6 6 Antibodies (Ab) against Human Antigens New Fluorochrome-Conjugated Antibodies The violet laser is becoming a standard component of flow cytometers and is often used for common phenotyping markers in multicolor staining panels. Cytognos SL offers two new fluorochromes for use with the violet laser, which provide maximum resolution of antigenic determinants when used in a multicolor panel. Their narrow emission peaks result in little spectral overlap, which require minimal intra-laser compensation. CF TM Blue is a dye with similar spectral properties to Pacific Blue which emits at 450nm. It is characterized by: Narrow and well separated emission profiles. The relative stain indexes, which reflect the effective brightness of the antibody to differentiate between the positive and negative population, are optimal. As no excitation is detected in a 488nm laser, compensation requirements are minimal. Compatible with the standard filter sets of cytometers equipped with violet lasers. 18 18 Reagents for DNA Content and Cell Cycle Analysis Cycloscope TM is an exclusive product line for flow cytometric cell DNA content measurements in patients with different diseases. The detection of DNA aneuploidies and cell cycle phase study offers relevant clinical information for prognostic evaluation and follow-up of patients with solid tumors or hematological malignancies. It has been demonstrated for some type of diseases that the existence of DNA aneuploidy is characteristic of malignant neoplasms and depending on the type of disease can be associated with better or worse prognosis. A high proliferative rate of tumor cells is usually associated to poor clinical outcome and helps to evaluate disease progression. The main advantage of this product line is that allows the measurement of cell DNA content on tumor cells specifically identified on basis of their phenotype. Cycloscope TM kits include a specific pool of monoclonal antibodies, to increase the ability to identify the neoplastic cells and separate them from other normal cells present in the sample, and a ready to use Propidium Iodide Solution for DNA analysis. This patented double staining method is sensitive and specific, allowing the simultaneous assessment of antigenic stainings and cell DNA in a single tube. Cycloscospe TM MM: Kit focused on the measurement of plasma cell DNA content in Multiple Myeloma (MM) and Monoclonal Gammopathies of Undetermined Significance (MGUS). 23 Reagents for Monitoring Monoclonal Gammopathies 23 Clonalpath TM y Clonalpath TM Plus Clonalpath TM is the ideal tool for monitoring of MM and MGUS patients based on the quantification of bone marrow plasma cells. Normal plasma cells typically display strong reactivity for the CD38 antigen, intermediate FSC/SSC pattern, co-express the CD19 and CD45 antigen while they are generally negative for the CD56 antigen. The presence of immunophenotypically normal plasma cells, which is a constant finding in MGUS, is a rare event in MM and when it is present, its frequency is significantly lower (<3%) than that observed in MGUS. Pathological plasma cells can be distinguished from normal plasma cells based in their different immunophenotypic pattern of expression for the antibodies anti CD19, CD56, CD45 and CD38. 24 24 Reagent for Dendritic Cell Studies As a major component of the immune system, dendritic cells (DCs) have a critical role in stimulating a tumor-specific immune response. This stimulation is of great interest for its application in immunological-based therapies, such as antitumor cell vaccination with autologous DCs. Therefore, as immunotherapy using these cells has the potential to become a treatment option, it is important to have a greater understanding of the number and function of DCs and to know the optimal conditions for their collection and administration. Most of the studies reveal the difficulties in defining the distinct DCs populations. Dendritic Cells Exclusion Kit is a CD3+CD19+CD56+CD14 antibody combination conjugated with FITC designed to identify by exclusion DCs. Peripheral blood DCs can be distinguished from other leucocyte subsets by their characteristic lack of staining with this antibody combination and a pattern of FSC / SSC intermediate between lymphocytes and monocytes. The combined use of this reagent with other antibodies such as anti human HLA DR, allows a clear identification of total dendritic cells population. Additionally, it has been reported that the use of this dendritic cells exclusion kit combined with anti-human CD33 antibody, anti human CD123 and/or anti-human CD16 allows the identification of different subpopulations of dendritic cells. Dendritic cells Name Reference Format Use DCE Kit CYT-DENDF 50 TEST RUO

25 Absolute Counting Systems 25 The absolute counting of cell subpopulations by means of flow cytometry can be done using a double platform technique (in which the information provided by the flow cytometer and by a haematological counter are combined), or using a single platform technique for which only the flow cytometer is used. The microbead-based single platform technology has emerged as the method of choice for absolute cell enumeration in different clinical applications due to the accuracy of the results. Perfect-Count Microspheres TM Perfect-Count Microspheres TM is a microbead-based single platform system, which assures the accuracy of absolute count results. Its unique internal quality control system contains two types of beads with densities around the upper and lower densities of peripheral blood cells. Variations of the ratio between beads type A and B warn about problems during sample preparation and/or acquisition which could invalidate the final results. 26 26 Reagents LeucoFinder TM LeucoFinder TM is an efficient single-platform method for the enumeration of residual white blood cells (rwbc) in leucoreduced blood products for transfusion. An accurate rwbc absolute count in filtered blood products for transfusion is critical to prevent febrile reactions, micro organism transfer, alloimmunization, platelet refractoriness and immunosuppression. LeucoFinder TM combines the detection of residual leukocytes by DNA labeling of their nucleus with the use of Perfect-Count Microspheres TM as absolute count system that provides a unique method of internal quality control. rwbc Beads B Beads A Beads A Beads B Name Reference Format Use LeucoFinder CYT-LF test CE / IVD

27 Screening Kit for Paroxismal Nocturnal Hemoglobinuria (PNH) 27 Paroxismal Nocturnal Hemoglobinuria (PNH) is an acquired clonal hematopoietic stem-cell disorder related to the occurrence of a somatic mutation in the PIG-A gen, located in the human X chromosome. This genetic alteration results in a partial or total deficiency of all proteins normally linked to the cell membrane by a glycosylphosphatidylinositol (GPI) anchor (GPI-anchored proteins). Flow cytometry provides a direct diagnosis tests in which the lack of GPI anchored proteins is studied on the major cell populations present in blood. The screening kit for PNH includes the following antibody combination: CD16-FITC / CD64-PE / CD45-PerCP-Cy5.5 / CD14-APC. With this antibody combination, it is possible to detect the existence of GPI deficient cells with a 100% of sensitivity. Antibodies anti-cd45 and anti-cd64 allow the identification of the populations to be studied: neutrophils and monocytes. The further evaluation of a possible loss of expression of the CD16 molecule on neutrophils and loss of expression of the CD14 molecule on monocytes allows the identification of the PNH clone in a fast and simple way. monocytes HPN Monocytes Normal Monocytes PNH Neutrophils neutrophils Normal Neutrophils Name Reference Format Use HPN Screening CYT-HPN-1 25 TEST CE / IVD
