Caltag Medsystems provide a number of Euroflow validated reagents for immunophenotyping, including screening tubes and multi-colour antibody cocktails, along with the Infinicyt software for data integration and multidimensional analysis of flow cytometry files. We ve recently introduced The FlowStainer - the first fully automated platform that handles all stages of sample preparation for flow cytometry - more information can be found on page 3. Our Flow Cytometry portfolio includes an extensive number of controls, including Stabilised Human Whole Blood and CD4 Low and Normal Blood Controls. In addition, Caltag Medsystems supply a number of CE/IVD kits for the diagnosis of foetomaternal haemorrhage, heparin induced thrombocytopenia and bacterial sepsis by flow cytometry. The diverse portfolio available comes from a combination of carefully selected suppliers and products manufactured onsite by our Cytomark scientists in Buckingham. 20 Immunophenotyping: Antibodies and Multicolour Cocktails Plasma Cell Studies MGUS and MM Differentiation Clonalpath and Clonalpath -Plus have been optimised in order to differentiate between Monoclonal Gammopathy of Undetermined Significance (MGUS) and Multiple Myeloma (MM) based on the percentage of immunophenotypically normal plasma cells. Clonalpath contains the following antibody combination for the identification and characterisation of plasma cells in a single vial: Normal plasma cells typically show strong reactivity to CD38 antigen, intermediate FSC/SSC pattern, co-express CD19 and CD45 antigen while being generally negative for CD56 antigen. In MGUS, the presence of immunophenotypically normal plasma cells is a constant finding, whilst in MM its frequency is significantly lower (<3%). 21 Immunophenotyping: Antibodies and Multicolour Cocktails Paroxysmal Nocturnal Haemoglobinuria The PNH Screening Kit The Paroxysmal Nocturnal Haemoglobinuria (PNH) Screening Kit is a 4-colour reagent used to evaluate the possible loss of expression of two GPI-anchored proteins, CD16 molecule on neutrophils and CD14 molecule on monocytes. Anti-CD45 and anti-cd64 antibodies allow identifying neutrophils and monocytes. Further evaluation of the loss of CD16 and CD14 in these populations allows the identification of the PNH clone in a fast and simple way. 22 Immunophenotyping: Antibodies and Multicolour Cocktails Kappa & Lambda Antibodies Kappa and Lambda antibodies are widely used in routine diagnostics to evaluate a series of clinical conditions in which lymphoid populations are involved. Kappa/Lambda antibodies manufactured by Cytognos are the reagents of choice for EuroFlow antibody panels (FITC and PE), providing comparable results to polyclonal gold standards. These antibodies also show a high performance in extracellular and intracellular staining. Flow cytometric analysis of blood cells using this reagent allows for the evaluation of TdT expression and the concomitant presentation of cell surface markers indicative of a disease. Quiklysis Quicklysis is an erythrocyte lyse non-wash solution without fixatives, for the testing of leucocyte subpopulations by flow cytometry. BulkLysis protocol allows greater sensitivity in flow cytometry studies where a higher cell count is required.
