Customized sample input size and elution volume for optimized DNA/RNA extraction result Introduction LabTurbo 24/48 compact systems are liquid handling workstations equip with innovative vacuum manifold and individual spin column for DNA/RNA extraction. LabTurbo systems provide complete automation solutions for the workflow of DNA/RNA preparation and reduce the needs of manpower and manual operations. By using individual spin columns with the innovative vacuum manifold, LabTurbo enables customized volume of sample input and wash buffer for optimized extraction results. In addition, the elution volume is independent of the sample input volume and can also be customized. Users can elute with only 50 ul to get ultra-high DNA/RNA concentration for superb detection sensitivity. 4 LabTurbo Extraction Protocol and Worktable The workflow of DNA/RNA preparation involves a lot of processes, such as sample aliquoting, DNA/RNA extraction, and assay (PCR) setup. As a completely automated system, LabTurbo provides comprehensive functions for liquid handling and DNA/RNA extraction. LabTurbo robotic set equips with spanned 6-channel pipette system and ultrasonic device for fast and flexible liquid handling functions and avoid cross-contamination from residual carry-over. By using the individual spin columns and innovative vacuum manifold, LabTurbo protocol is optimized for nucleic acid extraction with combined benefits of the manual method and automation. The worktable consists of six units to automate the steps of the extraction protocol (as shown in the picture. At first, sample aliquoting can be performed by the system or manual transfer. For auto-aliquoting, the system can automatically transfer sample from primary tubes placed on pos.1 to the working tube located at pos.4; Proteinase K is placed on the position 2; There are five reagent tanks at position 3 and all of the reagents will be automatically transferred to the reagent tanks by LabTurbo auto-vending system; the thermolysis of sample takes place at position 4. Samples are thoroughly mixed by pipetting and incubated for a defined time, even overnight; the binding and washing steps are performed on the vacuum manifold at position 5; after washing steps, the spin column will be transferred to another vacuum manifold with a removable elution cover at position 6 for elution. During the extraction processes, the robotic set will discard the plastic consumables through the trash channel and recover the worktable for the next run. After the extraction protocol is finished, LabTurbo can continue to run another extraction protocol or assay (PCR) setup.

5 LabTurbo Chemistry and Plastic Consumables LabTurbo uses individual spin column and vacuum technology to extract DNA/RNA from different types of samples; the spin column includes a silica membrane for chromatographic binding of DNA/RNA, allowing the removal of non-specific materials and elution of DNA/RNA products under optimized conditions. 4 plastic consumables are basically necessary for LabTurbo systems, including the 96-tip plates for liquid handling, 6-tube strips for sample loading and thermolysis, 6-spin column sets for binding and washing, and individual micro tube or 6-tube strip for elution. As the plastic consumables are in the formats of 96-tip plate or 6-tube strip, it does not need to be loaded one by one; so the worktable can be quickly set up in less than five minutes. All of the reagents, components, and Proteinase K in the LabTurbo kits are recommended to store at room temperature and are qualified for DNase-free and RNase-free tests. The kits contain all of the reagents, plastic consumables, and components except 96-tip plate and ethanol for LabTurbo extraction protocols. β-mercaptoethanol or isopropanol are not provided in the RNA extraction kits. The carrier RNA is supplied as a ready-to-use solution and can be stored at room temperature. It is not needed to prepare fresh solution from powder every time.

6 LabTurbo Kits and applications LabTurbo kits are suitable for various applications; Please see the following for the guidelines: LGD480: LabTurbo Genomic DNA extraction kit for 480 preparations; it is for extracting genomic DNA from blood, buffy coat, cells, tissues, synovial fluids, and pathogens. There are 4 sets of LGD kits for various applications; LGD is for 3 x 10 6 cells, 30 mg of tissues, or 300 ul of whole blood. The yield of DNA is up to 20 ug. LGD is for 5 x 10 6 cells, 50 mg of tissues, 500 ul of whole blood, or 200 ul of buffy coat prepared from 2 ml of whole blood. The yields of DNA can reach 30 ug. LGD is for 1 x 10 7 cells, 100 mg of tissues, 1000 ul of whole blood, or 500 ul of buffy coat prepared from 5 ml of whole blood. The yields of DNA can get to 60 ug. LGD is for 2 x 10 7 cells or 2000 ul of whole blood. The yields of DNA can get to 100 ug. For tissues or challenging samples, the LTL buffer must be purchased separately for pre-treatment. The purities of OD 260/280 and 260/230 are among and , respectively. The elution volume can be set between 50ul ul. LVN480: LabTurbo Viral nucleic acid extraction kit for 480 preparation; it is for extracting viral DNA/RNA and circulating DNA from cell-free fluids, such as serum, plasma, CSF, urine, swab, etc. There are 3 sets of LVN480 kits for different applications; LVN , LVN , and LVN are for 300 ul, 500 ul, and 1000 ul of cell-free fluids, respectively. LVN480 has widely been used for the detection of HBV, HCV, Norovirus (stool), HPV (swab), Influenza (swab), EBV, BKV, CMV, etc. The elution volume can be set at 60 ul. LVX480: LabTurbo nucleic acid extraction kit for 480 preparations. It is suitable for extracting DNA/RNA from multiple types of samples simultaneously with the same protocol. It is also suitable for samples that are difficult to extract DNA/RNA by standard kits and protocols, such as stool, soil, swabs, animal/fish tissues, FFPE-fixed tissues, plants, samples of low DNA/RNA

7 contents, etc. There are 4 sets of LVX kits for various applications: LVX is for 30 mg of tissues or 300 ul of fluids. LVX is for 50 mg of tissues or 500 ul of fluids. LVX is for 100 mg of tissues or 1000 ul of fluids. LVX is for 2000 ul of fluids. The sample input volume of this kit can be customized to up to 8 ml. Please inquire us directly for more information. The elution volume can be set at 50 ul. LSD480: LabTurbo Swab DNA kit for 480 preparations. It is for extracting DNA from swabs and solid forensic samples; swabs such as buccal swab, pharyngeal swab, vaginal swab, etc., and solid forensic samples including cigarette butts, straw, blood stain, glove, chewing gun, etc., for forensic DNA analysis. The swab or solid forensic samples can be directly applied to LabTurbo to start without manual pre-lysis procedures. The elution volume can be set at 50 ul. LTR480: LabTurbo Total RNA kit for 480 preparations. It is for extracting total RNA / micro RNA from cells, tissues, and cell-free fluids. There are 2 sets of LTR kits for different applications; LTR is for 5 x 10 6 cells (WBC), 15 mg of animal tissues, or 100 mg of plant tissues. LTR is for 1 x 10 7 cells (WBC), 30 mg of animal tissues, or 200 mg of plant tissues. The elution volume can be set between 50 ul to 100 ul.

8 Cross-Contamination Free LabTurbo was tested for cross-contamination using samples having HBV titer up to 10 9 copies / ml. Consistency LabTurbo are outstanding systems and can provide consistent extraction results of yields and purity for replicated samples of 1000 ul whole blood.

9 Which system is more sensitive for HBV DNA detection? Parallel comparison of HBV DNA detection of serum samples extracted by X-auto and LabTurbo. X-auto provided 12 highest values and LabTurbo discovered 23 highest values. Is it useful to use LabTurbo extraction systems to increase sample input volume for better detection sensitivity? Yes it is! The sensitivity test was performed with replicated serum inputs of 300 ul, 500 ul, 1000 ul, and 2000 ul. The elution volume was 50 ul for the whole test. The Cts values show a negative linear correlation to the serum inputs; it shows that the detection sensitivity is proportional to the increased sample input.

11 LabTurbo 96 Standard System Introduction LabTurbo 96 Standard System is an ultrafast, fully automated DNA/RNA extraction system for high throughput applications. It can process up to 96 samples within 70 minutes. By using the 96-well spin column plate, it can process various sample types and provide high DNA/RNA yield, purity, and recovery rate. It accommodates both single tubes and 96-well plates; it takes only 10 minutes for setting up 96 samples. LabTurbo software recognizes excel, txt, and csv file formats. 17 VacEZor VacEZor Introduction The VacEZor is an innovative vacuum manifold for simplifying nucleic acid extraction using manual spin column kits. It equips with a vacuum manifold, vacuum pump, waste bottle, and timer. It is designed for extracting DNA/RNA on a single platform.
