The advances in Proteomics have been possible through the development of advanced high resolution WITA VISION 2-DE systems allowing resolution up to 10000 protein spots of entire cell lysates in combination with protein identification by new highly sensitive mass spectrometric techniques. The knowledge of the human genome and its polymorphisms is the basis for an understanding of the gene functions in cellular processes. However, the data of the genes describe only the static state of the inherited information. To understand the dynamic processes within the cells, e.g. during cell differentiation and regulation or during disease development, it is mandatory to study the proteins involved. Their presence, actual concentrations, and possible modifications must be known in order to gain clues about their functional activities. This means that entire protein extracts of cells or expression pattern of the proteins, among them stress, alterations in temperature, different growth conditions, and the effects of various treatments. COMPARISION OF WITA S NEPHGE TECHNOLOGY WITH IPG WITA uses a NEPHGE (Non Equilibrium PH Gradient Technology for the Gel Electrophoresis) which gives certain advantages over the IPG technology. Separation gradient within the gel is forming by soluble ampholyte during electrophoresis. IPG 1st dimension separation according to IP of proteins. Fixed ph-gradients on pre-casted paper strips.
