Following are the Products and services from WITA For more information please visit : 2-DE is the most potent methods of analyzing the complete proteome of cells, cell lines, organs and tissues in proteomic studies. The advances in Proteomics have been possible through the development of advanced high resolution WITA VISION 2-DE systems allowing resolution up to protein spots of entire cell lysates in combination with protein identification by new highly sensitive mass spectrometric techniques. The knowledge of the human genome and its polymorphisms is the basis for an understanding of the gene functions in cellular processes. However, the data of the genes describe only the static state of the inherited information. To understand the dynamic processes within the cells, e.g. during cell differentiation and regulation or during disease development, it is mandatory to study the proteins involved. Their presence, actual concentrations, and possible modifications must be known in order to gain clues about their functional activities. This means that entire protein extracts of cells or expression pattern of the proteins, among them stress, alterations in temperature, different growth conditions, and the effects of various treatments. The analysis of malfunctioning cellular processes would allow the identification of biomarkers useful for the design of drugs to targets under specific disease situations. Furthermore, the individual genotypic differences of proteins, such as enzymes, under drug application cause differences in the tolerance against certain drugs. As they may be of therapeutic

2 use for some of the patients and toxic or inefficient for others, individualized therapies can be evaluated through proteomics. WITA is a corporation, having their Head office in Germany Teltow. 3 WITA s 1-DE GEL ELECTROPHORESIS SYSTEM : Advantages of WITA VISION 1-DE chamber Processing of 10 simultaneous 1-D gels Security cover with electric protection Storage of processed and re-buffered 1-D gels on the gel transfer modules at 80 degree C 1st dimension:iso-electric Focusing (NEPHGE technique) using proprietary ampholines (WITAlyte) ph 2 11 Gel casting using WITA s Gel Casting stand is very simple In Other Systems 2000 to 3000 ml of electrode solution is required where as in WITA s system only 400 ml electrode solution is required. 7 COMPARISION OF WITA S NEPHGE TECHNOLOGY WITH IPG WITA uses a NEPHGE (Non Equilibrium PH Gradient Technology for the Gel Electrophoresis) which gives certain advantages over the IPG technology. WITA Separation based on NEPHGE Technology Separation by IPG Technology NEPHGE 1st dimension separation according to IP of proteins. Separation gradient within the gel is forming by soluble ampholyte during electrophoresis. IPG 1st dimension separation according to IP of proteins. Fixed ph-gradients on pre-casted paper strips. NEPHGE 1st dimension separation standard sizes 24 and 41 cm. Larger sizes are possible in principle. IPG In the 1st dimension due to physical limitations no separation above 24 cm in length is possible.

8 NEPHGE NO endpoint method. 1st dimension electrophoresis is terminated BEFORE proteins reach their IP and point of minimal solubility: Proteins stay in solution, ready to enter 2nd dimension gel. IPG Endpoint method. 1st dimension electrophoresis is terminated AFTER proteins reached their IP and point of minimal solubility: Many proteins precipitate; substantial losses of proteins from 1st to 2nd dimension of gel. NEPHGE No limitations for DIGE 2-DE IPG No limitations for DIGE 2-DE NEPHGE standardized gel sizes 24 x 30 cm and 41 X 30 cm IPG maximal gel size 24 x 18 cm commercially available In combination with the WITA VISION 2 nd dimension apparatus gel size of 24 x 30 cm possible NEPHGE up to individual spots per gel, suitable for MS identification of selected proteins IPG No more than spots per gel.
