead and a slightly slower separation speed. Beads with a mean size of 1.2 µm are used for genomics and proteomics applications where highest magnetic strength and fast protocols are needed. MagSi-STA efficiently binds to biotinylated molecules, e.g. peptides, proteins, antibodies, sugars and lectins. The magnetic properties enable easy and quick washing steps. Because beads are in suspension, incubation times can be shortened compared to alternative techniques. Binding of molecules onto MagSi-STA is based on the strong non-covalent interaction between streptavidin and biotin (K a = 10 15 M -1 ). MagSi-STA beads are coated with recombinant streptavidin (53 kda) with shortened N- and C-terminus for improved solubility and accessibility of the sites. MagSi-protein A and MagSi-protein G Protein A and Protein G are recombinant proteins that bind with high affinity to the Fc portion of several classes and subclasses of immunoglobulin from a variety of species. Both proteins form reversible complexes with different IgG subclasses and species. MagSi-protein A and MagSi-protein G are ideal tool for efficient and economical purification and immunoprecipitation (IPs) from small sample volumes. Via cross-linking of IgG this product it becomes a tool for directed pull down of desired peptides and proteins. The recombinant protein G (~26kDa) on the surface of our beads contain only IgG binding sites. The albumin, cell wall and cell membrane binding domains, present on the wild-type protein G, have been removed to optimize binding properties. The 42kDa protein A is originating from Staphylococcus aureus. The advantage of MagSi-protein A and MagSi-protein G, compared to MagSi-STA, is that the antibody does not need to be biotinylated and the binding is reversible. The magnetic properties allow easy and quick washing steps, and antibodies are easily crosslinked if interference in downstream processes is expected. This makes the products ideal for protein isolation and proteomics applications. The relatively low hydrophobicity of the C4 beads allows the purification and fractionation of larger biomolecules like proteins. MagSi-WCX and MagSi-WAX MagSi-WCX (weak cation exchange) and MagSi-WAX weak anion exchange are ideal tools for the reduction of protein or peptide complexity (e.g. cell lysates).
