1 CLEARING AGENT l Embalming Historically, toluene and xylene have been used as clearing agents throughout the histology lab in the processes of tissue and slide preparation to remove alcohol and other dehydrants from tissues prior to embedding (usually in paraffin wax), and from finished slides prior to mounting. They are also used after sectioning to remove paraffin wax after cutting on the microtome. However, the effectiveness of xylene and toluene as clearing agents are associated with the ring structures in their molecules, while their double-bonds are related to their hazardous side effects. HistoChoice TM Clearing Agent contains none of the harmful chemicals associated with xylene and toluene. Non-toxic. Non-carcinogenic. Non-flammable with a higher flashpoint than xylene/toluene. Virtually Odorless. Easy to use in confined areas. Non-irritating to human skin**. **Can cause drying of skin after prolonged contact with the liquid. HistoChoice TM Clearing Agent directly replaces xylene and toluene. It is a much more effective alternative to xylene and toluene substitutes including: chlorinated hydrocarbons, terpenes, and other related solvents. Additionally, HistoChoice TM is about twice as effective as parafinnic hydrocarbons. Though less hazardous than xylene and toluene, these substitutes can be effective solvents, but can also leave an oily residue, dry very slowly, and lack the strong solvent characteristics necessary to produce good tissue preparations. They are also considered toxic chemicals posing serious health risks and often have strong odors, making work in confined, poorly ventilated areas difficult. Most are also highly flammable. Compatible with many organic mounting media. Faster wax dissolution, slower evaporation than xylene. No oily residue. Can be used in automated processors. Coverslips easily removed by immersion in HistoChoice. NOTE: Dispose by using standard procedures for xylene disposal. Follow normal laboratory procedures for handling non-polar solvents. HistoChoice TM Clearing Agent is an economical substitute for xylene and toluene. It also does not have the expensive hazardous waste disposal costs that are normally associated with some xylene and toluene substitutes containing harmful elements. Designed to work well with isopropanol. Non-toxic means it s less expensive to ship. Less-flammable for lower storage costs. Minimize compliance costs. Can be easily recycled. 6 The Next Generation of Environmentally Safe Molecular Biology Products. Molecular Biology FIX l Molecular Biology l DNA Procedures l mrna Procedures l In Situ Hybridization l Histopathology l Molecular Pathology AMRESCO s HistoChoice TM Molecular Biology (MB) Fixative is a safe, convenient solution for tissue fixation developed specifically for the molecular biologist. HistoChoice TM MB Fixative improves healthcare delivery because it is safe to use and is gentle on tissues and the environment. HistoChoice TM Tissue Fixative contains none of the harmful chemicals associated with Formalin-based products. Odorless and non-toxic. Gentle on your tissue samples. Can be safely washed down the drain upon neutralization (in most districts). HistoChoice MB Code Size H ml 100 ml H120-1L 1 L H120-4L 4 L HistoChoice TM MB is specifically formulated to preserve antigenic sites for antibody probes and nucleic acid sites for in situ hybridizations. HistoChoice TM MB directly replaces common fixatives including: formaldehyde-based, alcoholbased, Zenker s, B5 & B3, Bouin s and other fixatives with superior results. Tissues fixed in HistoChoice TM MB exhibit vibrant staining, better nuclear and cytoplasmic detail, and will retain a crisp appearance even after long-term fixation. HistoChoice TM MB fixed tissue also retains a much more natural look and feel than formalin fixed tissue. *Softer tissues mean easier, smoother cutting and sectioning. Superior sensitivity without predigestion. Markers can be observed at lower antibody levels. Vibrant staining with sharp visual intracellular detail. Full nuclear and cellular detail preserved. Very intense reactions, even after long-term fixation. No crosslinking problem of tissues as experienced with Formalin-based fixatives. Because this formula is designed specifically for molecular biology applications, HistoChoice TM MB fixed sections do not require predigestion or other recovery procedures to make important sites available. This means you spend less time preparing slides and more time doing research. More than a fixative, HistoChoice TM MB is a preservative that leaves antigens and nucleic acids in their native state, allowing binding for specific probes. Primary antibodies can be diluted several-fold due to the increased number of preserved antigenic sites, saving cost on every slide you process. Save costs on every slide processed. No more exposure monitoring costs. Less slide preparation time. Non-toxic, means it s less expensive to ship. Minimizes disposal and compliance costs. Available in multiple sizes. Ideal transport medium. *NOTE: Because soft tissues cut thinner than formalin-hardened samples, we recommend increasing the microtome settings by 1-3 microns.
