ChemiDoc XRS+ system (0 sec exposure) Comparison of chemiluminescent western blot detection on X-ray film and with the ChemiDoc XRS+ system. Blots of twofold serial dilutions of human serum were probed with rabbit anti-human transferrin polyclonal antibodies. A /,000 dilution of human serum was used to make the twofold serial dilutions. A 00 sec exposure on film does not reach the same limit of detection that is reached by a 0 sec exposure in the ChemiDoc XRS+ system. Chemiluminescence Visible Marker Merged Image Quantitative data. Film reached maximum detection limit at 00 sec, but the first two dilutions are saturated. Using the Quantity One -D analysis software -D viewer shows that the tops of the bands are flattened and are no longer quantitative, as there is no way to visualize their peaks and differentiate the exact volume of each. The 0 sec exposure on the ChemiDoc XRS+ system shows that the first two dilutions are not saturated; the peaks are visible on the -D viewer, enabling differentiation of the protein concentrations. Analysis Chemiluminescent sample detection and visible markers. Visible markers are commonly used to monitor any problems with SDS-PAGE and blot transfer. To determine the molecular weights, users trace the pattern of the visible marker from the gel on the blot or film. This adds additional steps and can possibly introduce errors. The ChemiDoc XRS+ system images the chemiluminescent samples and visible markers to provide a digital record; the images are then merged for molecular weight determination. 0 sec 0 sec 0 sec Live acquisition. The ChemiDoc XRS+ system tracks progressive image development, allowing users to visualize the chemiluminescent reaction in real time and capture the image they want. The total acquisition time and the number of images taken during this time can be customized, and each image can be saved. This removes the tricky exposure bracketing often needed even with a single film exposure. Multifluorescent western blot detection of maltose binding protein (MBP) and Profinity exact fusion-tagged MBP bands using Qdot and Qdot fluorescent conjugated secondary antibodies (Invitrogen Corp.), respectively, imaged with the ChemiDoc XRS+ system. The blot was imaged with excitation via UV, and separate images were taken by switching between the SYBR Green (Qdot ) and SYPRO Ruby stain (Qdot ) filters installed in the ChemiDoc XRS+ imager. Multifluorescent western blotting enables detection of two antibody-specific proteins at once, removing the variability involved with two blots or the loss of protein with stripping. Live image acquisition records real-time development of the chemiluminescent reaction, removing the guesswork often involved with single-event film capture of blot signals. Quality optics engineered for optimal signal-to-noise ratio ensure antibody-specific detection of low-abundance proteins. Automatic digital capture of chemiluminescent blot signals eliminates the chemical use and disposal problems involved with film developing. A self-contained lighttight enclosure means you don t need a separate darkroom and won t have to bother with scheduling an experiment around darkroom availability. Sophisticated analysis features support a variety of samples, including cell lysates and purified proteins. Acquisition settings can be stored for repeated use during your experiment or project, such as for efficacy studies over a period of time. Quality digital images, numerical data, and sample annotations are easily exported for publication in grant proposals, papers, and presentations or for sharing with colleagues, and they can be printed for documentation in laboratory notebooks. Total protein staining with Coomassie Blue stain and GFP fluorescence can be overlaid using the multichannel viewer, and purity analysis is performed using the band analysis software feature.
