1 Chemistry Notices For the Week of March 4 to March 8, 2013 Submissions: Please send to newsletter@chem.ualberta.ca by Wednesday for the following week s edition VISITING SPEAKERS & SEMINARS Analytical Division Visiting Speaker Dr. Ulrich Krull GRL/Bonnie x3477 Last printed :56 AM Professor, Department of Chemical and Physical Sciences University of Toronto Mississauga will speak on " Quantum Dots as Multiplexed Optical Transducers: From Microfluidic Bioassays and CMOS Sensors to Interrogation of Chemical Dynamics within Cells" There are significant analytical advantages that can be achieved by combining the unique optical properties of quantum dots (QDs) with bioconjugation of oligonucleotide probe molecules to create nano-scale biosensing elements. Of particular relevance are advantages in size scale and in multiplexing using optical technology. We have been exploring QDs that are decorated with oligonucleotides as the basis for bioprobes that operate using fluorescence resonance energy transfer (FRET), where the QD excites a nearby dye that is associated with the formation of double-stranded DNA. By extension, the principles of function have bearing on use of other selective binding agents such as based on proteins and aptamers. Migration from use of optical fiber technology to place solid-phase bioassays into microfluidic channels has allowed for transduction of sub-femtomole levels of target nucleic acid within minutes, rather than hours as typically required in previous assay designs. Moreover, the rate of mass transport of an injection of oligonucleotide targets to the in-channel interface that was coated with immobilized QDs enabled the quantitative transduction of target nucleic acid by measuring the length of the microfluidic channel that exhibited FRET sensitized photoluminescence. This represents a new approach to quantification that is not dependent on the proportionality of fluorescence intensity to concentration, and that allows for accurate quantification without use of fluorescently labeled target. This presentation will explore the design of QD-bioprobes, multiplexing of QD-bioprobes that are immobilized on optical fibers and in microfluidic channels, integration of optical nanoparticle probes on CMOS technology, the use of microfluidics to construct nano-scale biosensing elements for intracellular applications, and some early results of QDbioprobes in intracellular response. Reaction monitoring on micro-second timescales by nuclear magnetic resonance: Postdoctoral Research Fellow in organometallic photochemistry w/ Professors Simon Duckett, Robin Perutz Applications are invited for a postdoctoral research fellow to work on photochemical studies of organometallic complexes by NMR spectroscopy. This position is for a period of 2 years in the first instance and available from the 15th of April The post holder requires an organometallic or coordination chemistry background with experience of NMR. The PDRA will be located in the York Centre for Hyperpolarisation in Magnetic Resonance and in the Department of Chemistry. Laser irradiation methods have provided the means to study the evolution of many chemical systems. In proof-of-principle experiments, we have shown that it is possible to record NMR spectra on milli and microsecond timescales. We seek to use NMR to follow fast reactions using parahydrogen. The post holder will work alongside a Postdoctoral Research Fellow responsible for theory to achieve this aim. He/she will prepare complexes, examine their role in catalysis and study their photochemical reactivity by NMR in quantitative, data-rich experiments. 2. The position is tenable for a period of 2 years in the first instance and available from the 15th of April The post holder requires an excellent theoretical appreciation of NMR spectroscopy. He/she will be located in the York Centre for Hyperpolarisation in Magnetic Resonance and in the Department of Chemistry. In chemistry, laser irradiation methods have provided the means to study the evolution of many chemical systems. In proof-of-principle experiments, we have shown that it is possible to record NMR spectra on milli and microsecond timescales. We seek here to develop a method for using NMR to follow fast reactions using parahydrogen. The post holder will develop a theoretical framework to account for the evolution of parahydrogen-derived magnetisation after the photochemical addition of parahydrogen to a metal complex. He/she will then make measurements in conjunction with a second Postdoctoral Research Fellow to test their conclusions, and ultimately seek to produce NMR spectra that encode reaction rate information on timescales down to microseconds. The new methods will be used to study catalysis. 3. Postdoctoral research fellow exploring MRI and SABRE w/ Professors Simon Duckett, Gary Green Applications are invited for a postdoctoral research fellow to work in the York Centre for Hyperpolarisation in Magnetic Resonance on the development of SABRE methods for MRI. The position is tenable for a period of 3 years and available from the 15th of April The successful candidate will be responsible for the development of SABRE based methods to probe blood flow, the blood-brain barrier, cellular function and cellular metabolism in vivo. The post holder requires a good practical knowledge of MRI. 4. Postdoctoral research fellow in the synthesis and MRI detection of labelled materials w/ Professors Simon Duckett, Gary Green Applications are invited for a postdoctoral research fellow to work in the York Centre for Hyperpolarisation in Magnetic Resonance on the synthesis of labelled materials for subsequent use in imaging studies. The position is tenable for a period of 3 years and available from the 15th of April The successful candidate will be responsible for the synthesis of a range of organic and inorganic systems that are enriched in NMR active nuclei. As part of this work they will be involved in aspects of the development of imaging agents for MRI. The post holder requires a good practical knowledge of organic synthesis and a familiarity with NMR.
