Cryopreservation, is the most effective way of preserving biological materials, as all biological ageing is stopped at extreme low temperatures. In cryobiology there is no such thing as too cold, but there is a critical temperature for most biological materials. This temperature, known as the glass transition temperature (Tg), is approximately -130 C. When storing at a minimum of 10 to 30 C below Tg, i.e. at -150 C to -160 C, the biological material can be stored almost indefinitely with a very low percentage decomposition. Special freezers are cooled using liquid nitrogen. Liquid nitrogen ( ) has a temperature of -196 C, generating a freezer temperature of -150 C to -196 C, which is an ideal range for cell storage. Cells can either be stored submerged in the in the vapour phase above the liquid phase, or in recently developed systems in the dry phase. The quality of the vessel is the most important storage factor, assuming the samples have been frozen adequately. The better the insulation from external factors, the better the temperature stability inside the vessel will be. Cryo storage vessel features are developed to minimise the influence of the outer temperature: High vacuum multi-layered super insulation Composite neck holding the inner vessel and outer vessel tightly together, without heat transfer Hooks connected with composite bars to the racks, also to minimise heat transfer Small neck openings minimise non-vacuum insulated area of the vessel Plug with insulating cork Optional electronics give the researcher ongoing feedback. Auto-fill systems mean that there is no need to check manually the level of on a regular basis. For safety of the samples, all parameters exceeding the limits will generate an alarm on the vessel or other external system. A variety of different storage systems are available to suit the conservation of different types of specimens including straws, cryo tubes or bags in different sizes. Safety considerations is dangerous when in contact with the skin or eyes so requires careful handling by trained personnel. Protective gloves, glasses and if possible an apron must be worn when handling it. For larger systems we advise you to install an oxygen monitoring system, as the evaporation of large quantities of nitrogen in closed rooms may deplete oxygen levels and can lead to asphyxiation. 4 Cryo storage Canister storage system The CSS-series of tanks offer you storage of small to medium capacity applications in efficient, narrow neck designs, with extremely low consumption. The XSS-series, offers you similar benefits, and has extended storage capacity. The tanks are manufactured from an aluminium alloy with fibreglass/epoxy neck, and the high vacuum multi-layered super insulation from the inner space limits the nitrogen evaporation. 6 Cryo storage Box storage system The BSS-series tanks offer various solutions for storage of vials or straws in liquid or gaseous phase. 8 Cryo storage Dry shippers Dry shippers are used for safe transportation of samples at cryogenic temperatures. A hydrophobic absorbent material is fixed in the inner capacity of the vessel. After complete saturation of the absorbers with, the vessels are ready for transport of your samples at cryogenic temperatures, without the risk of flowing out, if the container is overturned. These vessels are not meant to be used on airplanes (non-iata). Model DS-24 can be equipped with removable absorbent cartridges to improve the holding time. The bigger units (25, 35 and 50) are fitted with a NW50 PNEUROP flange for mounting a pressure head for withdrawal. The small LD-2M is fitted with a pouring spout. The fixed withdrawal elbow with phase separator offers ease of filling small vessels eliminating splashing. Small amounts of can be withdrawn from the dewar, until the inner pressure of the vessel has dropped to ambient pressure. A pressure gauge and relief valve of 0,5 bar assures safe operation. The head needs to be removed before filling of the vessel. The stainless steel plunger tube needs to be cut to the inner height of the vessel : With the use of the TS head, small amounts of can be withdrawn from the dewar, till the inner pressure of the vessel has dropped to ambient pressure. The vessel can be filled via the filling valve, without the need to remove the head. A stainless steel flexible diameter 10 mm or rigid transfer elbow can be connected to the filling/withdrawal valve. The head is equipped with a NW50 flange, a filling/withdrawal valve, a pressure build up valve, a relief valve set at 0,5 bar and a pressure gauge. 12 Cryo storage cylinders These non-magnetic stainless steel cylinders are designed for and argon applications. They can be connected permanently to the AF-series sample storage tanks, or be used as storage cylinders to fill smaller vessels. The standard pressure building kit ensures that can be withdrawn at any moment. All models are provided with manometer, safety relief valve, fill and withdrawal valve (with standard connection for flexible) and vent valve. The control module is designed to manage the cryogenic tank automatically. The units need supply from a stand-alone pressurised vessel, or from a central supply system. AF-75, -200 and -250 have an aluminium inner and outer vessel. The units can be mounted on optional castors. The S-170 controller has a separate housing attached to the side of the vessel. The lids can be locked with a padlock (padlock not included, not possible on AF-75) AF-110 has a round aluminium inner vessel and a square outer casing of powder coated metal. The unit is mounted on castors. AF-400 has a stainless steel inner vessel. The user can choose a rotating platform on this model (AF-400P). This facilitates the access to the racks in the back, and reduces the number of racks. Castors and lock are standard mounted. On both models the S-170 controller is built into the front panel for easy setting and reading. AF-75, AF-200 and AF-250 series AF-800P and AF-1200P full stainless steel biological containers are designed for storage of samples in the most efficient and safe conditions in liquid or gaseous phase. Consumption of liquid nitrogen is reduced and holding time is increased thanks to the narrow neck diameter. Your valuable samples will be stored in gaseous phase even at -150 C right under the lid. The samples are very accessible thanks to an inner rotating tray, which may be turned around by means of a turning handle located on the top of the container. The S-170 controller is mounted in a separate housing on the top of the vessel.
