The platform has been optimized to capture as few as 10s of tumor cells from millions of blood cells. With this approach the platform can capture target cells from most cell suspensions with high sensitivity and reproducibility. This device uses immunoaffinity-based capture. It contains 58,000 nano-post structures that are coated with an antibody against surface antigens expressed on the target cells. As the sample flows through the device target cells are captured on the posts while unwanted cells flow out of the device. Captured cells can then be immunofluorescently stained, imaged and analyzed to quantify target cells on-chip.

Large sample can be processed to improve the rare cell yield and increase processing speeds. This device uses immunoaffinity-based capture. It contains bean-shaped microposts that are coated with an antibody against surface antigens expressed on the target cells. The device can be tailored to any target antigen of interest, including multiple antibody cocktails. As the sample flows through the device target cells are captured on the posts while unwanted cells flow out of the device. Captured cells can then be immunofluorescently stained, imaged and analyzed to quantify target cells on-chip.

The rapid processing time enables large sample to be processed in a timely manner. With the high purity of target cells, many down stream assays can be performed. This device uses size-based cell sorting which is advantageous for samples in which the cells of interest have a distinct size range compared to other cells. As the sample flows through the device target cells are aligned at a unique streak along the device channel and separated from unwanted cells. Cells are collected in suspension after which they can be immunofluorescently stained, imaged and analyzed as well as undergo cell assays of interest.

As a result of combining both size-based and immunoaffinity based rare cell isolation, the system allows for target cells to be isolated with remarkable purity. The system is ideal for samples from which removal of unwanted cells is desired, such as with sample preparation for sensitive molecular and genomic assays. This device relies on size based sorting followed by immunomagnetic isolation of the pre-sorted target cells. The approach also facilities negative depletion of unwanted cells.
