1. Introduction Abcam s Gold Conjugation Kit allows antibodies or proteins to be covalently attached to ultra-stable Gold nanoparticles at very high OD quickly and easily. The hands-on time for the Gold conjugation procedure is about 1 minute and the conjugate is ready to use within 30 minutes. While antibodies in their native state do not contain aldehyde groups, it is easy to introduce aldehydes by mild oxidation of the sugar chains located in the Fc domain. Covalent attachment of aldehydemodified antibodies to hydrazide surfaces orients the two antigenbinding arms in space, which can lead to enhanced assay performance. GOLD-Hydrazide Conjugation Kit (40 nm, 10OD) (ab188284) provides all the materials required to conjugate three antibodies in site specific manner to GOLD. Sufficient material is provided for small scale trial conjugations and then scale up of the best reagent. 2

3. Storage and Handling The activated antibody should be used within 2 hours. For longer term storage (weeks) divide the activated antibody in aliquots and store at -80 C to avoid multiple freeze-thaw cycles. 4. Additional Materials Microfuge Tubes (0.5 or 1.5 ml) Microfuge 5. General Guidelines A. Prior to Labeling Antibody must be purified Avoid glycerol or sugars (e.g. sucrose, glucose) Avoid amino acids (e.g. glycine and histidine) Avoid other primary amines (e.g. Tris) Avoid sodium azide and BSA It is strongly recommended that you dialyze your antibody against 100mM phosphate buffer ph 7.0 before using this kit. Stock antibodies at concentrations of >1mg/ml are recommended; as this allows potentially interfering substances 4

in the antibody preparation to be diluted out (see Step 6.2). You should pay particular attention to the composition of the stock antibody if you require less than a 1 in 5 dilution in step 6.2 of the protocol. If you are in any doubt about the suitability of buffers/additives in your preparation of antibody please contact our technical team for advice. B. Activation of Aldehyde-Antibody Adjust (if required) the concentration of the antibody in phosphate buffer to 1 mg/ml using the antibody diluent provided in the kit. Add 100 µl (1 and 3 reaction optimization kit) or 300 µl (validation kit) of the antibody into the vial of GOLD-Hydrazide Antibody Oxidation Reagent. Mix gently and incubate for 30 minutes in the dark at room temperature. Meanwhile proceed to desalting procedure (Section C., below). C. Desalting Procedure Use one column per antibody. The columns are designed for single use. Discard after use. 1. Secure a separating column in a vertical position. Remove the two caps (remove the upper one first) and allow the storage liquid to flow through the column to waste. 5

2. Equilibrate the column with the GOLD-Hydrazide Wash/Coupling Buffer by filling the column up to the very top and allow the liquid to flow through under gravity. Discard the flow-through. Repeat a further 4 times. When the last portion of buffer has passed into the column, move to the next step. 3. After the oxidation reaction (Step B above) add all the activated antibody to the top of the column and allow the liquid (i.e. ~100 µl or 300 µl) to pass completely into the column. 4. Add 600 µl (1 and 3 reaction optimization kit) or 400 µl (validation kit) of GOLD-Hydrazide Wash/Coupling Buffer to the top of each column. This liquid is required to push the activated material to the base of the column. Allow this liquid to pass completely into the column before proceeding to step 5. 5. Place a 0.5 ml or 1.5 ml collection tube (not supplied) under the column. Add 200 µl (1 and 3 reaction optimization kit) or 450 µl (validation kit) of GOLD-Hydrazide Wash/Coupling Buffer to the top of the column. 6. Collect the eluate from the column into the collection tube. This tube now contains the activated material free of excess GOLD-Hydrazide Antibody Oxidation Reagent and other chemicals. 6

D. Concentration of Aldehyde Antibody We strongly recommend that protein values should be determined using an absorbance at 280nm. For an IgG using a 1cm light path an OD280 of 1.0 is equivalent to an antibody concentration of 0.714 mg/ml. When using protein assay reagents it is important to use an IgG standard curve. The absorbance generated by this type of reagent is dependent on the protein used. For example, using a BSA standard curve to determine the protein concentration of an IgG solution will result in a 2.3-fold under-estimate of the IgG concentration. 6. Labeling Protocol The kit may be used in different ways to suit the particular aims of your experiment. The antibody can be tested at different antibody-gold ratios and/or at different scales of reaction. The optimum amount of antibody (which will influence the number of antibody molecules per particle) may be application-dependent and must be determined by experimentation. Around 10-30 µg of aldehyde-activated IgG antibody per ml of 10 OD GOLD Hydrazide is likely to give optimal results. The dilution of the GOLD upon addition of the antibody has no effect on conjugation efficiency. 7

1. Remove the required amount of GOLD-Hydrazide from the stock bottle and allow to warm to room temperature. 2. If required dilute the aldehyde-activated biomolecule in the GOLD-Hydrazide Wash/Coupling Buffer. Mix gently. 3. Take the required amount of GOLD-Hydrazide to be labeled and add the aldehyde modified antibody (dilution of the GOLD nanoparticles with the antibody up to 1:20 are acceptable). 4. Leave the mixture standing for 30 minutes at room temperature (20-25 C); longer incubation time has no negative effect on the conjugate. 5. After the conjugation, if you wish to exchange the GOLD conjugate into a specific buffer for your assay or test, centrifuge the conjugate in a microfuge at 9,000g for 6 minutes. Carefully remove the supernatant and add your preferred buffer. The buffer should not contain thiols; all other common lab materials are acceptable after the conjugate has formed. 8

7. Frequently Asked Questions What can I do if my antibody preparation contains interfering substances? The simplest procedure is to dialyze the antibody against a suitable buffer. Relatively weak buffers (e.g. 20mM) are preferred so that the ph conditions of the covalent reaction are not significantly altered upon addition of the antibody. See also the comments in Section 5 of the protocol. If dialysis is not practical you can use other popular methods of buffer exchange if required e.g. desalting columns. Does the antibody bind to the metal surface? No. The protective surface coat completely shields the metal surface and prevents direct metal-antibody interactions. For this reason, you cannot use Gold Conjugation Kits for passive conjugation of antibodies. Can antibodies from different species be used? Yes, the system has been tested with antibodies from a variety of species including mouse, rabbit, goat and sheep. What type of linkage to gold is formed? The antibody becomes covalently and irreversibly attached via lysine residues to the GOLD surface. 9

Can antibody fragments be conjugated? Yes. One of the advantages of the protective coat is that it is less likely than a bare metal surface to cause denaturation and loss of affinity of the antibody fragments. What if I need bulk material? The kit that you have purchased is a convenience product for rapid production of small quantities of conjugate for screening purposes. Abcam offers the kit in a range of sizes. Please enquire. Is the kit suitable for conjugating analytes and other small molecules? Yes, but please contact technical services. Depending on the functional groups on your analyte you may need a different type of surface or different chemistry for optimal results. 10
