Overview UK Biobank UK Biocentre NIHR National Biosample Centre
UK Biobank: Overview Study Design Participant recruitment Sample Logistics Automation Design and Implementation Sample Processing Sample Storage UK Biobank Conceived in ,000 participants aged years 700 participants per day 6 tubes of blood + 1 tube of urine ~20,000 aliquots 15 million aliquots in total Funded by leading research organisations World leading experience and innovation in: Sample Analysis 3
UK Biobank baseline recruitment 503,000 participants recruited 9 million letters, 800,000 telephone calls 21 assessment centres 20 clinic years and ~ 800 staff 4.5 million biological samples collected ~4,000 blood tubes per day 20-25,000 aliquots generated daily 15 million sample aliquots 5,400 courier journeys (~432,000 miles) 1,050 operational sample processing days To successfully achieve a scientific endeavour at this intensity to high quality needs a different approach
UK Biocentre Visiting scientists to UK Biobank Desire to employ similar methodology Sole remit to deliver UK Biobank target recruitment Baseline recruitment completed October 2010 Downtime whilst preparing the cohort for access Infrastructure Funders UK Biocentre incorporated 2011 Grown quickly over the last 5 years
NIHR National Biosample Centre Desire to have a national processing and archiving centre for large scale studies NIHR released funding of 25 million The NIHR National Biosample Centre ( NIHR NBC ) is a result of a collaboration between the University of Oxford and UK Biocentre ( UKBC ). It is: Established to leverage the expertise built up by the UK Biobank project Required to reinvest all profits generated back into NIHR NBC Set up to support the BRC s, BRU s and academic research groups Committed to providing world-class biorepository services at the best possible price NIHR NBC opened in January 2015 and has been operating successfully for 18 months (UK Biocentre for 5 years).
The core resource High throughput, flexible, predominantly automated sample processing. Very large ultra-low temperature automated sample archiving capacity ~20 million samples. -20 C (2 million) -80 C (18 million) Manual archiving capacity -80 C -196 C Collaborator owned freezer and tank management Nucleic acid isolation DNA and RNA Automated quantification and normalisation systems Haematology analysis Business continuity through: Full back up of power systems in the event of mains power failure Duplicate samples stored off-site in liquid nitrogen storage facility. Full backup of IT infrastructure HTA license ISO 9001:2008 certified
Typical projects that could benefit from the new facility Consolidation and curation of legacy collections Sample processing and archiving for new studies Specific assay programmes e.g. clinical chemistry, DNA extraction and assay, genotyping Secure back-up of sample collections at other sites Projects requiring bespoke protocol development or technology development to support large-scale Projects requiring advice on upstream activities or sample collection protocols
INTERVAL Project Commenced 2012 Investigate blood donation interval period 50,000 participants at 2 time points Samples received daily Up to 200 sample sets per day 3 tubes per participant EDTA Serum EDTA for haematology Up to 1,000 aliquots per day 500,000 aliquots in total Retrieval and reformatting tasks COMPARE project
Genomics England Project Commenced ,000 genomes 70,000 patients Rare disease and cancer Samples received weekly from Genomic Medical Centres (GMCs) Quality control performed on each DNA sample ~500 samples per week A260/280 Pico Green measurement Agilent Tapestation (Agarose gel) Delta Cq Samples plated for sequencing Samples shipped to Illumina
