1 *ANYONE MORE THAN 15 MINUTES LATE WILL HAVE TO RESCHEDULE TRAINING* The University f Arizna Institutinal Animal Care and Use Cmmittee (IACUC) has adpted the fllwing cntrl measures: Certificatin: All persns requesting access t the Animal Facility wh will be wrking in a muse r rat rm whether the animals are hused in IVC r cnventinal cages must cmplete the IACUC Mandated Hands n "MI certificatin" curse prir t access being granted. Respnsibilities: Principle investigatrs are respnsible fr mnitring the training and abilities f thse persns they supervise. University Animal Care persnnel are charged with the authrity t bserve and dcument failure(s) t fllw MI prcedures. SPF/ Clean animals must be kept under cnditins where bacteria, viruses, fungi, and ther ptential pathgens are minimized; therefre the micr-islatin/ivc caging acts t prtect these animals frm a "dirty" envirnment. IVC caging als reduces wrker expsures t animal allergens. Prper techniques assist in prviding bi-cntainment t prtect animals frm diseases. Any break in technique may cmprmise the health status f these animals. Planning yur wrk day: T insure that diseases are nt brught int the animal rm, always fllw the rm letter designatin (A-F), ensuring t enter higher designated areas first, and wrk with the animals befre ding anything else such as: autpsies n either animals r humans, surgery n either animals r humans, r prviding care fr yur wn pet rdents (try t switch t evening hurs fr pet care). If yu have pet rdents at hme, even if yu d nt care fr them and will be wrking inside the BIO-5 Barrier Facility yu MUST shwer prir entry. IF additinal caging is needed, yu must place yur request via t the Facility Crdinatr at least 2 business days prir t the day the cages are needed. HOW TO ENTER THE MI ROOM: Enter the animal rm and dn PPE per sign n dr, if yu are ging t be pening cages in the hd. New supplies are kept in the PPE bin in the rm. Prtective clthing must be thrwn away after each use r if it becmes dirty r trn. Remve PPE befre yu exit the rm. Handling pans in the laminar flw hd/wrk statin: The gal is t maintain bi-cntainment and keep the animals in the cage frm being expsed t any ther animals r pssible diseases that may be in the rm. It is recmmended that nly ne cage f animals be placed in the hd and pened at a time. Any time yur glved hands cme ut f the hd r tuches the utside f the animals cage yu must wet yur glved hands with disinfectant slutin when yu bring yur glved hands back int the hd BEFORE tuching anything, which is cnsidered clean. Step 1- Turn n the laminar flw hd if it is nt a cntinuusly perating hd Step 2- CLEANING HOOD: Spray the wrk surface f hd with disinfectant wipe with paper twels leaving hd still wet with disinfectant. (IF USING a chlrine based slutin, such as Clidx, it is crrsive t stainless steel, immediately after cleaning hd wipe the stainless steel surfaces with alchl.) Step 3- Use the spray r pump bttle f disinfectant t disinfect glved hands s that YOUR GLOVED HANDS ARE WET WITH DISINFECTANT AT ALL TIMES. If using frceps, use the pump r spray bttle wet the frceps between each use. 1

2 Step 4- Get pan f animals frm rack and place pan n "dirty" side f hd. Step 5- Remve MI lid and place lip dwn in hd. Step 6- Wet glves with disinfectant slutin (Yu just tuched the utside "dirty" surface f the MI lid) Step 7- Place clean equipment in the hd as needed, making sure t nt blck air flw while maintaining a clean and dirty wrk area. D nt place feeder with fd and Hydr-pac (r just fd) directly n surface f hd. Feeder shuld be placed in a clean empty pan if full access t the pan is needed r feeder can be placed n a clean paper twel. Step 8- If pssible use the rubber tipped frceps t handle mice (if yu can't use frceps, use glved hands which are wet with disinfectant slutin). Make sure t pick up animals (rats/mice) by the base f the tail r scruff f the neck. Step 9- In sterile rms use autclaved pans, r in standard rms use clean pans, bth can be fund in the rm r btained frm the clean side f the cage wash area t separate rdents, r t perfrm prcedures. Extra caging in rm is there fr all researchers, if yu require mre than ne r tw cages yu must request additinal cage set-ups. Send requests via t the Facility Crdinatr at least 48 business hurs in advance t ensure cages are in the rm when yu need them. NOTE: NEVER PUT RODENTS, FEEDERS, HYDROPACS OR BOTTLES ON TOP OR INSIDE OF M.I. LID; if the lid gets wet it will nt prperly prtect the animals in the cage. Step 10 - In IVC rms with aut water, the water valves inside the pans must be charged n the charging statin befre use t ensure the valve is wrking prperly. Step 11- Replace MI lid. BE SURE LID IS ON CORRECTLY! (The side f the MI lid labeled Air/Exhaust Rear shuld be n the side f the cage that has the water valve). The Rat lids have a plastic clip n the frnt f the MI lid. Step 12- Clean hd with disinfectant and alchl (if using Clidx) when finished wrking in rm. Discnnect charging statin frm the water hse. Turn ff lights and blwer in hd, unless therwise requested by the PI and hd is signed stating t be left n. ROOM RULES: University Animal Care technicians have first pririty in the rm. If they are wrking in the hd yu will be asked t wait until they are finished. Research staff must clean up the rm after themselves. This includes cleaning f hd and taking dirty caging t dirty cage wash area unless stated t leave caging in rm. N supplies can be stred in hd. FACILITY RULES: Investigatrs and their technicians are respnsible fr cvering animals with a sheet, lab cat, twels, pillw cases, bxes r ther suitable cver prvided by their lab. DO NOT use new gwns frm the PPE bin. Anytime yu have animals t be PTS, DO NOT cmbine any mre than 5 adult r 10 weanling mice in ne pan. Fr rats, make sure t fllw the NIH guidelines psted n the dr in the animal rm fr adults and n mre than 6 weanlings per cage. This is per Federal Plicy. Place the apprpriate PTS card n the cage and leave in the rm fr UAC. Take the Cage card and put in the black deactivatin bx fund by the facility exit drs. 2

3 THE MOST COMMON MISTAKES INCLUDE THE FOLLOWING: Nt wearing the crrect prtective dispsable clthes, r wearing dispsable clthing imprperly int the animal rm. Opening a cage i.e. remving the micr-islatr tp utside f the laminar flw bench. Taking animals and hlding them utside f the laminar flw bench Allwing the filter paper t get wet, trn r dirty Nt wearing glves, r nt keeping glves wet with disinfectant Nt dipping r spraying glved hands with disinfectant after tuching "dirty" surfaces Nt disinfecting equipment between grups f animals Frgetting t turn the water bttles arund during transprt t lab and back t rm r nt placing a Hydrpac in an IVC cage when transprting them t the lab, if needed. Frgetting t charge new IVC pans befre placing n the rack with animals. Placing the cage int the rack incrrectly. Nt ensuring cage is dcked prperly. Cage is put in backwards i.e.: the lixit is tward the frnt f the cage. Nt disinfecting the hd befre and after use. Frgetting t turn ff lights in hd when dne, especially in breeding rms where this can adversely affect breeding perfrmance. Vilatin Penalties: FIRST VIOLATION: On the first ccasin that ANY persn fails t fllw prper MI prcedures, that persn will be required t repeat individualized MI certificatin within tw weeks f the vilatin. PI is respnsible fr ensuring persn receives the required re-training and a fee fr the training will be charged t the PI s accunt. The principle investigatr fr the prject will be ntified f the nature f the vilatin, the persn invlved, and the date f cmpletin f individualized re-certificatin. Cntinued vilatins will be reprted t the IACUC and further penalties will be determined by the IACUC. Please reprt all suspected vilatins t The Facility Crdinatr, CAF Miguel Diaz ( ), AHSC Cheryl Jhnsn ( ) r BIO5 Cindy Richner ( ) (); they will investigate and reprt t the IACUC. Remve the next card in sequence. 3) Recrd the fllwing infrmatin n the activatin strip (bttm) f the cage card: Circle the card designatin: Wean = Animals cunted against prtcl Split = Animals nt cunted against prtcl CIRCLE Split FOR NEW BREEDER SET UPS Date and Initial Recrd number f animals in cage CIRCLE UAC IF CARD IS ACTIVATED BY UAC PERSONNEL. 2) Recrd the fllwing infrmatin n the main bdy (middle) f the cage card: Sex (Male, Female, Either, Bth) DOB (Date f Birth) Rec d =Date card put int use AND Initials f persn using card Per Cage = Number f Animals DO NOT LEAVE BLANK Other ntes as desired Activatin Strip (bttm f card) 1. Remve activatin strip frm main bdy at perfratin. Place int any black cage-card bx near entrance/exit t facility. Cards are cllected and activated daily. 2. Place main bdy f card and deactivatin strip (leave attached) int cage card hlder. 3. DEACTIVATION: Once cage is emptied f riginal mice, remve deactivatin strip (tp) frm main bdy f cage card. Place int any black cage-card bx near entrance/exit t facility fr deactivatin. Cards are cllected and deactivated daily. 4

6 IF AN ACTIVATION OR DEACTIVATION STRIP IS MISSING INFORMATION, IT WILL BE HANDLED AS FOLLOWS: Activatin strip NOT DATED OR SUBMITTED: Card will be activated back t first date f last scan. This will result in up t tw weeks additinal per diem charged. Deactivatin strip NOT SUBMITTED: Card will remain active until it has missed three cnsecutive scans. This will result in a minimum f 4 (maximum f 6) weeks additinal per diem charged. Number f animals NOT INDICATED: Five mice will be cunted against IACUC prtcl.
