5 Cassettes and Accessories Bio-Wraps Bio-Wraps are sections of tissue paper cut to a convenient, ready-to-use 63 x 100 mm size. They are colored blue to help locate small biopsy specimens following paraffin infiltration. They will not bleed into specimens. Bio-Wraps are available in convenient dispenser packs x 75 mm Box / Biopsy Pads Biopsy Pads are made from open-celled foam, which allows fluids to transfer safely and thoroughly around tissue while holding the specimen securely. To prevent floating and avoid artifacts, Biopsy Pads should be presaturated in fixative. The thin design helps eliminate excess pressure from being applied to unfixed specimens, reducing the risk of tissue sticking to the pads or fibers coming apart during embedding. 7 Mounting Media a) Mounting Media Micromount Micromount is a rapid-drying, xylene-based mounting medium. Its low viscosity facilitates coverslipping and prevents air bubble formation. An antioxidant is added to inhibit stain fading and prevent the formation of annual rings. Micromount sets up quickly, allowing slide preparations to be handled immediately after mounting. Micromount is soluble in xylene, toluene, and most xylene substitutes. Refractive Index (Wet), (Dry) Micromount, 473 ml Each Micromount, 473 ml Case / MM 24 MM 24 is a low viscosity, ultra-fast drying, toluene based mounting medium that is compatible with both aliphatic (eg Sub-X) and aromatic (eg xylene) clearing agents. It is specially formulated to provide optimum results with automatic glass coverslippers, yet is also effective when manually preparing slides. Set up time before permanent filing can be as quickly as 24 hours when used with aromatic agents and 48 to 72 hours when used with aliphatics. This unique product eliminates the need to use hazardous xylene or xylene substitutes as a clearing agent in histology and cytology laboratories. When using Clearium, simply do away with the final step of clearing and coverslip directly from absolute isopropyl alcohol. 8 b) Frozen Section Media FSC 22 Frozen Section Compound FSC 22 Frozen Section Compound is a water soluble embedding compound used in frozen sectioning. The compound bonds and encapsulates tissue specimens to the object holder for cryosectioning. It provides excellent sectioning consistency with minimal curling of sections at a working temperature of -20 C. FSC 22 is available in clear or in light blue for better visualization of small specimens. 9 Paraffin Wax Paraplast Paraplast is recommended for general tissue embedding. It is composed of a refined mixture of highly purified paraffin containing plastic polymers of regulated molecular weights. This supports improved tissue infiltration and superior quality sections with minimal compression. Paraplast gives brittle, difficult to section tissue an innate elasticity, promoting wrinkle-free sections. 10 Safety & Hygiene D-Formalizer Pads D-Formalizer Pads reduce formaldehyde exposure by absorbing and neutralising any excess formalin. The durable blue surface of the pads provides an excellent contrast to even small pieces of tissue. The Pads are also useful for soaking up leakage and deodorizing vapors in other applications. They can be used to line shelves which hold specimen containers that may seep formaldehyde around the lid, or to line transport boxes where leaks may occur D-Formalizer Pads, 280 x 356 mm Pack / D-Formalizer Powder D-Formalizer is a safe chemical treatment for neutralizing spent formalin. It leaves a clear solution, free of solids and aldehyde odor, that can be disposed of without clogging the drains. Simply add one packet of D-Formalizer to 3.8L of used formalin (or 4% glutaraldehyde) and agitate gently. After 15 minutes, the product can be safely poured down the drain in compliance with local ordinances. 12 Slide Folders & Boxes 100 Slide Box The 100 Slide Box is made of high impact polystyrene and secured with rust resistant, nickel-plated clips and hinge pins. The bottom of the slide box is lined with cork. To ensure proper slide identification, each slot is numbered to correspond to the index on the inside cover. Boxes will hold 3 x 1 or 25 x 75 mm microslides White Each Slide Folder The 20 Slide Folder is a robust chipboard folder, handmade using a double wall construction. The front slide protection flaps are made to fold back without risk of damage to the hinge. Each slide is identified with a number and by an index printed on the front. The slides are easily removed simply by touching either end of the slide. 13 Stains & Reagent Solutions a) Hematoxylin Stains Gill Hematoxylin Gill II is a medium strength hematoxylin. The Gill formula hematoxylins are used in the progressive method of staining. The Gill formulations do not contain mercury Gill II, 3.8 L Each 96 Harris Hematoxylin Harris Hematoxylin produces a deep, purple-blue stain. It is generally used in regressive methods, but may also be used progressively both in cytology and histology preparations. 14 b) Counter Stains Eosin Eosin Y is an alcohol-based, ready-to-use secondary stain used primarily in histology for routine hematoxylin and eosin staining. It may also be used in cytology for fast cytoplasmic staining of rush specimens also using the H&E technique Eosin, 3.8 L Each 60 EA-50 EA-50 is a light green, alcohol-based stain primarily used in the cytology laboratory for routine cytoplasmic staining. 15 c) SeleTech Staining System SelecTech SelecTech is a hematoxylin and eosin staining system consisting of Hematoxylin 560, Blue Buffer 8, Define, Alcoholic Eosin Y 515 or Eosin Trichrome 515. SelecTech standardizes the reagents and stains used in the hematoxylin and eosin staining procedure. The reagents and stains that make up this system must pass vigorous analytical chemistry standards and performance tests. The built-in quality assurance parameters assure consistent results. 16 d) Other Confix Blue (Australian Biostain) Confix Neutral Buffered Formalin is Easy and Quick, saving time and providing consistent quality The ph and Osmolality of the Australian Biostains Confix solutions are strictly controlled to ensure the best in tissue preservation and morphology as well as improved antigen demonstration and retrieval. The blue colour helps immediately identify that the sample is in formalin. The 5 L container is easy to lift and pour. Confix is a concentrate. Mix 1 part to 5 parts of RO water makes 10% NBF. 5 L makes 25 L ACFB.5L Confix Blue, NBF Concentrate, 5 L Each 63 Schiff s Reagent Schiff s Reagent is produced in small batches, ensuring maximum consistency and shelf life. It is used in the standard Periodic Acid-Schiff (PAS) reaction for neutralmucosubstances, glycogen, basement membrane and fungal cell walls. A positive reaction produces a purplish red/bright rose color Schiff, 473 ml Each 50 Decalcifier Decalcifier I is a unique decalcifying solution that has the ability to fix and decalcify at the same time. Needle biopsies may be completely fixed and decalcified in one hour or less. Small bone specimens and calcified arteries may be completely fixed and decalcified in approximately 2-4 hours. It is recommended to use 20 times the volume of the specimen. Following decalcification, the specimen is ready for processing without having to wash first. Tissues decalcified in this solution cut with ease and routine H&E techniques produce excellent stained sections. 17 Scott's Tap Water Substitute Concentrate Scott s Tap Water substitute is a blueing reagent designed for histology and cytology. It blues hematoxylin gently and minimizes loss of tissue sections and cells from glass slides. Scott s Tap Water Substitute is prepared to supply a gentle but appropriate blueing solution for staining. This formulation provides rapid and precise blueing of nuclear chromatin and nuclear membranes.
