Njen pekoči okus povzroča snov kapsaicin iz skupine alkaloidov, kateri ima veliko že raziskanih in še neraziskanih delovanj na človeka in je edinstven za rod Capsicum. Nedavno posekvenciran genom paprike omogoča nadaljnje raziskave v rodu paprik in njenih sorodnic. Uporaba genetskih orodij veliko prispeva h kvalitetnemu žlahtnjenju novih sort paprik. Z uporabo DNK markerjev lahko določimo oz. sledimo kapsinoidni biosintezni sposobnost rastline. Preverjali so uporabnost razvitih SCAR markerjev v žlahtniteljskem programu paprike in nukleotidno zaporedja regije, kjer je prisotna delecija v ne-pekoči papriki, uporabili za filogenetske študije. Razviti genetski markerji imajo neposredno uporabno vrednost za razločevanje pekočih in ne-pekočih paprik v stadiju sadik v žlahtniteljskih programih. III

Its spicy taste is caused by a substance called capsaicin, from the group of chemicals named alkaloids, which has a lot of documented and undocumented effects on humans and is unique to Capsicum genus. Recently sequenced pepper genome enables further research in the genus of peppers and its relatives. The use of genetic tools makes a significant contribution to the quality of breeding new varieties of peppers. By using the DNA markers we can determine capsinoid biosynthetic ability of plants. Researchers have discovered the usefulness of SCAR markers for pepper breeding programs and used nucleotide region of sequence hrbouring a deletion present in mild peppers for phylogenetic studies. Developed genetic markers have direct practical value in breeding programs for distinguishing spicy and non-spicy peppers at the seedling stage. IV

8 1 UVOD Paprike izvirajo iz Srednje in Južne Amerike, in so sedaj pomembna zelenjadnica, ki jo gojijo po vsem svetu. Uživa se jih kot svežo zelenjavo in predela v prah, omake in salse. Pekoč okus je edinstvena lastnost rastlin iz rodu Capsicum in ena izmed glavnih lastnosti za gojitelje paprik. Eden od ciljev žlahtniteljskih programov paprike je nadzor pekočega okusa ploda, da bi izpolnili različne potrebe predelovalne industrije in samih potrošnikov (Yoon in sod., 1992). Skoraj vse divje pekoče paprike so pekoče, medtem ko so sladke paprike rezultat udomačitve (Andrews, 1995). Večina paprik sodi v vrsto C. annuum, ki je najbolj gojena vrsta. Za vzgojo paprike, je natančna izbira različno pekočih linij pomembna in tudi zaželena. 2 PAPRIKA Paprika je član družine Solanaceae oz. razhudnikovk. To je diploidna, fakultativna, samoprašna rastlinska vrsta in je tesno povezana s krompirjem, paradižnikom, jajčevci, tobakom in petunijami, ki spadajo v isto družino. Številne rastline družine razhudnikovk imajo enako število kromosomov (n = 12) vendar se bistveno razlikujejo v velikosti genoma. Paprika je ena izmed najstarejših udomačenih rastlin na zahodni polobli (Aguilar- Meléndez in sod., 2009), je najbolj razširjena začimba na svetu in je glavna sestavina v večini svetovnih kuhinj (Bosland in sod., 2012). Paprika ima širok spekter uporabe, vključno v farmacevtiki, industriji naravnih barvil in kozmetiki, kot okrasna rastlina in kot aktivna sestavina v večini obrambnih repelentov. Paprika ponuja številne bistvene vitamine, minerale in hranilne snovi, ki imajo velik pomen za zdravje človeka (Marín in sod., 2004). V letu 2011, je 20 največjih držav, ki proizvajajo papriko pridelalo 33,3 milijonov ton pekoče paprike, zasajene na 3,8 M ha zemljišč (statistika Združenih narodov za prehrano in kmetijstvo (FAO), glej URL). V zadnjem desetletju se je svetovna proizvodnja paprike povečala kar za 40 %. 2.1 KAPSAICINOIDI Do pekočega okusa paprike pride zaradi kopičenja kapsaicinoidov, skupine alkaloidov, ki so edinstveni za rod Capsicum. Kapsaicin, dihidrokapsaicin in nordihidrokapsaicin predstavljajo primarne kapsaicinoide, ki se proizvajajo izključno v žlezah na placenti ploda. Organoleptični občutek toplote nastane, ko se kapsaicin veže na prehodne receptorje sesalcev. Številni encimi, ki sodelujejo v biosintezi kapsaicina niso dobro opisani, in regulacija poti ni povsem jasna. Z več kot 22 kapsaicinoidov izoliranih iz pekoče paprike, ta rod zagotavlja odličen primer za raziskovanje razvoja sekundarnih metabolitov v rastlinah (Bosland in sod., 2012). Kapsaicinoidi so v naravi pokazali, da imajo protiglivične in antibakterijske lastnosti, da delujejo kot svarilo za živali plenilcev ob zaužitju in imajo značilne lastnosti, ki pomagajo ptičem pri širjenju semen. Kapsaicinoidi imajo številne koristi za zdravje ljudi: so učinkoviti pri zaviranju rasti različnih oblik raka, (Surh, 2002; Ito in sod., 2004) so analgetik za artritis in druge bolečine (Fraenkel in sod., 2004), zmanjšujejo apetit in spodbujajo zmanjšanje teže (Lejeune in sod., 2003; Smeets in sod., 2005). Presenetljivo je, da je popolno razumevanje biosintetske poti kapsaicina na 1

9 molekularnem nivoju pomanjkljivo, glede na gospodarski in kulturni pomen kapsaicinoidov. 2.2 GENOM PAPRIKE Nedavno so določili genomsko zaporedje visoke kvalitete za pekočo papriko ʹC. annuum cv. CM334ʹ (ʹCriollo de Morelos 334ʹ), ki je domača sorta iz mehiške države Morelos. Ta sorta je stalno prikazovala visoko stopnjo odpornosti na različne patogene, vključno z glivo Phytophthora capsici, virusom pegavosti paprike in koreninske nematode. Ta domača sorta je bila v veliki meri uporabljena v raziskavah pekoče paprike in pri žlahtnjenju novih kultivarjev. Pridobili so tudi nukleotidne podatke dveh gojenih paprik in za divjo vrsto C. chinense. Primerjalna genomika članov družine Solanaceae, ki vključuje pekočo papriko, predstavi evolucijski pogled v širitev genoma, poreklo pekočega okusa, ločenega procesa zorenja in odpornosti proti boleznim pekoče paprike. Ta visoko kakovosten referenčni genom paprike bo služil kot platforma za izboljšanje hortikulturnih, hranilnih in zdravilnih vrednosti Capsicum vrst (Seungill in sod., 2014). Ta študija predvideva, da je lastnost pekočega okusa paprike nastala z razvojem novih genov, zaradi neenakega podvajanja obstoječih genov in zaradi sprememb v izražanju genov v plodu po speciaciji oz. nastanku novih vrst. CM334ʹ. Uporabili so Illumina tehnologijo določevanja nukleotidnih zaporedij genomskih knjižnic z inserti velikosti od 180 bp do 20 kb. Na podlagi analize 19 bp dolgih DNA zaporedij (K-mer analiza) so ocenili velikost genoma paprike na 3,48 Gb. 2

10 Po filtraciji podatkov in sestavljanju so dobili 3,06 Gb velik genom v 37,989 skafoldih in 90 % dela genoma je bilo predstavljenih s samo imi skafoldi (Preglednica 1). Ponovno so sekvencirali dva kultivarja paprike ('Perrenial' in 'Dempsey') in de-novo sekvencirali tudi divjo vrsto (C. chinense ʹPI159236ʹ, v nadaljnjem besedilu: C. chinense), da bi zagotovili celovit pregled genetske variabilnosti in razlike v strukturi genoma med različnimi sortami paprik. Divergentne sekvence so zelo razpršene vzdolž kromosomov paprike. Število malo pokritih regij (190 v 500 kb premikajočih oknih zaporedja), ki niso bili usklajeni med C. annuum in C. chinense, kažejo genomske razlike v teh dveh vrstah (Slika 1). Genom pekoče paprike deli veliko ohranjenih regij z genomom paradižnika, njegovemu sorodniku znotraj družine Solanaceae. Vendar pa je genom paprike približno štirikrat večji od genoma paradižnika, zaradi večjega kopičenja ponavljajočih se sekvenc v obeh, tako heterokromatinskih kot evkromatinskih regijah genoma (Slika 2). Najpogostejša ponavljanja genomu pekoče paprike so bili LTR retrotranspozoni, kot v mnogih drugih rastlinskih genomih (Feschotte in sod., 2002). (Seungill in sod., 2014). 3 MARKERJI PRI ŽLAHTNENJU PAPRIKE Pekoč okus je odvisen od enega samega prevladujočega gena (Andrews, 1995), kot smo že omenili v predhodnem tekstu. Molekularne genetske karte paprike so razvili: Lefebvre in sod., 1995; Livingstone in sod., 1999; Kang in sod., 2001 ter Blum in sod., Lokus C, gen ki določa pekoč okus, se nahaja na kromosomu 2 (Blum in sod., 2002; Lefebvre in sod., 2002). Prav tako so bili nekateri geni za biosintezo kapsaicina že izolirani. Curry in sod., 1999 so izolirali gene, ki kodirajo domnevno aminotransferazo (pamt) in sintazo 3- keto-acil-acp (KAS). Kim in sod., 2001, so izolirali 39 cdna iz placent plodov čilijev z metodo subtraktivne cdna knjižnice (SSH, angl. suppression subtractive hybridization). Opisali so tri gene, ki kodirajo encime biosinteze kapsaicina: SB2-66, določena domnevno kapsaicin - sintazo (CS), SB2-149, aminotransferazo in SB1-58, A-sintaza 3-keto-acil- ACP. Analizirali so segregacijo SB2-66 in ugotovili, da je tesno povezana z lokusom C. Razvili so tudi pet SCAR markerjev, ki temeljijo na CS zaporedju in testirali njihovo uporabnost pri žlahtnjenju oz. kot marker za pekoči okus. 3.1 SCAR MARKERJI, KI TEMELJIJO NA CS ZAPOREDJU Določitev segregacije lastnosti pekočega okusa F2 populacije iz križanja C. annuum ʹCM334ʹ in C. annuum ʹECW123Rʹ so izvedli v opisanem poskusu. Rastline so gojili v rastlinjaku od leta 2002 do leta Plodove paprik 121-ih rastlin so pobrali v primerni 4

12 zrelosti in opravili test degustacije (2002) in analizo s HPLC metodo (v letih 2002 in 2003) za kvalitativno in kvantitativno določanje vsebnosti kapsaicina. Preučili so medsebojno dedovanje lokusa CS (na osnovi nukleotidnih zaporedij) in lokusa C za potrditev povezave lokalizacije CS z lokusom C, z F2 populacijo iz križanja z nepekočim C. annuum, ʹECW123Rʹ in rahlo pekočim C. annuum, ʹCM334ʹ. HPLC analiza kapsaicinoidov od 121-ih F2 posameznikov je pokazala, da je razmerje med pekočimi, in ne-pekočimi posamezniki 3: 1 (χ2 = 0,03). Tako se je lastnost pekočega okusa obnašala kot tipična dominantna monogenska lastnost. Pregledali so genomsko DNA populacije F2 in staršev s PCR z začetnimi oligonukleotidi CSF1 in CSR4. PCR produkt je bil en sam fragment dolg približno 1,6 kb, kot je bilo pričakovano iz CS zaporedja. Fragmenti so se jasno ločili v vseh testiranih posameznikih (Slika 3). Gen CS je bil izražen različno v tkivu placente (Kim in sod., 2001) pri pekoči in ne-pekoči papriki (Slika 4). Tako genetsko kartiranje in specifična ekspresija v placenti pekoče paprike potrjuje ugotovitev, da gen CS ustreza lokusu C. Slika 3: Prikaz povezanosti zaporedja CS s pekočim fenotipom. Uporabljen par začetnih oligonukleotidov je CSF1 in CSR4. Elektroforezo produktov so izvedli na 0,8 % agaroznem gelu. Številke linij so številke F 2 paprik. C je pekoča paprika ʹCM334ʹ in E nepekoča paprika ʹECW123Rʹ, ki sta bili uporabljeni v križanju kot starševski liniji. Pekoč fenotip je označen kot "P" in ne pekoč kot "N". Na sliki se lepo vidi povezanost fragmenta DNA s pekočim fenotipom (Choong in sod., 2004). 5

Linija 1, Capsicum annuum, ʹHaehwaIIIʹ; linija 2, C. annuum, ECW; linija 3, C. chinense, ʹHabaneroʹ; linija 4, C. annuum, ʹCM334ʹ (Choong in sod., 2004). 3.2 CS GEN NE-PEKOČE PAPRIKE IMA 2529 BP DELECIJO Nadalje so bili PCR začetni oligonukleotidi izdelani iz celotne dolžine cdna zaporedja CS (SB2-66) za primerjavo istih genomskih regij pekoče paprike ʹHabaneroʹ z ne pekočo papriko ʹHaehwaIIIʹ. Slika 5 prikazuje PCR produkte, pridobljene s šestimi različnimi kombinacijami začetnih oligonukleotidov pri vzorcih pekočih in ne-pekočih paprik. Struktura CS gena in mesta začetnih oligonukleotidov so prikazani na Sliki 5. PCR pomnoževanje z začetnimi oligonukeleotidi CSF1 in CSR4, in CSF1 in CSR2 je ustvarilo in 434 bp dolga produkta pomnoževanja pri pekoči papriki, toda nobenega fragmenta pri ne-pekoči papriki. Na drugi strani pa so pri pomnoževanju z začetnima oligonukleotidoma CSF3 in CSR4 pridobili 615 bp dolg fragment tako iz pekočih in ne pekočih rastlin. To pomeni, da je C-terminalna regija CS enaka v pekoči in ne-pekoči papriki, medtem ko ima N-terminalna regija delecijo v ne-pekoči papriki. Za bolj natančno definicijo izbrisane regije, so oblikovali kombinacije začetnih oligonukleotidov, ki temeljijo na zaporedju klona BAC667I16 (Yoo, 2003; Yoo in sod., 2001, 2003). Produkti pomnoževanja genomske DNK pekoče paprike ʹHabaneroʹ in ne pekoče ʹHaehwaIIIʹ paprike z začetnima oligonukleotidoma BF5 in CSR4 so bili klonirani, sekvencirani in medsebojno primerjani. Rezultati so pokazali 2529 bp dolgo delecijo v zaporedju ʹHaehwaIIIʹ, kot je prikazano na sliki 5. Ta ugotovitev potrjuje naše prejšnje rezultate, ki kažejo, da N-terminal regija gena CS vsebuje 2,5 kb delecijo v ne pekoči papriki. Da bi ugotovili, ali je odkriti polimorfizem pekoče in ne-pekoče paprika, C. chinense cv. ʹHabaneroʹ in C. annuum cv. ʹHaehwaIIIʹ, reprezentativen tudi pri drugih sortah paprik, so nadalje tri pekoče in tri ne-pekoče sorte testirali s PCR z uporabo kombinacij začetnih oligonukleotidov, ki potrjujejo dolžinski polimorfizmu za CS. Kot so že opazili pri pekoči sorti ʹHabaneroʹ in ne pekoči ʹHaehwaIIIʹ (Slika 6), so pri vseh šestih kultivarjih 6

14 pomnožili 615 bp dolg fragment z začetnima oligonukleotidoma CSF3 in CSR4. Z začetnima oligonukleotidoma BF6 in BR8 so pri pekočih paprikah pomnožili 2800 bp dolg fragment in pri ne-pekočih paprikah kratek fragment dolg le 272 bp. Z začetnima oligonukleotidoma BF7 in BR9, se je pri pekoči papriki pomnožil 896 bp dolg fragment, medtem ko se pri ne-pekoči papriki ta ni pomnožil (Slika 5). Zato so kombinacije začetnih oligonukleotidov BF6 / BR8 in BF7 / BF9 uporabni SCAR markerji za gen CS in za fenotipsko napovedovanje pekočega okusa paprike. Kombinacija začetnih oligonukleotidov BF7 in BR9 pomnoži 896 bp dolg fragment samo v pekoči papriki. "P" je oznaka za pekočo papriko, linija 1, ʹCM334ʹ; linija 2, ʹPI159236ʹ; linija 3, ʹTabascoʹ. "N" je oznaka za ne-pekočo papriko, linija 4, ʹECWʹ; linija 5, ʹSladka Bananaʹ; linija 6, ʹPepperociniʹ (Choong in sod., 2004). Tako so pokazali uporabnost SCAR markerjev za žlahtniteljske programe paprike in filogenetske študije zaporedja regije, kjer je prisotna delecija CS gena pri ne-pekoči papriki. Razviti genetski markerji imajo neposredno uporabno vrednost za razločevanje pekočih in ne pekočih paprik v fazi sadik v žlahtniteljskih programih. Razvitih je bilo šest SCAR markerjev in kombinacije začetnih oligonukleotidov CSF1 in CSR4, CSF1 in CSR2 in BF7 in BR9 so pomnožile dominantne markerje, medtem ko so kombinacije BF5 in BR8 ter BF6 in BR8 pomnožile kodominantne markerje. Kodominantni SCAR markerji so veliko bolj informativni pri ne-pekoči papriki, saj vključujejo vidno delecijo fragmenta. Pomanjkanje dominantnih SCAR markerjev je ta, da ne prepoznajo ne-pekoče paprike od potencialne eksperimentalne napake. Ti markerji so bili zelo učinkoviti pri opredelitvi pekočega fenotipa različnih sort. V nekaterih primerih čilijev, ki so bili v testu okušanja razvrščeni kot ne-pekoči so se pokazali kot zelo malo pekoči s testi z uporabo SCAR markerjev in HPLC analizo. Izredno nizke vsebnosti kapsaicina ni lahko zaznati z metodo okušanja, vendar jih lahko zaznavamo z bolj občutljivimi metodami, kot je HPLC in tudi z genetsko analizo. Pomanjkljivost HPLC metode določevanja pekočega okusa je ta, da moramo počakati na zrele plodove, medtem ko lahko SCAR markerji določijo pekočo lastnost prej (npr. v fazi sejanca) in z neznatnimi količinami rastlinskega materiala. SCAR markerji, ki so bili razviti in predstavljeni, zagotavljajo natančen, enostaven, hiter in gospodaren način zgodnjega odkrivanja pekočega okusa pri rodu Capsicum. Ti SCAR markerji imajo tudi potencial za uporabo pri filogenetskih študijah rodu Capsicum in širše družine Solanaceae. 8

16 3.3 UPORABA MARKERJEV PRI GOJENJU NOVE SORTE SVEŽE PAPRIKE, KI VSEBUJE KAPSINOIDE Kapsinoidi so manj pekoči analogi kapsaicina, ki so jih prvič izolirali iz manj pekočega kultivarja Capsicum annuum ʹCH 19 Sweetʹ (Yazawa in sod., 1989; Kobata in sod., 1998). So podobne strukture kot kapsaicinoidi, vendar imajo estrsko skupino namesto amidne skupine. Kapsinoidi imajo podobne bioaktivnosti in bistveno manjšo stopnjo pekočega okusa kot kapsaicinoidi (Sasahara in sod., 2010; Luo in sod., 2011). Pekoč okus kapsinoidov je približno 1000-krat nižji v primerjavi s kapsaicini. Številne študije so pokazale, da kapsinoidi spodbujajo termogenezo z aktivacijo kapsaicin receptorja (Kawabata in sod., 2009). Poleg tega kapsinoidi kažejo delovanje proti debelosti, indukciji apoptoze, proti raku in imajo antioksidativne lastnosti (Rosa in sod., 2002; Haramizu in sod., 2011). Kapsinoidi so zaradi nižje stopnje pekočega okusa bolj ugodni kot sestavine v živilih in dodatkih kot kapsaicini. Za vnos kapsinoidov iz plodov paprik je treba zeleno papriko zaužiti surovo. Obstajata dva razloga za to. Prvič, ker so kapsinoidi nestabilni v vodi in ob segrevanju (Sutoh in sod., 2001), zato bi se morali izogibati termični obdelavi plodov paprik. Drugič, vsebnost kapsinoidov se zmanjšuje, ko plodovi dozorijo od zelenih do rdečih (Yazawa in sod., 1989). Vendar pa plodovi sorte paprike ʹCH-19 Sweetʹ niso primerni za uživanje surovi, ker so neprijetnega okusa in majhni. Poročali so o nekaterih drugih manj pekočih kultivarjih, ki vsebujejo kapsinoide (Tanaka in sod., 2010). Te sorte prav tako niso primerne za uživanje še ne-dozorele. Zato obstaja potreba po razvoju nove sveže sorte s primernim okusom in vsebnostjo kapsionidov. Takšno novo sorto se lahko razvije s križanjem med obstoječo svežo sorto in sorto, ki vsebuje kapsinoide. Vendar pa bi bil klasičen postopek križanja časovno zelo dolg in delovno intenziven, zaradi sajenja velikega števila križancev na različnih področjih in zaradi kemičnih analiz delov ploda, ker bi populacija kazala različne vsebnosti kapsaicina in kapsinoida. Namen te raziskave je bil torej oceniti uporabnost molekulskih markerjev, ki temeljijo na p-amt in Pun1 genih za pohitritev žlahtniteljskega programa za vzgojo nove sorte s primernim okusom za uživanje in dovolj veliko vsebnostjo kapsinoidov Križanje sort Poznamo papriko sorte ʹCH-19 Sweetʹ, ki vsebuje kapsinoide vendar ni primerna za uživanje. V opisanem žlahtniteljskem programu so razvili novo sorto, ki so jo poimenovali ʹMaru Saladʹ in katere rodovnik je prikazan na Sliki 7. Uporabili so pristop žlahtnjenja s pomočjo markerjev, pristop ki temelji na uporabi molekulskih DNA markerjev, ki pomagajo izbrati željene genotipe. Križali so papriki ʹCH-19 Sweetʹ in ʹMurasakiʹ. ʹCH-19 Sweetʹ je malo pekoč kultivar z veliko kapsinoidi, vendar ima majhne sadeže, ki so neprijetnega okusa. ʹMurasakiʹ je ne-pekoč svež kultivar, ki ne vsebuje kapsinoidov. Za določitev genotipa p-amt, je bil uporabljen pristop dcaps markerjev. Za dcaps analizo so genomsko DNA ekstrahirali iz listov mladih rastlin paprik. dcaps, je preprosta tehnika za genetsko analizo posameznih nukleotidnih polimorfizmov, ki temelji na restrikciji pomnoženih fragmentov. Kapsinoide in kapsaicine v zeleni zreli papriki so določili s HPLC metodo. Določili so vsebnost v 9-ih paprikah. Za populacijo, ki so jo križali 9

17 ʹMurasakiʹ ʹCH-19 Sweetʹ, so tri plodove iz ene posamezne rastline uporabili za oceno vsebnosti kapsinoidov. Za potrditev stabilnosti kapsinoidne vsebine, so gojili več kot 20 rastlin ʹMaru Saladʹ dve leti, in določili vsebino s tremi ponovitvami devetih plodov iz vsake rastline Razvoj novih kultivarjev Razvoj novih svežih kultivarjev, ki vsebujejo kapsinoide, z uporabo DNK markerjev. Pri dcaps analizi za gen p-amt so dobili 317 bp dolg fragment pri sorti ʹMurasakiʹ in 269 bp dolg fragment pri sorti ʹCH-19 Sweetʹ. Analiza markerja za gen Pun1 je pokazala da je sorta ʹMurasakiʹ brez fragmenta medtem ko ima sorta ʹCH-19 Sweetʹ 1670 bp dolg fragment. Gena p-amt in Pun1 lahko označimo kot A in B, torej sta genotipa ʹMurasakiʹ in ʹCH-19 Sweetʹ AAbb in aabb (Slika 8). F1 hibrid, ki izhaja iz križanja ʹMurasakiʹ ʹCH-19 Sweetʹ ima torej genotip AaBb, fenotipsko v glavnem proizvaja pekoče kapsaicine. Po križanju dobimo F2 rastline, rastline genotipa --bb (recesivna oblika Pun1 gena) niso proizvedle niti kapsaicina niti kapsinoidov (slika 8). Rastline genotipov AABB, AaBB, AABb in AaBb so pekoče, ki proizvajajo v glavnem kapsaicine. Paprike genotipov aabb in aabb pa so bile manj pekoče rastline, ki proizvajajo v glavnem alkaloid kapsinoid, ki je prijetnejšega okusa. Rezultati analize z DNA markerjema na podlagi dveh genov so se skladale z rezultati HPLC analize (Slika 8). Tako sta se p-amt in Pun1 gen izkazala za učinkovite markerje za izbor malo pekočih rastlin, ki proizvajajo kapsinoide. Potomci izbranih aabb in aabb rastlin so bili združeni, medsebojno oprašeni in DNA selekcija s pomočjo markerjev je bila ponovljena. S ponavljanjem selekcije in samoopraševanja je bila razvita nova sorta, ki je vsebovala želeno stopnjo kapsinoidov in je bila prijetnega okusa. Novo sorto so poimenovali ʹMaru Saladʹ (slika 7). 10

Rezultati analize z uporabo DNK markerjev so se skladali z rezultati HPLC analize, zato sta se gena p-amt in Pun1 izkazala za učinkovite markerje za izbiro malo pekočih rastlin, ki proizvajajo kapsinoide. Surovi plodovi novo nastale paprike so bili primerni za uživanje in tako primeren vir kapsinoidov. Plodovi so bili večji kot pri papriki ʹCH-19 Sweetʹ. V tej študiji so bili p-amt in Pun1 markerji uporabljeni za žlahtnjenje nove sveže paprike, ki vsebuje kapsinoide imenovane ʹMaru Saladʹ, tako da so uporabili DNK markerje za detekcijo kapsinoidne biosintetične sposobnosti. Vse rastline genotipa --bb niso proizvajale ne kapsinoidov ne kapsaicinov. To je potrdilo da je dominantni alel Pun1 gena odgovoren za sintezo le teh, ob hkratnem predpogoju, da je rastlina recesivni homozigot za p-amt gen. Ugotovili so tudi, da če ima rastlina kapsinoidno biosintezno sposobnost, vendar je ne izrazi, jo lahko še vedno določimo z uporabo DNK markerjev. 4 SKLEPI Nedavno določeno zaporedje genoma paprike lahko služi kot pomemben genomski vir za izboljšanje hranilne in farmacevtske vrednosti, ki jo ima pekoča paprika in za podporo evolucijske in primerjalne genomike družine Solanaceae, enega od svetovno najbolj raznolikih rastlinskih družin. Capsicum je edini rod, ki je razvil biosintezo kapsaicinoidov, ki so sestavljeni iz več kot 20 sorodnih alkaloidov, ki povzročajo pekoč okus pri papriki. Določitev zaporedja genoma pekoče paprike omogoča razumevanje biosintezne poti kapsaicina in predstavlja odličen model za raziskovanje razvoja ostalih sekundarnih metabolitov v rastlinah. Genom paprike zagotavlja trdno osnovo za nadaljnje študije biosintezne poti kapsaicina pri rodu Capsicum z uporabo primerjalne genomike, presnovnega inženiringa in transgenih pristopov. V kombinaciji z nedavno objavljenimi genomi sorodnih vrst paradižnika in 12

20 krompirja, bo zaporedje genoma paprike pomagalo pojasniti evolucijo, raznovrstnost in prilagoditev več kot vrst družine Solanaceae, ki so prilagojene na širok spekter habitatov, ki segajo od suhih puščav do tropskih deževnih gozdov. Ponovno sekvenciranje dveh kultivarjev in de-novo sekvenciranje zaporedja vrste C. chinense zagotavlja prikaz genomske raznolikosti med Capsicum vrstami. Genom paprike bo omogočil napredek novih vzgojnih tehnologij z raziskovanjem genoma in izborom genomskih študij o hortikulturno pomembnih lastnostih, kot so velikost ploda, donos, pekoč okus, odpornost na abiotske strese, hranilnost in odpornost proti boleznim. SCAR markerji, ki so bili razviti in predstavljeni zagotavljajo natančen, enostaven, hiter in gospodaren način zgodnjega odkrivanja pekočega okusa pri rodu Capsicum. Ti SCAR markerji imajo tudi potencial za uporabo pri filogenetskih študijah rodu Capsicum in širše pri družini Solanaceae. Ker se v današnjem svetu vse bolj poudarja zdrava prehrana, bodo hranilne vrednosti in biološke funkcije paprike, zelo pomembne za pridelavo, saj poraba paprike v svetu še naprej narašča. V pomoč pri žlahtnjenju pa nam bo predstavljen genom paprike, poznavanje biosintezne poti kapsaicina ter molekulski markeji za odkrivanje pekočega okusa pri papriki. 13
