Overview Cell-based production of active pharmaceutical ingredients generally requires supplementation of cell culture media to promote cell growth and boost protein production. Every cell line can offer unique challenges for developing processes that yield desired performance criteria. One bioprocess challenge is the selection and optimization of a cell culture supplement or feed that will result in desired titer, growth characteristics, and protein quality. Therefore, the availability of a family of chemically defined (CD) supplements offering a diverse performance profile can significantly improve process development timelines. D iosciences dvanced ioprocessing has designed and formulated a diverse set of fully chemically defined (CD), animal free (F), and protein free cell culture supplements, D Resurge TM CD-CD supplements, without glucose and L-glutamine. This set of supplements can be used in batch and fed-batch processes to enhance recombinant protein production in mammalian cells while maintaining product quality. These supplements have been developed with multiple CHO host cell lines and in various culture systems including shaker flasks and bench-scale bioreactors. single supplement may not be ideal for enhancing performance across multiple cell line conditions; therefore a wider array of CD supplement formulations may be more effective for process development. pplication in atch and Fed atch Modes-Shaker Flasks D Resurge CD-CD supplements can be used in batch and fed batch modes in cell culture processes. D Resurge CD Supplements in ioreactor Performance of D Resurge CD supplements is scalable to bioreactors. The five D Resurge CD supplements produced a --fold increase in protein production over media control in bench top bioreactors in batch or fed batch mode. Equivalent results were obtained in shaker flask studies, indicating successful scalability.

Protein Quality with D Resurge CD Supplements How a CD supplement impacts protein quality of a therapeutic antibody is dependent on multiple factors: the type of expression host, the type of base media, and the cell culture process. The end user would have the best understanding of the critical quality attributes of their product, and ultimately preclinical and clinical studies would determine the therapeutic impact of any glycan changes. The N-glycan profiles of monoclonal antibodies from D Resurge supplemented and control conditions were analyzed. typical elution profile is shown for the D Resurge CD supplemented condition in CHO line (Figure ). The structures and relative composition of the most prevalent glycans from each cell line are depicted in Figures (CHO line ) and (CHO line ). Differences between the supplemented and control conditions were similar to those seen when using other supplements, such as peptones (data not shown).
