2 Scharlau microbiology History Scharlau Chemie S.A. has been manufacturing laboratory chemicals since In 1979 ADSA-MICRO started the manufacture of dehydrated media. This company was acquired by Scharlau Chemie S.A. in 1992 and the brand changed to Scharlau. A new facility for the manufacture of dehydrated media was set up in 2002, which allowed us to increase capacity and to further improve the quality of our media. 3 Prepared solid media used for water analysis Filtration plates Prepared 55 mm filtration plates in blister packs Each blister pack comes in a sealed flowpack bag. This kind of packaging increases the shelf life to over 6 months, avoiding fast dehydration and contamination. The latest directives, relative to the analysis of water for human consumption, indicate the use of media with agar. Such media allow more precise enumeration, since without an excess of water, the colonies can grow perfectly separated and with the typical morphology. Formulations follow specifications of the various Pharmacopoeiae and ISO regulations. Packaging into blisters inside the clean room to avoid contaminations. 4 Water analysis according to EN-ISO directives Membrane filtration technique The water sample is passed through a 0,45 μm membrane filter. Microorganisms are retained on the surface of the membrane. Membrane with microorganisms is placed on a Petri dish of 55 mm that contains the appropriate agar. After incubation at right temperature and time, colonies that have grown on the membrane surface can be directly counted. This MF technique offers a series of advantages over the traditional MPN technique (Most Probable Number): Much more representative sampling. The membrane allows reco very of very low concentration of microorganims in large volumes of liquid. Therefore this technique is suitable for very low contaminated water. Definitive results in less time. More precise and direct enumeration of the colonies. Indicated for strongly mineralized waters that give wrong reactions in the colimetry in liquid media. Membranes can be kept for a long period of time for testimonial, medical or legal purposes. Considerable savings in space, labour and laboratory material as compared to the MPN technique. This technique has been adopted also for the control of water not destined to human consumption, e.g. process water in manufacturing. 6 Water analysis according to EN-ISO directives Coliforms and Escherichia coli Chapman TTC Agar (Tergitol 7 Agar) (Lactose TTC Sodium Heptadecylsulfate Agar) Intended use Selective medium for the detection and enumeration of coliforms, heat-tolerant coliforms and Escherichia coli following the membrane filtration method. This medium is mentioned in the EN ISO :2000 directives. Comments Tergitol 7 inhibits the growth of Gram positive bacteria. TTC solution added to the medium serves as bacterial growth indicator and for an earlier recognition of Escherichia coli and Enterobacter aerogenes. Instructions Filtrate 100 ml of sample water through membrane of pore size 0,45μm. Use sterile forceps to carefully place the membrane, filtered side up, on top of the agar of the filtration plate. E. coli Incubation Incubate for (21 ± 3) hours at (36 ± 2) C and avoid excessive dehydration of the plate. Interpretation Colonies capable of fermenting the lactose and the consequent production of acid that leads to a colour change of the medium from green to yellow are to be considered suspicious. Supplements TTC Other bacteria Confirmation All suspicious colonies have to be confirmed with Indole and Oxidase reagents. Count as coliforms all characteristic colonies that are oxidase-negative. Colonies with oxidase-negative and indole-positive reations should be considered E. coli. 7 Water analysis according to EN-ISO directives Enterococci Slanetz & Bartley Agar Intended use Selective medium for enumeration and detection of Enterococci in drinking water, waste water and beverages following the membrane filtration method. According to EN ISO :2000 directives. Comments The medium contains sodium azide to inhibit the growth of Gram negative bacteria. TTC solution added to the medium allows better counting. Instructions Filtrate 100 ml of sample water through membrane of pore size 0,45 micron. Use forceps to carefully place the membrane, filtered side up, on top of the agar of the filtration plate. Slanetz-Bartley no growth Enterococcus sp. Incubation Incubate for (44 ± 4) hours at (36 ± 2) C and avoid excessive dehydration of the plate. Interpretation Colonies capable of growing in presence of sodium azide and of reducing TTC to form red, brown or pink colour. Confirmation Carry out confirmation of suspicious colonies using Bile Esculin Azide Agar (01-592) observing those colonies that are capable of hydrolizing the esculin within one to two hours forming colours that may range from brown to black. 8 Water analysis according to EN-ISO directives Enumeration of culturable microorganisms Tryptone Yeast Extract Agar Intended use Solid medium for the enumeration of culturable microorganisms by colony count in water. According to EN ISO 6222:1999. Over 300 colonies Comments The nutrients of the Tryptone and the vitamins of the yeast extract favour the growth of most bacteria. Instructions Mass inoculation technique. 2 sets each of sterile Petri dishes. In series 1 we will place 1 ml of water. In series 2 we will place 0,1 ml of water. 10 to 15 ml of medium, once melted and kept at (45 ± 1) C, must be poured over the samples avoiding bubbles colonies Incubation One set of inoculated plates at (36 ± 2) C for (44 ± 4) hours and the other set at (22 ± 2) C for (68 ± 4) hours. Interpretation All colonies are counted, expressing the result in amount of colonies per ml. Only plates containing colonies are used for enumeration. If there are over 300 colonies per ml, test will have to be repeated with higher dilutions. 9 Water analysis according to EN-ISO directives Clostridium perfringens (inclusive of spores) m-cp Agar Intended use Solid medium for enumeration and isolation of Clostridium perfringens in water for human consumption by membrane filtration method.according to European Directive 98/83/CE. Comments Clostridia hydrolyse the saccharose forming yellow colonies. After exposure to ammonia vapours, the colonies change to red colour. Instructions Filtrate 100 ml of sample water through membrane of pore size 0,45 micron. Use forceps to carefully place the membrane, filtered side up, on top of the agar of the filtration plate. Incubation Incubate under anaerobic conditions for (21 ± 3) hours at (44 ± 1) C. Clostridium perfringens before development Interpretation Colonies of opaque yellow colour are to be considered suspicious. Confirmation Plate has to be submitted to the effects of vapours of ammonia for seconds. Those colonies that change from yellow to pink or red will be counted. Suitable for membrane filtration method. Principle Based on the detection of two enzyme activities: ß-D-Glucuronidase (ß gluc) ß-D-Galactosidase (ß gal) Improved formula with better inhibition of the Gram positive accompanying flora, using bile salts instead of Tergitol. After incubation at 37 C for 24 hours, E. coli colonies appear in blue colour and the balance coliforms in salmon colour. The remaining enterobacteria do no present any coloration. These are the media used in water testing. Coliforms and E. coli E. coli bacteria is a fecal contamination indicator. Traditional methods for detecting E. coli are extremely tedious and usually require a heavy workload since many suspect colonies have to be tested. CHROMagar E. coli. Is a culture medium which directly marks E. coli colonies in blue colour EC EC169 5 litres* 25 litres* CHROMagar ECC. Will additionally show the other coliforms as red colonies. 13 Chromogenic Media Chromogenic Media CHROMagar CHROMagar Liquid ECC. Is used as a broth for the pad technique for the detection of E. coli and coliforms in water samples. The inoculated filtration membrane is put on top of a pad presoaked with CHROMagar Liquid ECC EL382 5 litres* CHROMagar Vibrio. V. parahaemolyticus, V. vulnificus & V. cholerae are pathogenic bacteria which can cause serious seafood poisoning. For the detection of those bacteria, traditional methods (TCBS) are time consuming, require heavy workload and are not very sensitive. On the contrary, CHROMagar Vibrio medium helps to easily differentiate V. parahaemolyticus, V. vulnificus & V. cholerae, from other Vibrio directly during the isolation step by colony colour with a sensitivity higher than conventional methods VB912 5 litres* CHROMagar Pseudomonas. For the simultaneous detection and enumeration of Pseudomonas aeruginosa with markedly different colouring (blue colonies) PS822 5 litres* Presence/Absence test Coliforms Presence/Absence Broth Chromogenic Medium Intended use Direct and easy detection of presence or absence of total coliforms and of Escherichia coli in water samples. Comments Chromogenic and fluorogenic broth that combines the test of the enzymes ß-D-Glucuronidase and ß-D-Galactosidase and the indole test* in a single flask, ready for use. 14 Diagnostic tests Oxidase Test For detection of cytochrome oxidase in oxidase producing microorganisms like Pseudomonas sp. Sterile swabs drenched with Oxidase reagent, ready for use. This detection has been improved with a new mixture of chromogenic substrates. Presence of coliforms gives a blue-greenish colour, and presence of E. coli also gives fluorescence under UV light; and it can also been confirmed with the adition of Kovacs reagent (indole test). Kovacs Indole Reagent Intended use Classical test to differentiate Escherichia and Enterobacteria. Also used for the differentiation of other non enteric microorganisms. Comments Many microorganisms can produce Indole from Tryptophan thanks to a Tryptophanase, in a process favoured by oxygen and inhibited by glucose. Media used in this test should contain no glucose, should present a high Tryptophan content and should be incubated aerobically. Instructions The pure culture to be verified has to be inoculated in the medium. Weak positive Negative Positive Incubation Incubate sample for hours at 35 C. This incubation can be reduced to 4 hours, if you part from a massive inoculum in solid medium and if you inoculate in a small volume of culture medium (Indole Test Cat. no. 064-TA0132).
